微量Bradford法测定提纯禽结核菌素蛋白含量   总被引:4,自引:0,他引:4  
研究用96孔微量板Bradford法测定提纯禽结核菌素蛋白含量。通过不同浓度的待测禽结核菌素与考马斯亮蓝G-250溶液混合后,在595nm光波下测定各孔样品的OD值。微量Bradford法检测禽结核菌素的线性范围是50—1000μg/mL,相关系数为0.999,最低定量限为50μg/mL。试验用3支禽结核菌素国际参照品,分别配制成125、250、500μg/mL三种不同浓度,每支每种浓度测定4次,计算实际测得的蛋白质浓度。结果显示,批内相对标准偏差为1.6%~4.4%;批间相对标准偏差0.7%-2.2%。研究结果表明微量Bradford法可作为测定禽结核菌素蛋白含量的一种方法。  相似文献

Teat lesions are common in dairy cattle in New Zealand. Most of the infectious cases are caused by pseudocowpox virus, but lesions similar to those caused by bovine herpesvirus (BHV)-2 have been described clinically and confirmed serologically in New Zealand (Horner and Raynel 1988). BHV-2 and BHV-4 cause an acute viral disease in cattle known as bovine herpes mammillitis or ulcerative mammillitis (Hillerton et al 2001). Presumptive cases have been reported in New Zealand based on the nature and distribution of lesions (Daniel 1970). The disease is probably endemic in New Zealand but is poorly documented and generally considered to be mild and sporadic. The main differential diagnosis is pseudocowpox, which is often also present in herds affected with bovine herpes mammillitis, and dual infections of individual cattle have been reported (Gibbs et al 1972). To my knowledge, neither BHV-2 nor BHV-4 have yet been isolated in New Zealand.  相似文献

Airborne in situ observations of molecules with a wide range of lifetimes (methane, nitrous oxide, reactive nitrogen, ozone, chlorinated halocarbons, and halon-1211), used in a tropical tracer model, show that mid-latitude air is entrained into the tropical lower stratosphere within about 13.5 months; transport is faster in the reverse direction. Because exchange with the tropics is slower than global photochemical models generally assume, ozone at mid-latitudes appears to be more sensitive to elevated levels of industrial chlorine than is currently predicted. Nevertheless, about 45 percent of air in the tropical ascent region at 21 kilometers is of mid-latitude origin, implying that emissions from supersonic aircraft could reach the middle stratosphere.  相似文献

Objective To determine reference ranges for healthy yearling farmed saltwater crocodiles by performing routine biochemical and haematological laboratory tests on blood samples.
Design A clinico-pathological study.
Procedure Blood samples were collected from 120 healthy yearlings from four Northern Territory crocodile farms and body weight and length were measured. All animals had been fasted for 2 days before sample collection. Routine biochemical analytes were determined on 120 samples and haematological values determined on 30 samples (from one farm).
Results Reference ranges for biochemical and haemato-logical values were determined for farmed yearling saltwater crocodiles in the Northern Territory.
Conclusion The results were comparable with published reference ranges for other crocodilian species. Other published results of haematological values from saltwater crocodiles were from very young (6-week-old) hatchlings and older (2- to 4-year-old) crocodiles. Differences in values were presumed to be caused by age-related factors.  相似文献

AIM: To estimate the incidence of clinical mastitis and the frequency of isolation of mastitis-causing organisms from clinical cases in one lactation season (July 2005 to May 2006) on 14 dairy farms from the Northland region of New Zealand.

METHODS: Cases of clinical mastitis were determined by trained farm personnel who recorded the identity of affected cows. Pooled milk samples from affected quarter(s) were aseptically collected by the farm personnel, for microbiology. Mean numbers of affected cows and quarters were compared at the population and farm level per 305 cow-days-at-risk (DAR).

RESULTS: One or more cases of clinical mastitis occurred in 559/3,765 (14.8%) lactating cows. The average incidence of clinical mastitis was 0.19 cases per 305 DAR. The incidence in rear quarters (56.2%) was 1.3 times (p=0.027) that of front quarters (43.8%). The incidence of clinical mastitis and numbers of affected quarters were signifi cantly infl uenced by the stage of lactation (higher in early lactation), age (higher in older cows) and farm. At the cow level, the most common isolates were Staphylococcus aureus (23.7%) and Streptococcus uberis (23.3%). No causative organisms were identifi ed in 19.9% of the samples. Each cow had an average of 1.8 quarters affected during a case of clinical mastitis.

CONCLUSIONS: This study demonstrated a higher incidence of staphylococcal clinical mastitis on dairy farms from Northland than has been reported in other regions of New Zealand.  相似文献

In various species, sex, hormonal treatments and oestrous-cycle stage have been shown to affect the animal's response in behavioural tests. Few such studies have been performed in the horse. The main aim of the present study was to investigate whether oestrous-cycle stage affects mares' response to a novel object test and isolation test and, in part, to study whether mares, assumed to suffer from oestrous-related behavioural problems, respond differently in these tests when compared with controls. Twelve mares were tested twice, in oestrus and dioestrus, in a crossover design. Seven behavioural and two heart rate variables were measured for the novel object test and two heart rate variables for the isolation test. Oestrous-cycle stage and whether a mare was classified as a 'problem' mare did not affect the mare's response. However, test order, i.e. the cycle stage a mare was tested in first, affected its reaction. This effect could partly be explained by significant differences between test occasions 1 and 2 in three behavioural variables and one heart rate variable (p < 0.05) in the novel object test. The mares explored the novel object more and had a higher mean heart rate in the first test. Exploring the novel object more could largely be attributed to those mares tested in dioestrus first, perhaps indicating that the mares in oestrus were less receptive to the novel object. The reason for the differences between test occasions could be an effect of learning or habituation.  相似文献

Drought is becoming a major threat to rice farming across the globe owing to the depletion of water tables in rice-growing belts. Drought affects rice plants at multiple stages, causing damage at morphological and physio-biochemical levels, leading to severe losses that exceed losses from all other stresses. The amalgamation of conventional breeding methods with modern molecular biology tools and biometrical methods could help accelerate the genetic gain for drought tolerance in rice. Many drought-tolerance traits with genetic determinants have been identified and exploited for tolerance rice variety breeding. The integration of genome-wide association study and genomic selection tools with speed breeding shortened the breeding cycle and aided in rapid improvement of genetic gain. In this review, we emphasized the progress made through classical breeding as well as the limitations and usefulness of current genomic methods in improving drought tolerance. We briefly addressed methods for identifying genetic determinants for drought tolerance and deploying them through genomics-assisted breeding programmes to develop high-yielding drought-tolerant rice cultivars.  相似文献

AIMS: The principle aim of this study was to examine the association between infection with Theileria orientalis Ikeda type and growth rates of suckled beef calves on four beef farms. In addition, associations between calf sex, sampling time, and individual farm and T. orientalis Ikeda type infection intensity and haematocrit (HCT) were investigated.

METHODS: The study was a prospective longitudinal study in which 240 calves from four purposively selected beef farms in the North Island of New Zealand were blood sampled and weighed in late spring, mid-summer and early autumn. Two farms were from high-risk (A and B) and two from low-risk (C and D) tick areas. Blood samples were analysed to determine HCT, and the number of T. orientalis Ikeda type organisms/µL of blood (infection intensity) using a quantitative PCR assay. A calf was defined as infected if >415 organisms/µL were detected in a blood sample. Linear mixed models were used to examine associations between infection intensity, mean daily liveweight gain (MDG), HCT, calf sex and time of sampling on the four farms.

RESULTS: On Farms A and B nearly all calves were infected at each sampling time, on Farm C <30% were infected at any sampling and on Farm D infection prevalence increased from 32 to 79% between late spring and early autumn. On Farms C and D, from mid-summer to early autumn, mean MDG was 0.127 (95% CI=0.072–0.183) kg/day less for infected than uninfected calves (p<0.001). On all farms MDG was negatively associated with infection intensity for mid-summer and early autumn sampling times (p=0.037). The relationship between time of sampling and infection intensity varied between farms (p<0.001), and between male and female calves (p=0.018). Females had a higher infection intensity than males at the mid-summer and early autumn samplings. The association between HCT and infection intensity varied with sampling time and farm (p=0.018). There was a strong negative association between infection intensity and HCT at the late spring sampling, but in mid-summer there was no association, and in early autumn only a weak association.

CONCLUSIONS AND CLINICAL RELEVANCE: This study has shown that beef farmers in the North Island of New Zealand should be concerned about the welfare effects and economic impacts of T. orientalis Ikeda type infection in suckled beef calves.  相似文献

AIM: To compare the ability of four strains of Streptococcus uberis at two doses to induce clinical mastitis in lactating dairy cows after intramammary inoculation in order to evaluate their usefulness for future experimental infection models.

MATERIALS AND METHODS: Four field strains of S. uberis (26LB, S418, and S523 and SR115) were obtained from cows with clinical mastitis in the Wairarapa and Waikato regions of New Zealand. Twenty-four crossbred lactating cows, with no history of mastitis and absence of major pathogens following culture of milk samples, were randomly allocated to four groups (one per strain) of six cows. Each cow was infused (Day 0) in one quarter with approximately 104 cfu and in the contralateral quarter with approximately 106 cfu of the same strain. The other two quarters remained unchallenged. All four quarters were then inspected for signs of clinical mastitis, by palpation and observation of the foremilk, twice daily from Days 0–9, and composite milk samples were collected from Days 0–8 for analysis of somatic cell counts (SCC). Quarters were treated with penicillin when clinical mastitis was observed. Duplicate milk samples were collected and cultured on presentation of each clinical case and on Day 4 from challenged quarters with no clinical signs.

RESULTS: Clinical mastitis was diagnosed in 26/48 (54%) challenged quarters. Challenge with strain S418 resulted in more cases of mastitis (12/12 quarters) than strains SR115 (7/12), 26LB (6/12) or S523 (1/12), and the mean interval from challenge to first diagnosis of mastitis was shorter for S418 than the other strains (p<0.001). The proportion of quarters from which S. uberis could be isolated after challenge was less for strain 26LB (1/6) than SR115 (6/7) (p<0.05), and SCC following challenge was lower for strain S523 than the other strains (p<0.05).

CONCLUSIONS: There were significant differences between the strains in the proportion of quarters developing clinical mastitis, the interval to mastitis onset, SCC following challenge and the proportion of clinical cases from which S. uberis could be isolated. These results illustrate the difference in the ability of S. uberis strains to cause mastitis and the severity of the infections caused.

CLINICAL RELEVANCE: Experimental challenge models can be used to compare infectivity and pathogenicity of different strains of mastitis-causing bacteria, the efficacy of pharmaceutical products and host-responses in a cost-effective manner.  相似文献
