[目的]筛选馕面团优质酵母菌.[方法]从新疆乌鲁木齐、阿尔泰,喀什3个样品点采集样品,采用PDA和YEPD培养基分离纯化了馕面团发酵酵母菌,并进行了26S rDNA D1/D2区域序列测定和系统发育分析.[结果]试验得出,29株菌相互之间的26S rDNAD1/D2基因序列同源性比例为99%.在这29株菌26S rDNA D1/D2区序列的基础上采用邻近法构建了系统发育进化树,从中选出了11株代表菌,它们被认为是该地区特殊而独特的酵母品系,完成了初筛.进一步测定11株菌的发酵活力、发酵前后的pH与面团醒发时间,完成了复筛并获得了6株菌.接着对6株酵母菌进行了感官测试,最后获得了一株优质酵母菌K24(KM454437),馕感官评分为97.1分.[结论]研究筛选出来的一株馕面团酵母菌发酵效率高,为新疆馕加工产业提供技术依据.  相似文献

利用PCR技术对从日本引进的Tos17插入水稻极限糊精酶基因变异体后代进行了插入位点鉴定和纯合体筛选分析。结果表明,利用Tos17末端2 6bpDNA序列和插入位点上下游各2 0 0bp和30 0bp处的目标基因序列片段设计引物,可以有效地进行Tos17插入突变纯合体筛选,为突变体的进一步利用提供了分子依据。  相似文献

Palm esters were synthesized through enzymatic transesterification of various palm oil fractions with oleyl alcohol using Lipozyme RM IM as the catalyst. At the optimized alcoholysis reaction condition, after 5 h reaction time all palm oil fractions exhibited a high percentage yields of esters (>80%). Simultaneous differential scanning calorimeter-thermal gravity analysis showed a high thermal stability profile of palm esters. Other physicochemical properties of palm esters such as refractive index, density, surface tension, slip melting point, saponification value, iodine value and acid value were analyzed following standard test methods modified from the American Oil Chemists’ Society standards. The dermal irritation assay of palm oil esters shows the non-irritancy of the esters with a Human Irritancy Equivalent (HIE) score below 0.9. Furthermore, an increase in skin hydration of 40.7% after 90 min after application in an acute moisturizing test, has proven the suitably of palm oil esters to be used in the cosmetics formulation.  相似文献

Summary Inheritance of spikelet number per ear and rate of spikelet initiation in wheat (Triticum aestivum L. em. Thell.) was studied in the land race spring wheats, 8–23 and 8–27 from Afghanistan, under controlled temperature and photoperiod. Spikelet number per ear was found to be under simple genetic control with dominance for high spikelet number. It is suggested that the gene determining spikelet number does so by determining the rate of spikelet initiation.  相似文献

ABSTRACT

1. Sex-specific variations in early embryonic development rates may pre-empt later variations in embryonic development through to pipping and hatching. Given that erythropoiesis (blood production) can be equated with early embryonic growth rate, it was hypothesised that blood pigment haemoglobin can act as a specific spectral fingerprint for changes in growth rate. Moreover, by measuring longitudinal, rather than lateral, spectral transmission through the egg, a more consistent spectrum with a higher signal-to-noise ratio could be captured.

2. Longitudinal visible transmission (T575/T598 ratio), which is sensitive to haemoglobin, was used to monitor sex-specific early embryonic development rate in white layer chicken eggs from d 0 to 8 of incubation. The sex of these eggs was subsequently confirmed two days after hatching.

3. Embryonic development was detectable from d 3 (72 h) of incubation, 36 h earlier than previously reported lateral spectral measurements, supporting the greater sensitivity of longitudinal measurements.

4. At d 3, the mean T575/T598 ratio for male embryos was significantly lower (P < 0.001) (i.e. higher absorbance of haemoglobin) than for female embryos, which was thought to be due to sex-differences in early embryogenesis. On the other hand, female embryos had a significantly lower (P < 0.05) mean T575/T598 ratio than male embryos at d 7 of incubation, presumably due to the combined effects of oestrogen synthesis receptors and enzymes on erythropoiesis in female embryos at this time.

5. In conclusion, the proposed methodology has the sensitivity to differentiate sex-specific embryonic development rates during early incubation and the potentiality to advance precision incubation management and poultry research.  相似文献

Abstract

AIM: To describe the seasonal pattern of development of third-stage infective larvae (L3) from eggs of Teladorsagia (=Ostertagia) circumcincta, Trichostrongylus colubriformis and Haemonchus contortus on pasture in the North Island of New Zealand.

METHODS: Sheep faeces containing known numbers of eggs of all three nematode species were deposited on, or buried in, pasture plots at three sites, viz coastal Manawatu, Upper Hutt Valley, and East Cape hill country. Development was measured by recovering L3 from faeces, herbage and soil 28–31 days after deposition on 13–18 occasions, between January 2005 and July 2006. Analysis of the number of larvae recovered used a mixed model including number of eggs deposited, weight of faeces recovered (an assumed indicator of earthworm activity), site, contamination date, and position of deposited faeces, i.e. on the surface or buried.

RESULTS: There was a significant effect of contamination date on development of all three species, with maximum numbers ofL3 developing between late spring (November) and early autumn (March), and minimum numbers in June and July. There were large differences between species, with H. contortus exhibiting a long period (April to October) where development was close to zero, whereas T. circumcincta developed to some extent all year round. Development of T. colubriformis was intermediate between the other two species.

Burying faeces containing nematode eggs increased the number of L3 recovered compared with surface deposition (p≤0.001), although there were a small number of exceptions involving only T. colubriformis. The weight of faeces recovered at harvest, which was assumed to be an indication of earthworm activity, was correlated with the number of L3 recovered for all species (p<0.001). In a separate analysis, earthworms were assumed tohave been active if <5 g faeces remained at harvest. Where this occurred, the number of L3 of T. colubriformis and T.circumcincta recovered was reduced by 56% and 58%, respectively (p<0.001).

CONCLUSIONS: A marked seasonal pattern of development was observed for all three species, with the most larvae developing in spring-early autumn and the least in winter. This seasonal pattern was most pronounced in H. contortus and least obvious in T. circumcincta. Burying faeces containing eggs generally resulted in more L3 being recovered, whilst the apparent activity of earthworms resulted in fewer larvae being recovered.  相似文献

Diversity of chemical composition, in vivo digestibility and in situ dry matter degradability of oat hay (Avena sativa) were studied. Oat hay samples obtained from 22 batches imported were analyzed for chemical composition. The result showed a wide range of variations in crude protein (3.9–8.8%, on a dry matter basis), crude fiber (22.1–35.2%), nitrogen free extract (50.0–66.0%), organic cellular contents (23.4–40.0%), organic cell wall (53.1–70.8%), organic a (4.1–9.8%), organic b (49.0–63.8%) fractions and the estimated total digestible nutrients (50.3–63.8%) from the organic components. Of these, four oat hay samples were used for the measurement of dry matter intake, in vivo digestibility and in situ dry matter degradability in sheep. Significant differences were observed for the dry matter intake, in vivo digestibility of dry matter, crude protein, crude fiber and nitrogen free extract, dry matter disappearance at 48 h and effective degradability at k = 0.05 h?1 of the samples (P < 0.05). The results revealed a wide range of diversity of nutritional quality of the imported oat hay that could have an affect on production potential in animals.  相似文献
