Poor sperm viability post-thaw has resulted in constant research into methods of cryopreservation of canine semen. One factor that may be involved in poor viability is sperm oxidative stress caused by excessive formation of reactive oxygen species. The present study was performed in order to evaluate the effect of different concentrations of ascorbic acid (AA) and glutathione (Glu) added to an extender for the freeze-thawing of dog sperm. Semen from five mature dogs was collected and frozen in two studies. Prior to and after freezing, sperm motility, morphology and membrane status were examined. In addition, sperm motility was examined up to 120 min after thawing to evaluate thermo-resistance. In study I, semen was collected twice from each dog. On both occasions, semen was divided into three aliquots: control, Glu 1 m m and Glu 5 m m . In study II, semen was collected twice and divided into three aliquots; control, AA 50 μ m and AA 250 μ m . Initial sperm motility was significantly higher in sperm diluted with AA 50 μ m ; sperm longevity, however, measured by a thermal-resistance test (TRT), was higher for Glu treatments. Higher concentration of Glu produced significant improvement in TRT and membrane status, whereas higher concentration of AA had a negative impact in sperm longevity. Antioxidant supplementation to semen freezing extenders improved semen quality post-thaw. Moreover, Glu had the most beneficial effect when supplemented at 5 m m . 相似文献

Ablation of follicles ≥ 6 mm in diameter and treatment with PGF2α 10 days after ovulation were used to induce the development of ovulatory waves. Comparisons were made between induced waves with one (33 waves, 72%) and multiple (13 waves, 28%) ovulatory follicles. Diameter deviation was defined as the separation of follicles into dominant and subordinate categories. Multiple ovulatory follicles were preceded by more (p < 0.001) follicles ≥ 20 mm at the beginning of deviation, higher LH preceding deviation (approached significance, p < 0.08), lower (p < 0.05) concentrations of FSH on the day of deviation and thereafter, and higher (p < 0.0003) oestradiol by 2 days after deviation. During the peri-ovulatory period, systemic hormone concentrations for waves with multiple ovulations involved higher oestradiol before ovulation (approached significance, p < 0.07), lower FSH (p < 0.04) before and after ovulation, and both higher progesterone (p < 0.05) and lower LH (p < 0.05) beginning the day after ovulation. Results indicated that by the beginning of deviation there were more follicles ≥ 20 mm and subsequently greater oestradiol production in waves that led to the development of multiple ovulatory follicles, and during the peri-ovulatory period differences between one and multiple ovulations were consistent with the negative effects of the ovarian hormones on the gonadotropins. 相似文献

Despite numerous assessments of the sensitivity and resilience of drylands to degradation, there has been little research into the way affected communities innovate and adapt in response to land degradation. This paper shows how local and scientific knowledge can be combined to identify rangeland management strategies to reduce or adapt to land degradation. To achieve this, we have developed and applied a four‐stage social learning approach based on stakeholder participation in three degradation ‘hotspots’ in communal rangelands of the Kalahari, Botswana. This approach aims to collate, evaluate and apply both scientific and local knowledge on rangeland degradation and management options. First, current practice and possible management options were identified from the literature. Second, a series of semi‐structured interviews with rangeland users identified local knowledge of strategies to reduce and adapt to land degradation. Third, these options were discussed and evaluated with rangeland stakeholders in focus groups held across each study region. Finally, the outputs from these focus groups were used to produce rangeland assessment guides for each region that provided management options agreed to be locally relevant by both researchers and local stakeholders. The study found that the majority of strategies reported in the literature were not suitable for use by pastoralists in the Kalahari. However, many of the strategies suggested by stakeholders could only be applied effectively under common property regimes, giving impetus to the growing literature encouraging institutional reform to strengthen common property management regimes. The research stimulated a social learning process that combined knowledge from local stakeholders (both pastoralists and extension workers) with the scientific knowledge of researchers to provide a range of management options that could help land managers reduce or adapt to land degradation. By combining participatory research with insights from scientific literature in this way, more relevant results were provided than either approach could have achieved alone. OBJECTIVE: To report a technique for fluoroscopically guided closed reduction with internal fixation of fractures of the lateral portion of the humeral condyle (FLHC) and determine the long-term results in 10 clinical cases. STUDY DESIGN: Prospective clinical case study. ANIMALS: Ten dogs with 11 fractures. METHODS: Fractures of the lateral portion of the humeral condyle were stabilized with transcondylar screws and Kirschner wires. Closed reduction and implant placement were achieved using intraoperative fluoroscopic guidance. After fracture repair, postoperative radiographs were evaluated for articular alignment and implant placement. Dogs were evaluated after surgery by means of lameness scores, elbow range of motion (ROM), radiographic assessment, and owner evaluation of function. RESULTS: Postoperative reduction was considered anatomic in 6 fractures with all other fractures having <1.5 mm of malreduction. Follow-up was available for 9 patients from 9 to 21 months after surgery. All of the fractures had healed. One minor (wire migration) and one major (implant failure) complication occurred. Mean lameness scores were 0 (n = 6), 0.5 (n = 2), and 1 (n = 1) at the time of final follow-up. No significant differences were found in follow-up ROM values between affected and unaffected elbows. All of the dogs in this study regained 90-100% of full function, based on owner assessment. CONCLUSIONS AND CLINICAL RELEVANCE: Fluoroscopic guidance for closed reduction and internal fixation of FLHC in dogs is an effective technique. 相似文献

Observations of outflow velocities in coronal holes (regions of open coronal magnetic field) have recently been obtained with the Solar and Heliospheric Observatory (SOHO) spacecraft. Velocity maps of Ne7+ from its bright resonance line at 770 angstroms, formed at the base of the corona, show a relationship between outflow velocity and chromospheric magnetic network structure, suggesting that the solar wind is rooted at its base to this structure, emanating from localized regions along boundaries and boundary intersections of magnetic network cells. This apparent relation to the chromospheric magnetic network and the relatively large outflow velocity signatures will improve understanding of the complex structure and dynamics at the base of the corona and the source region of the solar wind. 相似文献

The purpose of this series of experiments was to determine the effect of various types of immunosuppressive treatments (cyclophosphamide, infectious bursal disease virus [IBDV], chicken anemia virus [CAV], and combination infection with IBDV and CAV) on susceptibility of chickens to challenge with avian polyomavirus. In the first experiment, chickens were chemically bursectomized with intraperitoneal injections of cyclophosphamide; in the second study, chickens were orally inoculated with IBDV; in the third study, birds were intramuscularly inoculated with CAV; and in the final study, birds were inoculated with both IBDV and CAV. In all experiments, chickens were challenged with 10(4.7) tissue culture infective doses of polyomavirus intraperitoneally. Only chemically bursectomized chickens developed lesions similar to those found in the naturally occurring multisystemic fatal form of polyomavirus infection seen in psittacine nestlings, including hepatic necrosis and large pale intranuclear inclusions. 相似文献

X-ray absorption spectroscopy and x-ray fluorescence microprobe studies of 35-million-year-old calcite from a Mississippi Valley-type zinc ore deposit indicate substitution of tetravalent uranium for divalent calcium. Thus, tetravalent uranium has a stable location in calcite deposited under reducing conditions. This result validates uranium-series dating methods (including uranium/lead dating) for ancient calcite and shows that calcite provides a sink for uranium in deep groundwater aquifers and anoxic lacustrine and marine basins. 相似文献

The herbicide‐binding region of the chloroplast psbA gene from a total of 20 biotypes of Poa annua L resistant and susceptible to metribuzin and diuron was selectively amplified using PCR. Sequence analysis of the fragment from six herbicide‐resistant biotypes of P annua exhibited a substitution from valine to isoleucine at position 219 of the D1 protein encoded by the psbA gene. This is the same mutation as reported for Chlamydomonas and Synechococcus through site‐directed mutagenesis and in cell cultures of Chenopodium rubrum L. To our knowledge this is the first report of a higher plant exhibiting resistance in the field to photosystem II inhibitors due to a psbA mutation other than at position 264. The existence of additional biotypes of P annua resistant to diuron or metribuzin but lacking mutation in the herbicide‐binding region indicates that resistance to these herbicides can also be attained by other mechanisms. © 2000 Society of Chemical Industry 相似文献

Learning from a “benign neglect strategy” in a national park: Response of saproxylic beetles to dead wood accumulation 总被引:1,自引:0,他引:1
Increasing demands for firewood owing to rising energy costs have accelerated discussions about the amount of dead wood needed for conservation. A sharp increase in dead wood caused by bark beetles in a German national park provides lessons for management of commercial and protected forests. We investigated the effects of dead wood due to bark beetle infestation as well as tree senility on abundance and richness of saproxylic species of beetles. Increasing amounts of spruce dead wood and opening of the canopy by bark beetles had positive effects on the abundance of host-generalist, conifer-specialist, and red-listed saproxylic beetles. Broadleaf specialists were positively associated with the amount of broadleaf dead wood and negatively associated with canopy openness. Gradient analysis of beetle assemblages revealed two major environmental axes: canopy openness and amount of dead wood. We found a threshold for community divergence at a canopy openness of 23% (confidence interval CI: 11-49) and at an amount of dead wood of 64 m3 ha−1 (CI: 35-160). Critically endangered species served as indicators of dense and open forests, but only when the amount of dead wood was high. Our results suggest that, to maintain saproxylic beetle assemblages, the amount of dead wood in commercial montane forests (at present ≈15 m3 ha−1) needs to be tripled, with a focus on broadleaf wood in dense stands and spruce wood in open stands. For large protected areas in Europe, our data suggest that bark beetle infestation and senescence without active forest management improves habitat conditions for saproxylic beetles. 相似文献
