Deoxynivalenol(DON)is considered to be the most harmful mycotoxin that affects the intestinal health of animals and humans.Phenethyl isothiocyanate(PEITC)in feedstuff is an anti-nutritional factor and impairs nutrient digestion and absorption in the animal intestinal.In the current study,we aimed to explore the effects of PEITC on DON-induced apoptosis,intestinal tight junction disorder,and its potential molecular mechanism in the porcine jejunum epithelial cell line(IPEC-J2).Our results indicated that PEITC treatment markedly alleviated DON-induced cytotoxicity,decreasing the apoptotic cell percentage and pro-apoptotic mRNA/protein levels,and increasing zonula occludens-1(ZO-1),occludin and claudin-1 mRNA/protein expression.Meanwhile,PEITC treatment ameliorated DON-induced an increase of the inducible nitric oxide synthase(iNOS)and cyclooxygenase 2(COX-2)mRNA levels and intracellular reactive oxygen species(ROS)level,and a decrease of glutathione peroxidase 1(GPx1),superoxide dismutase 2(SOD2),catalase(CAT)and heme oxygenase 1(HO-1)mRNA levels.Additionally,PEITC treatment significantly down-regulated autophagy-related protein 5(ATG5),beclin-1 and microtubuleassociated protein 1 light chain 3B(LC3-II)mRNA/protein levels,decreased the number of green fluorescent protein-microtubule-associated protein 1 light-chain 3(GFP-LC3)puncta and phosphatidylinositol 3 kinase(PI3K)protein expression,and up-regulated phospho-protein kinase B(p-Akt)and phospho-mammalian target of rapamycin(p-mTOR)protein expression against DON.However,the activation of autophagy by rapamycin,an autophagy agonist,abolished the protective effects of PEITC against DON-induced cytotoxicity,apoptosis and intestinal tight junction disorder.Collectively,PEITC could confer protection against DON-induced porcine intestinal epithelial cell injury by suppressing ROSmediated autophagy.  相似文献

两种杂交石斑鱼子一代杂种优势的微卫星标记分析   总被引:2,自引:3,他引:2  
使用6对微卫星引物对两种杂交子一代(青龙斑和虎龙斑)及其亲本(斜带石斑鱼、棕点石斑鱼和鞍带石斑鱼)共5个群体进行微卫星分析,计算等位基因频率、有效等位基因数、基因杂合度、多态信息含量、相似指数和Nei氏遗传距离。结果显示,在5种石斑鱼中,棕点石斑鱼的平均有效等位基因数最大(6.849 3),最小的是鞍带石斑鱼(2.608 6)。6对微卫星引物在这5个群体中具有丰富的多态性,平均多态信息含量(PIC)分别为0.666 7、0.751 3、0.441 9、0.664 0、0.542 6。在平均观测杂合度(Ho)中,虎龙斑和青龙斑的最高均为0.940 0,鞍带石斑鱼的最低为0.508 3。遗传距离和遗传相似率结果显示,青龙斑和虎龙斑均与父本鞍带石斑鱼的亲缘关系较近。杂交子一代群体的等位基因基本来自父母本群体双方,可推断杂交子一代的遗传物质来自父母双方,属两性融合生殖,是真正意义上的杂交种。杂交后代的遗传变异水平明显增强,这是杂种优势得以形成的重要遗传物质基础。  相似文献

Marine fisheries target and catch fish both for direct human consumption (DHC) as well as for fishmeal and fish oil, and other products. We derived the fractions used for each for 1950–2010 by fishing country, and thus provide a factual foundation for discussions of the optimal use of fisheries resources. From 1950 to 2010, 27% (~20 million tonnes annually) of globally reconstructed marine fisheries landings were destined for uses other than DHC. Importantly, 90% of fish destined for uses other than DHC are food‐grade or prime food‐grade fish, while fish without a ready market for DHC make up a much smaller proportion. These findings have implications for how we are using fish to feed ourselves or, more appropriately, how we are not using fish to feed ourselves.  相似文献

Thyroxin (T4) is proven to have various functions in fish reproduction. In this study, we investigated oocyte maturation and spawning performance in the rabbit fish, Siganus guttatus, orally exposed to different doses of T4 (0.05, 0.25, and 0.5 mg/kg fish) during vitellogenesis. No statistically significant differences of reproductive characteristics were observed among the treatments. However, the group that was exposed to the dose of 0.5 mg/kg spawned after receiving a hormone injection. The findings suggest that feeding T4 during vitellogenesis did not advance oocyte maturation, but probably could induce spawning in rabbit fish. In a separate experiment, we investigated the effects of orally administered domperidone (DOM) on reproductive performances of the female rabbit fish. The study showed that oral administration of DOM during the early stages of the reproductive cycle had no significant effects on oocyte maturation and spawning performance. Overall, these findings suggest that DOM may not be needed for the induction of maturation and spawning of this species under aquaculture conditions. These practical investigations were conducted with a view to attempt reducing drug use, production costs, and support sustainable aquaculture.  相似文献

In order to understand the stability of blue-tongue virus (BTV)storage in cold room for long time, 18 tissue samples which were kept in 4℃ for 10 to 15 years were test by Real-time PCR, and 10 blood samples which were kept in 4℃ for 15 years were subjected to virus isolation process, then was proved by monoclonal antibody staining,research results showed all old samples were positive in Real-time PCR, further more, 5 live virus were isolated from 10 old blood samples. The biochemistry of intermuscular variation in tenderness is not fully understood. To investigate the role of the calpains in this process we performed two experiments using bovine and ovine species. In the bovine experiment, two distinct muscles, longissimus thoracis et lumborum (LT) and psoas major (PM), were used. In the ovine experiment, four muscles, LT, PM, semimembranosus (SM), and semitendinosus (ST), were used. Muscles were sampled at death for the determination of the steady-state mRNA level of calpains and calpastatin and the activities of calpain 1, 2, and calpastatin. Muscles were also sampled to determine the temporal changes in pH, tenderness, and the activity of the ubiquitous calpain system during postmortem aging. The results of the relative rate of tenderization in both species was found to be related to muscle type; LT had the highest value in both species. Within species, the mRNA steady-state levels of calpain 1 and calpastatin were similar in various bovine and ovine muscles. Bovine calpain 2 mRNA level was significantly lower in the LT than in the PM. Ovine calpain 2 mRNA level was lower, but not significantly different, in the LT compared to the other muscles. The mRNA level of bovine calpain 3 was significantly higher in the LT muscle than in the PM. In the ovine, the mRNA level of calpain 3 was highest in the LT, followed by SM, PM, and ST. Results on the activity of the ubiquitous calpain system in various muscles at death were dependent on muscle type and species. Temporal changes in the activity of calpains and calpastatin during the first 24 h of postmortem aging were similar in the muscles studied: calpain 1 and calpastatin declined significantly and calpain 2 remained relatively unchanged. The temporal changes in muscle pH in both experiments indicated that the extent and rate of pH decline during aging was related to muscle type. Correlation analysis between the relative rate of tenderization and mRNA expression of calpains revealed a strong relationship with calpain 3 in both species.  相似文献

The frequency, distribution and repeatability of measurements related to infection with gastrointestinal nematodes have been estimated in dairy goats. Monthly faecal and blood samples were taken over two grazing seasons from a flock of 120 naturally infected goats. The frequency distribution of egg excretion was positively skewed at each sampling date, suggesting an aggregative distribution of parasites. Overdispersed distributions were also apparent for pepsinogen and inorganic phosphate values. Repeatabilities for each parameter were estimated within each year. The values ranged between 0.23 to 0.43 and were relatively similar during the 2 years, with the highest coefficients recorded for pepsinogen. For each parameter, moderate to high repeatabilities were also estimated between the 2 years. Overall, the data suggest that, in dairy goats, the distribution of worms is overdispersed with a few individuals repeatedly harbouring large worm populations. These results have practical implications for genetic selection for resistance to worm infection and for selective administration of anthelmintics.  相似文献
