Relationship Between Dynamic and Static Value of Elastic Modulus in Rock

Taking the deposited-layer rock and compacted-layer rock as study target,by elastic wave experiment and uniaxial compression experiment,the physics mechanics parameters and acoustics parameters of these rock are worked out; Based on it, by the correlativity is established between dynamic and static value of elastic modulus in rock of different lithology taking nonlinear fitting,it offers reference for the study of the drill and measuring well in the exploitation of oil. Influenced by the integrality and weathering breakage degree of rock,the biggish different is existed between dynamic elastic modulus gained by dynamical method and static elastic modulus obtained by static method,and the relationship between dynamic and static value of elastic modulus is approximatively expressed as a relationship of quadratic polynomial function.

豆天蛾核型多角体病毒F蛋白基因的克隆和序列分析

从豆天蛾幼虫虫体中分离纯化出一种新型的豆天蛾核型多角体病毒(CbNPV),提取基因组DNA,进行基因组测序。结果发现一个与杆状病毒F蛋白基因同源性很高的读码框序列,该长度为2 109 bp,编码702个氨基酸。氨基酸序列同源性分析表明:CbNPV F蛋白与II类NPV和I类NPV F蛋白的同源性分别为32.9%~61.8%和8.0%~16.1%,并且将它与多种I类NPV杆状病毒同功能蛋白GP64进行分析,同源性在9.1%~11.4%,表明CbNPV F蛋白与I类NPV的F及GP64蛋白的同源性很低,因此认为CbNPV属于II类NPV。     

产肠毒素性大肠杆菌K88与K99菌毛蛋白的串联表达

根据GenBank中发表的E.coli K88、K99基因序列,分别设计合成1对引物.利用PCR技术,以大肠杆菌C83907和C83644的质粒为模板分别扩增不含信号肽的K88及K99基因.通过分离、纯化、限制性核酸内切酶酶切,连接和转化,构建了含K88-K99串联表达载体的重组菌株BL21(DE3)(pETK88CK99).结果显示,经酶切,PCR鉴定和DNA序列分析,证实了构建的重组质粒pETK88CK99中含有K88K99融合基因,且基因序列和阅读框架均正确.经过SDS-PAGE分析,串联表达蛋白含量占菌体蛋白的40%左右,经Western blotting检测,该串联表达蛋白能被大肠杆菌K88、K99标准血清识别.结果表明,构建的重组菌株可以作为预防新生仔猪大肠杆菌性腹泻基因工程疫苗的候选菌株.     

鸭Ⅰ型肝炎病毒荧光定量RT-PCR方法建立及初步应用

根据GenBank中鸭Ⅰ型肝炎病毒的基因序列,设计合成一对引物和一条Taq-Man探针。进行优化后,建立了能够检测鸭Ⅰ型肝炎病毒的荧光定量RT-PCR方法。该方法敏感性好,检测敏感性达到20个模板拷贝数,比常规PCR灵敏度高100倍;该方法特异性强,对番鸭细小病毒、鸭圆环病毒和鹅细小病毒等6种病原体的检测全为阴性。对广西地区鸭群收集的118份病料进行检测,结果阳性率为1.69%。结果提示:在广西地区的鸭群中,存在鸭Ⅰ型肝炎病毒的感染,建立的荧光定量RT-PCR方法可用于鸭Ⅰ型肝炎病毒的临床快速检测。     

Differentiation of embryonic-like stem cells derived from human bone marrow into multinucleated fibers in vitro

AIM: To compare the capacity of in vitro differentiation into multinucleated fibers between embryonic-like stem cells (ELSCs) and mesenchymal stem cells (MSCs) derived from human bone marrow. METHODS: To isolate ELSCs, human bone marrow mononuclear cells were cultured in gelatin-coated flask with serum-free Knockout-DMEM medium designed for the expansion of human embryonic stem cells. MSCs were isolated from the same bone marrow by the traditional method. The morphological characters of both ELSCs and MSCs were observed under inverted phase-contrast microscope, and the expression of their multipotent antigen markers was identified by immunofluorescent staining. ELSCs and MSCs were cultured in myogenic differentiation medium. The protein levels of muscle-specific antigen markers myosin heavy chain (MHC), myogenin and MyoD were detected by the method of immunostaining. The mRNA expression of MHC, myogenin and MyoD was detected by RT-PCR. The capacity of in vitro differentiation into multinucleated fibers was compared between ELSCs and MSCs by calculating the proportion of MHC-positive multinucleated fibers. RESULTS: ELSCs, which weakly expressed the multipotential markers Oct-4, Nanog-3 and Sox-2, were isolated from bone marrow by the method of serum-free medium. ELSCs appeared smaller, slenderer and more homogeneous, and were morphologically different from MSCs derived from the same marrow. No multipotential marker in MSCs was expressed. ELSCs and MSCs were induced into long multinucleated fibers expressing MHC and myogenin at mRNA and protein levels by culturing in the myogenic differentiation medium. However, on the 10th day after induction, the proportion of the MHC-positive fibers in ELSCs was (25.7?4.1)%, and the proportion in MSCs was (15.8?7.6)%.The capacity for differentiation into muscle in ELSCs was significantly higher than that in MSCs (P<0.05). CONCLUSION: Bone marrow ELSCs are induced into multinucleated fibers and have the stronger myogenic differentiation capacity than MSCs derived from the same marrow. ELSCs are a more ideal candidate for muscular disease therapy.     

水稻丙酮酸激酶基因OsPK1的过表达转基因株系的获得与鉴定

利用农杆菌介导的转化方法研究OsPK1过表达对水稻的影响。将CaMV 35S启动子驱动的OsPK1的全长CDS序列导入到日本晴基因组中。选择3个过表达转基因系作为代表进行鉴定和分析。结果显示,过表达植株在株高、分蘖数、穗长和千粒重四方面接近野生型水稻,结实率略下降,但每穗饱满种子数明显增加。通过定量RT-PCR分析,发现有4个代谢酶在过表达植株嫩叶中表达存在着上调或下调。用GC-MS方法检测糖含量,显示OsPK1过表达株系中葡萄糖、果糖、半乳糖和蔗糖的含量与野生型差异不大。总之,对OsPK1过表达株系的鉴定,有助于更好了解OsPK1的功能,并且每穗饱满种子数明显增加具潜在的应用价值。     

朝鲜鹌鹑黑素皮质素受体1基因多态性及其在不同羽色个体中的表达

通过混合样品DNA测序方法寻找黑素皮质素受体l（MC1R）基因的突变位点,采用Alu1-RFLP对突变位点在4种羽色（栗羽、黄羽、白羽、黑羽）鹌鹑群体中的基因分布进行了研究;利用qRT-PCR技术测定了MC1R基因在12日龄时4种羽色鹌鹑胚胎皮肤组织中的表达情况。结果表明,在鹌鹑MC1R基因上发现1个T/C突变位点,该位点没有导致编码蛋白氨基酸序列改变,A1u1-RFLP分析发现,该突变位点的不同基因型在4种羽色鹌鹑群体间的分布有显著差异（P〈0.05）。4种羽色鹌鹑皮肤组织中MC1R基因的表达量存在明显差异,栗羽鹌鹑皮肤组织中该基因的表达量明显高于黑羽鹌鹑皮肤中的表达量（栗羽〉黄羽〉白羽〉黑羽）。本试验没有发现导致日本鹌鹑黑羽突变的Glu92Lys突变位点,表明朝鲜鹌鹑的黑羽突变与报道的日本鹌鹑黑羽突变的机制不同,朝鲜鹌鹑的黑羽可能与其他基因的突变有关。     

Preparation of Activated Carbon from Cotton Stalk Using Potassium Hydroxide as Activation Agent

以棉花秸秆为原料,采用KOH活化法制备活性炭样品,探讨了炭化、活化及后处理过程中各实验条件对活性炭样品性能的影响.综合考虑活性炭样品的性能及得率,得出较优的实验条件为:炭化温度450～500℃、碱炭比值1.0、活化温度800℃、活化时间120 min;在较优条件下制得活性碳的比表面积2 312m2/g,碘吸附值1 936 mg/g,亚甲基蓝吸附值392 mg,/g;孔径分布以微孔为主;表面含有羟基(-OH)、活泼氢(-H)等基团.
