Chickpea can be seriously affected by Pea enation mosaic virus (PEMV) in the Pacific Northwest region of the USA and other areas of the world. Use of pesticides to manage PEMV vector transmission is ineffective and PEMV-resistant chickpeas have not been identified. Therefore, the Cicer core collection consisting of 499 wild Plant Introduction (PI) accessions and improved cultivars representing 25 countries and two chickpea phenotypes (Desi and Kabuli) was evaluated for resistance to PEMV based on a 1 to 5 scale where 1 = symptomless and 5 = severe mosaic and stunting. Eight accessions were considered resistant to PEMV based on mean disease severity values equal to or less than 1·9 in repeated greenhouse tests. All of the resistant accessions to PEMV were of the Desi type and all originated from Iran or India. PI 450763 was the only Kabuli accession that demonstrated tolerance to PEMV. PEMV resistance was not detected in commercial chickpea cultivars or advanced breeding lines currently grown in the USA and Canada. This is the first report of chickpea germplasm with resistance to PEMV.  相似文献

The G(M2) gangliosidoses are a group of lysosomal storage diseases caused by defects in the genes coding for the enzyme hexosaminidase or the G(M2) activator protein. Four Jacob sheep from the same farm were examined over a 3-year period for a progressive neurologic disease. Two lambs were 6-month-old intact males and 2 were 8-month-old females. Clinical findings included ataxia in all 4 limbs, proprioceptive deficits, and cortical blindness. At necropsy, the nervous system appeared grossly normal. Histologically, most neurons within the brain, spinal cord, and peripheral ganglia were enlarged, and the cytoplasm was distended by foamy to granular material that stained positively with Luxol fast blue and Sudan black B stains. Other neuropathologic findings included widespread astrocytosis, microgliosis, and scattered spheroids. Electron microscopy revealed membranous cytoplasmic bodies within the cytoplasm of neurons. Biochemical and molecular genetic studies confirmed the diagnosis of G(M2) gangliosidosis. This form of G(M2) gangliosidosis in Jacob sheep is very similar to human Tay-Sachs disease and is potentially a useful animal model.  相似文献

The Russian wheat aphid (RWA), Diuraphis noxia (Mordvilko), has become a perennial, serious pest of wheat (Triticum aestivum L.) in the western United States. Current methodologies used to enhance RWA resistance in wheat germplasm could benefit from an understanding of the biochemical mechanisms underlying resistance to RWA. This study was initiated to identify specific polypeptides induced by RWA feeding that may be associated with RWA resistance. The effects of RWA feeding on PI 140207 (a RWA-resistant spring wheat) and Pavon (a RWA-susceptible spring wheat) were examined by visualizing, silver-stained denatured leaf proteins separated by two-dimensional polyacrylamide gel electrophoresis. Comparisons of protein profiles of noninfested and RWA-infested Pavon and PI 140207 revealed a 24-kilodalton-protein complex selectively inhibited in Pavon that persisted in PI 140207during RWA attack. No other significant qualitative or quantitative differences were detected in RWA-induced alterations of protein profiles. These results suggest that RWA feeding selectively inhibit synthesis and accumulation of proteins necessary for normal metabolic functions in susceptible plants. Fossil zooplankton fecal pellets found in thinly bedded marine and lacustrine black shales associated with phosphate, oil, and coal deposits, link the deposition of organic matter and biologically associated minerals with planktonic ecosystems. The black shales were probably formed in the anoxic basins of coastal marine waters, inland seas, and rift valley lakes where high productivity was supported by runoff, upwelling, and outwelling.  相似文献

Management of fertilizer phosphorus (P) is a critical component of potato production systems as potato has a relatively high P requirement and inefficiently uses soil P. Phosphorus promotes rapid canopy development, root cell division, tuber set, and starch synthesis. Adequate P is essential for optimizing tuber yield, solids content, nutritional quality, and resistance to some diseases. Although soil test P is the primary tool for assessing P fertilizer needs, in some areas petiole P analysis has been successfully utilized to guide in-season P applications. Potato has been shown in some studies to respond to fertilizer P at soil test levels considered very high for most other crops (100+ mg kg?1 Bray P1 or Mehlich I or III and 20+ mg kg?1 sodium bicarbonate) especially on medium- to finer-textured soils. Even on high-testing soils, fertilizer P rates for top yields sometimes exceed 150 kg P2O5 ha?1. In addition, many states/provinces continue to recommend half or more of the amount of P in the harvested portion of the crop irrespective of soil test P level. In most situations, few differences are expected among fertilizer P sources; however, high rates of diammonium phosphate (DAP) or urea-phosphate (UAP) should not be band-applied in contact or near the seed piece. Most research determined that fertilizer P was most efficiently used when band-applied at planting (e.g., 5 cm to each side of the seed piece); however, some western USA work on high-pH soils showed increased yields and petiole P levels with preplant broadcast applications. In-season applications with the irrigation water can be successful when the potato roots are sufficiently close to the soil surface; however, most research indicates that P applications are more effective when applied at planting or early in the season. Potato fertilizer phosphorus best management practices include: (1) apply the fertilizer P rate calibrated for local soils; (2) band-apply fertilizer P at least 5 cm from the seed piece, especially on very sandy soils or where DAP or UAP are used; (3) use petiole P tests to determine the need for in-season applications; (4) account for all P sources applied, including animal manures; and (5) utilize the best soil conservation practices to reduce P losses to surface waters.  相似文献
