Prestile is a late maturing potato variety with attractive, round to oblong, white-skinned, white-fleshed tubers with shallow eyes. Its major use is expected to be for tablestock in areas where Katahdin and Ontario are now grown. Prestile tubers are slow to sprout in storage and do not show the net necrosis caused by potato leafroll virus, but are quite susceptible to heat necrosis. This variety is immune to potato virus X, and is moderately resistant to common scab (Streptomyces scabies), verticillium wilt (Verticillium albo-atrum), and early blight (Alternaria solani). Prestile is susceptible to skinning and shatter bruise, and is difficult to kill, particularly when heavily fertilized.  相似文献

The objective of this research was to examine the population structure of full‐blood (100%) Wagyu cattle registered in the United States with the American Wagyu Association, with the aim of estimating and comparing the levels of inbreeding from both pedigree and genotypic data. A total of 4132 full‐blood Wagyu cattle pedigrees were assessed and used to compute the inbreeding coefficients (FIT and FST) and the effective population size (Ne) from pedigree data for the period 1994 to 2011. In addition to pedigree analysis, 47 full‐blood Wagyu cattle representing eight prominent sire lines in the American Wagyu cattle population were genotyped using the Illumina BovineSNP50 BeadChip. Genotypic data were then used to estimate genomic inbreeding coefficients (FROH) by calculating runs of homozygosity. The mean inbreeding coefficient based on the pedigree data was estimated at 4.80%. The effective population size averaged 17 between the years 1994 and 2011 with an increase of 42.9 in 2000 and a drop of 1.8 in 2011. Examination of the runs of homozygosity revealed that the 47 Wagyu cattle from the eight prominent sire lines had a mean genomic inbreeding coefficient (FROH) estimated at 9.08% compared to a mean inbreeding coefficient based on pedigree data of 4.8%. These data suggest that the mean genotype inbreeding coefficient of full‐blood Wagyu cattle exceeds the inbreeding coefficient identified by pedigree. Inbreeding has increased slowly at a rate of 0.03% per year over the past 17 years. Wagyu breeders should continue to utilize many sires from divergent lines and consider outcrossing to other breeds to enhance genetic diversity and minimize the adverse effects of inbreeding in Wagyu.  相似文献

The aim of study was to correlate tumour necrosis factor-α (TNF) infused doses used with the TNF concentrations achieved and with the secretory function of both the ovary and the uterus in cows. We evaluated the concentrations of progesterone (P4), prostaglandin (PG)F, PGE2 nitric oxide (NO) and TNF in the jugular vein and vena cava caudalis as parameters of exogenous TNF action on the female reproductive tract. Aortae abdominalis of cows (n = 18) were infused with saline or two doses of TNF (luteolytic – 1 μg or luteotrophic – 10 μg). In the peripheral blood, 1 μg TNF concentrations achieved within the range of 30–45 pg/ml, and 10 μg TNF provoked a sharp increase in achieved concentrations at a range of 250–450 pg/mL). The TNF concentrations achieved in vena cava caudalis were five to six times higher than that in peripheral blood (p < 0.001). One microgram TNF increased PGF and NO (p < 0.001) and decreased P4 (p < 0.05). The higher TNF dose stimulated P4 and PGE2 (p < 0.01). TNF infusion at luteolytic dose achieved its concentrations at the physiological range previously observed in cows. Luteotrophic TNF dose achieved the concentrations in vena cava caudalis that are much higher than physiological level and were previously noted in pathological circumstances (i.e. mastitis , metritis ).  相似文献

The objectives of this study were to evaluate the effects of immunization against recombinant GnRH fusion proteins and growth promotants on onset of puberty, feedlot performance, and carcass characteristics of beef heifers. Heifers were immunized against an ovalbumin fusion protein containing 7 GnRH peptides (oGnRH, n = 12), a thioredoxin fusion protein containing 7 GnRH peptides (tGnRH, n = 12), a combination of oGnRH plus tGnRH (otGnRH, n = 12), or a combination of ovalbumin and thioredoxin (control, n = 11). Each heifer received a primary immunization containing 1 mg of protein in 1 mL of adjuvant injected into the mammary gland at wk 0 (mean age = 38 wk) and booster immunizations at wk 6 and 12. Six heifers within each treatment received Synovex H implants at wk -2. Weekly blood samples were collected from wk -2 to 26 for determination of serum progesterone concentrations and GnRH antibody titers. In GnRH-immunized heifers, GnRH antibody titers increased after the first booster injection, peaked after the second booster injection, and remained elevated through the end of the study (P < 0.01). Heifers immunized against oGnRH achieved greater (P < 0.05) GnRH antibody titers than tGnRH heifers but did not differ (P = 0.20) from otGnRH heifers. During the 26-wk study, ovulation was prevented (P < 0.05) in 10 out of 12, 12 out of 12, 11 out of 12, and 0 out of 11 tGnRH, oGnRH, otGnRH, and control heifers, respectively. At slaughter, uterine weights were lighter (P < 0.01) for GnRH-immunized heifers than control heifers. Synovex H-implanted heifers had greater (P < 0.05) ADG from wk -2 to 26, greater LM area, and lesser percentages of KPH, yield grade, and quality grade than nonimplanted heifers, regardless of the immunization treatment. Immunization against GnRH fusion proteins resulted in production of antibodies against GnRH that prevented ovulation in 92% of the heifers without affecting feedlot or carcass performance. Implanting heifers with Synovex H improved ADG, LM area, and yield grade. Improvements in delivery of the oGnRH vaccine may provide a feasible alternative to surgical spaying of heifers.  相似文献

Selenium is an essential mineral micronutrient for animals, and significant evidence supports an association between supranutritional Se intake and a reduction in the incidence of some forms of cancer. Thus, supplemental Se intake may provide an avenue for reducing cancer incidence. However, an important issue to consider is the form of Se that should be provided in such a supplement, because the bioavailability and bioactivity of Se can vary dramatically depending on the chemical form in which it is delivered. Because wheat products are the largest source of Se in U.S. diets, the absorption of Se was evaluated in different fractions of milled wheat that exhibits very high Se levels, owing to its production on naturally Se-rich soils. An experiment was conducted to determine the bioavailability of Se from three milled fractions of high-Se wheat. The method used was the slope-ratio assay, which measures the ability of Se from the wheat fractions to regenerate Se-dependent enzyme activities and tissue Se concentrations in Se-deficient rats. The responses generated from wheat Se were compared to a standard response curve generated by feeding graded amounts of Se as sodium selenite (Na2SeO3; NaSelenite) or selenomethionine (SeMet) in an AIN-93G-Torula yeast-based diet. Results showed that Se from wheat flour ( approximately 75% extraction) was nearly 100% available by a number of measures including plasma, liver, kidney, and muscle Se concentrations and liver and erythrocyte Se-dependent enzyme activities when compared with similar measures in rats fed NaSelenite or SeMet. However, on the basis of similar criteria, Se from wheat shorts was only about 85% available and that from wheat bran was about 60% available for absorption. These results indicate that high-Se wheat products, mainly those made from refined flour alone, might be particularly well suited for use as dietary Se supplements.  相似文献

Diffuse reflectance Fourier transform mid infrared (FTMIR) and near-infrared spectroscopy (FTNIR) were compared to scanning monochromator-grating-based near-infrared spectroscopy (SMNIR), for their ability to quantify fatty acids (FA) in forages. A total of 182 samples from thirteen different forage cultivars and three different harvest times were analyzed. Three calibration analyses were conducted for lauric (C12:0), myristic (C14:0), palmitic (C16:0), stearic (C18:0), palmitoleic (C16:1), oleic (C18:1), linoleic (C18:2), and alpha-linolenic (C18:3) acids. When all samples were used in a one-out partial least squares (PLS) calibration, the average R (2) were FTNIR (0.95) > SMNIR (0.94) > FTMIR (0.91). Constituents C18:2 and C16:0 had among the highest R (2) regardless of the spectroscopic method used. The FTNIR did better for C12:0, C14:0, and C18:3. The SMNIR did better for C16:0, C16:1, C18:0, C18:1, and C18:2. A second set of calibrations developed with half of the samples as the calibration set and the rest as the validation set showed that all the methods produce acceptable calibrations, with calibration R (2) above 0.9 for most constituents. However, the SMNIR had a better average calibration relative error than the FTNIR, which was slightly better than the FTMIR. A third set of calibration equations developed using 100 random PLS runs with the 182 samples split randomly also shows that the three spectral methods are satisfactory for predicting FA. It is not clear whether any of the spectral methods is distinctly better than another. Calibration R (2) and validation R (2) were higher for most FA with the SMNIR than the FTMIR and FTNIR.  相似文献

We quantified spatial and temporal variations of the fluxes of nitrous oxide (N2O) and methane (CH4) and associated abiotic sediment parameters across a subtropical river estuary sediment dominated by grey mangrove (Avicennia marina). N2O and CH4 fluxes from sediment were measured adjacent to the river (“fringe”) and in the mangrove forest (“forest”) at 3-h intervals throughout the day during autumn, winter and summer. N2O fluxes from sediment ranged from an average of −4 μg to 65 μg N2O m−2 h−1 representing N2O sink and emission. CH4 emissions varied by several orders of magnitude from 3 μg to 17.4 mg CH4 m−2 h−1. Fluxes of N2O and CH4 differed significantly between sampling seasons, as well as between fringe and forest positions. In addition, N2O flux differed significantly between time of day of sampling. Higher bulk density and total carbon content in sediment were significant contributors towards decreasing N2O emission; rates of N2O emission increased with less negative sediment redox potential (Eh). Porewater profiles of nitrate plus nitrite (NOx) suggest that denitrification was the major process of nitrogen transformation in the sediment and possible contributor to N2O production. A significant decrease in CH4 emission was observed with increasing Eh, but higher sediment temperature was the most significant variable contributing to CH4 emission. From April 2004 to July 2005, sediment levels of dissolved ammonium, nitrate, and total carbon content declined, most likely from decreased input of diffuse nutrient and carbon sources upstream from the study site; concomitantly average CH4 emissions decreased significantly. On the basis of their global warming potentials, N2O and CH4 fluxes, expressed as CO2-equivalent (CO2-e) emissions, showed that CH4 emissions dominated in summer and autumn seasons (82-98% CO2-e emissions), whereas N2O emissions dominated in winter (67-95% of CO2-e emissions) when overall CO2-e emissions were low. Our study highlights the importance of seasonal N2O contributions, particularly when conditions driving CH4 emissions may be less favourable. For the accurate upscaling of N2O and CH4 flux to annual rates, we need to assess relative contributions of individual trace gases to net CO2-e emissions, and the influence of elevated nutrient inputs and mitigation options across a number of mangrove sites or across regional scales. This requires a careful sampling design at site-level that captures the potentially considerable temporal and spatial variation of N2O and CH4 emissions.  相似文献

The objective was to evaluate near-infrared diffuse reflectance spectroscopy (NIRS) for determining the composition of poultry manures. Poultry manure samples were obtained from a commercial testing laboratory along with conventionally determined analyte values for total N, NH(4)(+)-N, organic N (computed as total minus NH(4)(+)-N), minerals, and moisture. Samples were blended in a food blender and scanned in polyethylene bags on a FOSS-NIRSystems model 6500 scanning monochromator. Initial results indicated that changes had occurred in NH(4)(+)-N contents from the time of analysis, and samples were reanalyzed for NH(4)(+)-N. Using the new analyses, NIRS was able to accurately determine NH(4)(+)-N, organic N, total N, and moisture in 128 poultry manures. Results were more accurate when determinations were made on an "as is" as opposed to dry matter basis. Finally, results indicated that NIRS was not suitable for the determination of minerals (P, K, Ca, etc.) in poultry manures.  相似文献

Spinosad is a bacterially derived insect control agent consisting of two active compounds, spinosyns A and D. The objective of this paper is to describe the environmental fate of spinosad in aquatic systems. To this end, several studies performed to meet regulatory requirements are used to study the fate and degradation in individual environmental media. Specifically, investigations of abiotic (hydrolysis and photolysis) and biotic (aerobic and anaerobic aquatic) processes are described. Understanding developed from the laboratory-based studies has been tested and augmented by an outdoor microcosm study. Understanding of aquatic fate is a building block for a complete environmental safety assessment of spinosad products (Cleveland, C. B.; Mayes, M. A.; Cryer, S. A. Pest Manag. Sci. 2001, 58, 70-84). From individual investigations, the following understanding of dissipation emerges: (1) Aqueous photolysis of spinosad is rapid (observed half-lives of <1 up to 2 days in summer sunlight) and will be the primary route of degradation in aquatic systems exposed to sunlight. (2) Biotic transformations contribute to spinosad's dissipation, but less so than photolysis; they will be of primary importance only in the absence of light. (3) Spinosad partitions rapidly (within a few days) from water to organic matter and soil/sediment in aquatic systems but not so rapidly as to replace sunlight as the primary route of dissipation. (4) Abiotic hydrolysis is relatively unimportant compared to other dissipation routes, except under highly basic (artificial) conditions and even then observed half-lives are approximately 8 months. Degradation pathways are understood are follows: (1) Degradation primarily proceeds by loss of the forosamine sugar and reduction of the 13,14-bond on the macrolide ring under aqueous photolytic conditions. (2) Degradation to several other compounds occurs through biotic degradation. Degradation under anaerobic conditions primarily involves changes and substitutions in the rhamnose ring, eventually followed by complete loss of the rhamnose ring. Degradation under aerobic conditions was more extensive (to smaller compounds) with the loss of both the forosamine and rhamnose sugars to diketone spinosyn aglycon degradates. (3) Hydrolytic degradation involves loss of the forosamine sugar and water and reduction on the macrolide ring to a double bond at the 16,17-position.  相似文献
