The capacity for water diffusion in burrow walls (i.e. the coefficient of sorptivity) either burrowed by Lumbricus terrestris (T-Worm) or artificially created (T-Artificial) was studied through an experimental design in a 2D terrarium. In addition, the soil density of earthworm casts, burrow walls (0–3 mm around the burrow) and the surrounding soil (>3 mm) were measured using the method of petroleum immersion. This study demonstrated that the quantity of water which transits through burrows of L. terrestris in the soil matrix was lower than that transited through soil fractures, due to a reduction of soil porosity in burrow walls (compaction: cast > worms burrow walls > surrounding soil > artificial burrow walls). Earthworm behaviour, in particular burrow reuse with associated cast pressing on walls, could explain the larger burrow wall compaction in earthworm burrows. If water diffusion was lower through the compacted burrows, burrow reuse by the worms makes them more stable (worms would maintain the structure over years) than unused burrows. The present experimental design could be used to test and measure the specific differences between earthworm species in their contributions to water diffusion. Probably, these contributions depend on the presumed related-species behaviours which would determine the degree of burrow wall compaction.  相似文献

The uptake and metabolism of [14C]-2,4-dichlorophenol (DCP) and [14C]-2,4-dichloroaniline (DCA) were investigated in wheat and soybean. Seeds were exposed to a nutrient solution containing 50 microM of one of two radiolabeled compounds, and plant organs were harvested separately after 18 days of growth. In wheat, uptake of [14C]-2,4-DCP was 16.67 +/- 2.65 and 15.50 +/- 2.60% of [14C]-2,4-DCA. In soybean, uptake of [14C]-2,4-DCP was significantly higher than [14C]-2,4-DCA uptake, 38.39 +/- 2.56 and 18.98 +/- 1.64%, respectively. In the case of [14C]-2,4-DCP, the radioactivity absorbed by both species was found mainly associated with roots, whereas [14C]-2,4-DCA and related metabolites were associated with aerial parts, especially in soybean. In wheat, nonextractable residues represented 7.8 and 8.7% of the applied radioactivity in the case of [14C]-2,4-DCP and [14C]-2,4-DCA, respectively. In soybean, nonextractable residues amounted to 11.8 and 5.8% of the total radioactivity for [14C]-2,4-DCP and [14C]-2,4-DCA, respectively. In wheat, nonextractable residues were nearly equivalent to extractable residues for [14C]-2,4-DCP, whereas they were greater for [14C]-2,4-DCA. In soybean, the amount of extractable residues was significantly greater for both chemicals. However, in both species, nonextractable residues were mainly associated with roots. Isolation of soluble residues was next undertaken using excised shoots (wheat) or excised fully expanded leaves including petioles (soybean). Identification of metabolite structures was made by comparison with authentic standards, by enzymatic hydrolyses, and by electrospray ionization-mass spectrometric analyses. Both plant species shared a common metabolism for [14C]-2,4-DCP and [14C]-2,4-DCA since the malonylated glucoside conjugates were found as the final major metabolites.  相似文献

Real-time quantitative PCR (RT-qPCR) is a critical tool used to evaluate changes in gene expression. The precision of this tool is reliant upon the selection of reference genes whose expression remains unaltered in culture conditions and following stimulation. Stably expressed reference genes are used to normalize data so observed changes in expression are not due to artifacts but rather reflect physiological changes. In this study, we examined the expression stability of the porcine genes glyceraldehyde 3-phosphate dehydrogenase (GAPDH), succinate dehydrogenase complex subunit A (SDHA), eukaryotic elongation factor 1 gamma-like protein (eEF1), ribosomal protein L19 (RPL19), beta-actin (ACTB) and ATP synthase mitochondrial F0 complex (ATP5G1) in peripheral blood mononuclear cells (PBMCs), monocytes, monocyte-derived dendritic cells (MoDCs), blood isolated dendritic cells (BDCs) and T cells with or without stimulation with lipolysaccharide (LPS). An M value was used as a measure of gene stability as determined using geNORM software. Recommendations for the use of reference genes include using GAPDH and B-actin in PBMCs: RPL19 and SDHA in T cells; RPL19 and B-actin in monocytes; RPL-19 and SDHA in BDCs: and RPL-19 and ATP5GA in MoDCs.  相似文献

The anorexic or dysphagic adult horse often requires nutritional support. Providing nutrients by the enteral route is the safest and most economic choice, but the dietary options available for use in horses are somewhat limited. The objective of this study was to compare the physiologic response of normal horses with a low-residue liquid or normal diet over a 10-day feeding period. Two groups of 6 normal adult horses were maintained on 1 of 2 diets for a 10-day period. Diets were formulated to meet the caloric needs of a horse maintained in a stall. The control group was fed 70% timothy hay and 30% textured concentrate for the test period, and the experimental group received the low-residue liquid diet, similar to liquid nutritional formulas designed for human use. Clinical parameters, body weight, packed cell volume, total plasma solids, blood glucose, and serum electrolytes were recorded daily for each horse during the dietary trial period. On days 1, 5, and 10 of the study, a complete blood count, serum biochemical profile, and urinalysis were performed.Horses' serum total bilirubin concentration and pattern of weight loss differed between groups. All other physical parameters, blood chemistry, complete blood count, and urinalysis results remained within the normal reference interval for the horses regardless of diet, although some statistical differences were observed. Horses returned to pasture and free-choice grass diet without complications at the end of the dietary trial period. These results demonstrate that few differences of biologic significance were observed between horses being fed low-residue diet and horses receiving a normal diet of hay and grain over a 10-day period. Summary

Tree growth and water status throughout the growing season and after fruit removal were studied in container-grown peach trees. Trees with fruit (F) and defruited (DF) trees were sampled destructively at bud break (8 March), 1 month after fruit removal (3 June), at harvest (6 August), and before leaf fall (15 October) to determine the mass of leaves, current season shoots, branches, trunk, and the entire root system. Tree water status was determined from the mid-day stem water potential (SWP) the day before each sampling date. Root growth in DF trees was greater than that observed in F trees, while the above-ground biomass was similar in DF and F trees. DF trees therefore had lower leaf:root biomass ratios than F trees throughout the fruit growing season. Environmental factors did not fully explain the seasonal variations in SWP, but there was a significant correlation between leaf:root biomass ratios and SWP. Reductions in leaf:root biomass ratios were accompanied by increases in SWP and, ultimately, DF trees had higher SWP values than F trees in mid-Summer. Improvements in tree water status following fruit removal can be explained, in part, by additional root growth.  相似文献

Of the 13 genes described as affecting fruit color in Cucurbita pepo, one, D (Dark stem), also markedly affects stem color. The D allele confers dark stems and dark intermediate-age fruits, is dominant to the d allele for light stems and fruits, and epistatic to two recessive genes conferring light fruit coloration, l-1 and l-2. However, a gene for light fruit coloration, W (Weak fruit color), is epistatic to D in the fruits. We observed variation for stem color in a scallop squash cultivar having light-colored fruits, some plants having dark stems and others light stems. Two true-breeding inbreds of this cultivar, one having dark stems and the other light stems, were developed and, when these inbreds were crossed, the progeny had light stems. In order to elucidate the genetic basis of the dominant light-stem characteristic, these two inbreds were crossed with two near-isogenic tester lines, one of genotype D/D l-1/l-1 l-2/l-2 and the other of genotype d/d l-1/l-1 l-2/l-2. Also, the dominant light stem and light fruit color were introgressed into a third near-isogenic line of genotype D/D l-1/l-1 l-2/l-2, resulting in two new near-isogenic lines, and these lines were then intercrossed. The results showed that the variation in stem color of the scallop squash cultivar derives from segregation of alleles at the W locus, with a newly designated top-dominant allele, W S , conferring both, light stems and light fruits. In an attempt to transfer the Lr56/Yr38 resistance loci from Aegilops sharonensis to wheat, a 6A‐6Ssh chromosome translocation was produced. It involves essentially the entire chromosome 6Ssh with a small terminal segment of 6AL. Induced homoeologous recombination of the translocated chromosome with 6A produced numerous recombinants including three recombined chromosomes carrying Lr56 that could not be precisely mapped for lack of suitable markers. This study aimed to determine the chromosomal locations of the translocation breakpoints in these three recombinants using various DNA markers as well as physical and genetic mapping. The three recombinants Lr56‐39, ‐157 and ‐175 carry small segments of Ae. sharonensis chromatin distally to the Xgpw4329 and IWA5416 loci near the 6AS telomere. The Ae. sharonensis chromatin that remains in each line includes a homoeolocus of the wheat marker locus Xdupw217 (on 6BS) and its characteristic amplification product can be used as a dominant marker for the presence of Lr56. Of the three recombined chromosomes, Lr56‐157 retained the least alien chromatin and appears to be the best candidate for use in wheat breeding.  相似文献
