This study evaluated the effectiveness of a LHRH fusion protein vaccine on endocrine changes, feedlot performance, and carcass quality of bulls compared with steers and hormone-implanted steers. Crossbred bulls (n = 30; mean weight, 179 +/- 4 kg; mean age, 130 +/- 2 d) were randomly assigned to three treatment groups: 1) castrated (castrated; n = 10); 2) castrated-implanted with trenbolone acetate (implanted; n = 10); and 3) immunized against a cocktail of recombinant fusion proteins, ovalbumin-LHRH-7 and thioredoxin-LHRH-7 (immunized bulls; n = 10). Blood was collected every 2 wk to evaluate antibody and hormone concentrations. Serum LHRH antibodies (P < 0.001) were detected in animals of the immunized group, which had reduced serum LH concentrations (P < 0.001) compared with the castrated groups and serum FSH concentrations, which did not decrease but were significantly different when compared with castrated and implanted animals. Serum testosterone concentrations in the immunized bulls were not different from the two castrated groups (P > 0.05) by d 60 after primary immunization. Initial mean scrotal circumference of the immunized bulls was 18.0 +/- 0.6 cm on d 0 and increased to 22.6 +/- 1.3 cm by d 310. No differences (P > 0.05) in ADG were observed among treatment groups. Immunized animals had an intermediate BW gain (P > 0.05) when compared with the castrates, whereas the castrated groups differed (P < 0.05) from each other. Carcass characteristics were similar (P < 0.05) among the three groups. Vaccinating bulls against a LHRH fusion protein cocktail suppressed LH and testosterone, which led to reduced testicular development and no bullock carcasses. Growth and carcass characteristics of the immunized animals were similar to the steers.  相似文献

OBJECTIVE: To measure the ocular surface area, scleral thickness, and the aqueous and vitreous humor volumes in the canine and equine eye. Animals studied Fourteen canine and 16 equine cadaver eyes. PROCEDURE: Eyes were measured either fresh or following fixation in formalin. Ocular surface area was calculated using the fluid displacement method. Globes were hemisected and aqueous and vitreous humor were collected and quantitated. Scleral thickness was measured by digital caliper, by image projection, and by ultrasonic biomicroscopy (UBM). RESULTS: Mean +/- standard deviation (SD) scleral surface areas in canine and equine eyes were 12.87 +/- 2.24 and 57.23 +/- 5.63 cm2, respectively. Mean +/- SD aqueous humor volume was 0.77 +/- 0.24 in the dog and 3.04 +/- 1.27 mL in the horse. Mean vitreous humor volume was 1.7 +/- 0.86 mL for the canine eye and 26.15 +/- 4.87 mL for the equine eye. In canine and equine eyes, the sclera was thinnest at the ocular equator: 0.34 +/- 0.13 mm (canine) and 0.53 +/- 0.1 (equine). There were no significant differences between the direct caliper and projected thickness measurements or differences between measurements in the superior, inferior, nasal, and temporal quadrants of the eye. Scleral thickness in fresh tissue was greater than in fixed tissue at most sites. CONCLUSIONS: The UBM measurement method appeared to be most consistent and reproducible when compared to direct measurement techniques. Formalin fixation of the eyes was associated with significantly thinner scleral tissue than with fresh ocular tissue.  相似文献

The objective of this study was to determine whether an antiestrogen (enclomiphene) would shorten the interval to first estrus and conception in postpartum beef cows. Sixty postpartum Angus beef cows were stratified by age, body condition, and calving date and were randomly assigned to one of two treatment groups. Group 1 cows (n = 24) received three silastic implants, each containing 150 mg of enclomiphene, on d 20 postpartum. Implants were removed on d 30 postpartum. Group 2 cows (n = 28), received empty implants and served as controls. Cows were artificially inseminated at first detected estrus. Estrus detection and ovulation were further verified by increased serum progesterone. Concentrations and pulse frequencies of LH were determined from blood samples collected at 15-min intervals for 6 h on d 20, 25, 30, and 40 postpartum. Hypothalami and pituitaries were collected from four cows in each treatment group on d 30 postpartum and analyzed for concentrations of estradiol receptors. Concentrations of total and unoccupied hypothalamic and pituitary estradiol receptors were reduced by enclomiphene. Neither concentrations nor pulse frequencies of LH differed significantly between treatment groups on any of the 4 d. Days to first estrus did not differ (P greater than .05) between enclomiphene-treated (57 +/- 6; n = 24) and control (56 +/- 4; n = 28) cows. Days to conception did not differ between treated (81 +/- 9) and control (79 +/- 8) cows. The dose of enclomiphene used in this study reduced hypothalamic and pituitary estrogen receptors but did not alter secretion of LH or days to first estrus in the postpartum beef cow.  相似文献

The objectives of this study were 1) to evaluate the ability of trenbolone acetate (TBA) administered in tandem with LHRH immunization to suppress reproductive function in bulls and 2) to examine the effects of LHRH and androgen (TBA) signaling on pituitary gland function. Forty-four Angus × Hereford crossbred calves (BW=225 ± 2 kg; age=187 ± 6 d) received castration, LHRH immunization, or TBA administration in a 2 × 2 × 2 factorial design. Treatment groups receiving LHRH immunization contained 6 animals, whereas other treatment groups contained 5 animals. Animals immunized against LHRH received a primary injection and 2 booster injections of ovalbumin-LHRH-7 fusion protein on d 0, 42, and 196, respectively. Animals treated with TBA were implanted on d 224. Serum LHRH antibodies increased (P<0.05) after each booster for immunized animals, but were negligible in nonimmunized animals throughout the experiment. Serum testosterone concentration (P<0.001) and scrotal circumference (P<0.05) were depressed in LHRH-immunized bulls compared with nonimmunized bulls by d 84 and 168 of the experiment, respectively. Treatment with TBA tended (P=0.08) to decrease serum testosterone concentrations of nonimmunized bulls. Weights of testes at slaughter were decreased (P<0.001) for LHRH-immunized (232 ± 41 g) compared with nonimmunized (752 ± 45 g) bulls, but did not differ (P=0.80) between TBA-implanted (500 ± 49 g) and nonimplanted bulls (484 ± 36 g). Both LHRH immunization and castration decreased pituitary gland stores of LH and FSH (P<0. 001). There was no effect (P>0.10) of TBA on pituitary gland FSH content and only a tendency (P=0.09) to increase pituitary gland LH content. Immunization against LHRH decreased expression of LH β-subunit and common α-subunit genes (P<0.001). Castration increased expression of LH β-subunit and common α-subunit genes (P=0.02). Treatment with TBA further suppressed (P=0.04) α-subunit mRNA expression in LHRH-immunized steers. In summary, LHRH immunization decreased synthesis and storage of LH and decreased storage, but not synthesis of FSH in bulls. The increased synthesis of LH and FSH in nonimmunized, but not LHRH-immunized steers suggests that castration removes the negative feedback on gonadotropin synthesis but that LHRH is still needed for release of these hormones. Androgen replacement with TBA did not restore the negative feedback control of gonadotropin synthesis.  相似文献

Many aquatic habitats in coastal Oregon have been impacted by historic land use practices that led to losses of in-stream wood and associated degraded fish habitats. Many of these streams are now bordered by stands of dense second growth forests(30–80 years) that are incorporated into riparian buffer zones with low wood recruitment and storage. Thinning in riparian zones is one management option to increase the rate of large tree growth and eventually larger in-stream wood, however, it raises concern about impacts on current wood recruitment, among other issues. Using a forest growth simulation model coupled to a model of in-stream wood recruitment, we explore riparian management alternatives in a Douglas-fir plantation in coastal Oregon. Alternatives included:(1) no treatment,(2) single and double entry thinning, without and with a 10-m buffer, and(3) thinning combined with mechanical introduction of some portion of the thinned trees into the stream(tree tipping). Compared to no treatment, single and double entry thinning on one side of a channel, without a 10-m buffer, reduce cumulative instream wood volume by 33 and 42 %, respectively, after100 years(includes decay). Maintaining a 10-m buffer reduces the in-stream wood loss to 7 %(single entry thin)and 11 %(double entry). To completely offset the losses of in-stream wood in a single entry thin(on one or both sides of the stream), in the absence or presence of a 10-m buffer,requires a 12–14 % rate of tree tipping. Relative to the notreatment alternative, cumulative in-stream wood storage can be increased up to 24 % in a double-entry thin with no buffer by tipping 15–20 % of the thinned trees(increased to 48 % if thinning and tipping simultaneously on both sides of the stream). The predicted increases in in-stream wood that can occur during a thin with tree tipping may be effective for restoring fish habitat, particularly in aquatic systems that have poor habitat conditions and low levels of in-stream wood due to historic land use activities.  相似文献

Climate change and other anthropogenic disturbances can lead to the loss of genetic variation and thereby affect evolutionary potential and survival of plant populations in the wild. We examined these predictions in the primary wild relative of barley, Hordeum vulgare L. subsp. spontaneum (K. Koch) Thell., within its center of diversity, in Jordan. Changes in genotypic and phenotypic diversity were assessed using seed samples collected in 1981 and 2012 from the same 18 sites across Jordan. The overall population structure was conserved, but we observed an increase of within population genetic diversity and a reduction in population differentiation. Phenotypic variation differed among years and sites but the magnitude and direction of change variated among sites. While the sampled region became significantly hotter and drier during this period, simple correlation models did not support association between measures of climate change and the observed genetic and phenotypic changes. Agricultural activities that promote disturbance and demographic fluctuations may affect crop wild relatives that grow in agricultural landscapes, in unexpected ways. The observed increase in genetic diversity within populations might be explained by increased migration or by an advantage of increased genetic variation in the face of variable environmental conditions. This study provides a new perspective on the range of possible responses of crop wild relatives to environmental pressures.  相似文献

Genomewide data sets of single nucleotide polymorphisms (SNPs) offer great potential to improve ex situ conservation. Two factors impede their use for producing core collections. First, due to the large number of SNPs, the assembly of collections that maximize diversity may be intractable using existing, serial software algorithms. Second, the effect of the natural partitioning of the genome into linked regions, or haplotype blocks, on the optimization of collections, and the capture of diversity, is unknown. To address the first problem, we report the development of a parallel computer program, M+, for identifying optimized core collections from arbitrarily large genotypic data sets on high performance computing systems. With respect to the second problem, we use three exemplar data sets to show that, as haplotype block length increases, the number of accessions necessary to capture a predetermined proportion of genomewide haplotypic variation also increases. This relationship is asymptotic such that the minimum haplotype block length suitable for assembling core collections can be empirically determined, and the number of accessions necessary to capture a given percentage of the haplotypic diversity present in the entire collection can be estimated, even when true haplotype structure is unknown. Additionally, we test whether simple geographic or environmental information can be used to produce core collections with elevated genomewide haplotypic diversity. We find this opportunity to be limited, and dependent on natural history and improvement status.  相似文献

Complementary DNA clones were isolated that represent the 5' terminal 2.5 kilobases of the murine Duchenne muscular dystrophy (Dmd) messenger RNA (mRNA). Mouse Dmd mRNA was detectable in skeletal and cardiac muscle and at a level approximately 90 percent lower in brain. Dmd mRNA is also present, but at much lower than normal levels, in both the muscle and brain of three different strains of dystrophic mdx mice. The identification of Dmd mRNA in brain raises the possibility of a relation between human Duchenne muscular dystrophy (DMD) gene expression and the mental retardation found in some DMD males. These results also provide evidence that the mdx mutations are allelic variants of mouse Dmd gene mutations.  相似文献
