Type D retrovirus was isolated from rhesus macaques with simian acquired immunodeficiency syndrome (SAIDS) and transmitted to healthy rhesus macaques with tissue culture medium containing the virus. The clinical, immunologic, and lymph node morphologic changes were observed in 9 rhesus macaques for 52 weeks after inoculation. A spectrum of clinical signs developed including early death, persistent SAIDS, and apparent remission. Animals that died or developed persistent SAIDS had characteristic lymphoid depletion, persistently depressed peripheral blood mononuclear cell (PBMC) mitogenic response, and decreased serum immunoglobulins. The SAIDS retrovirus (SRV) was recovered from PBMC of 8 of the animals after inoculation. Virus could not be recovered from PBMC of one animal in remission, but this animal developed serum-neutralizing antibodies to SRV after inoculation. Seven of the animals seroconverted to SRV after inoculation, all 9 were seronegative for human T-lymphotropic virus-III, and 5 animals tested were seronegative to human T-lymphotropic virus-I. These findings support the etiologic role of the type D retrovirus in SAIDS and further define the pathogenesis of this disease.  相似文献

Cockle, a defect of lamb's pelts, has recently been shown to be associated with infestation of lambs with the louse Bovicoh ovis. The mechanisms by which infestation with the louse leads to cockle are not known, but immunological hypersensitivity has been suggested. The objectives of our work are to determine if lambs respond immunologically to lice and, if so, whether hypersensitivity occurs. Preliminary results are presented here.  相似文献

Six livers from one-year-old sheep were each sampled at 27 different sites and the concentrations of copper, vitamin B,, and zinc determined for each sample. Using a crossover design, the effects of the addition of ketamine to a previously determined optimal hand-injected immobilization dosage of carfentanil/xylazine were evaluated in 11 adult white-tailed deer (Odocoileus virginianus). Two i.m. ketamine dosages were evaluated: 0.15 mg/kg (low ketamine) and 0.30 mg/kg (high ketamine). Each deer was immobilized twice 2 wk apart. Inductions were video recorded and reviewed by observers, who had been blinded to drugs and dosages, who rated qualitative aspects. There were significant (P < 0.05) dosage-dependent decreases in heart rate, SaO2, and arterial pH, and a significant dosage-dependent increase in PaCO2. Induction times with both dosages were more rapid (mean 2.3 +/- 0.9 min for low ketamine and 2.3 +/- 0.6 min for high ketamine) than those reported for the same carfentanil/xylazine dosage used without ketamine. Mean quality ratings, though improved compared to those reported for carfentanil/xylazine alone, were considered "undesirable" for both dosages. Hyperthermia (temperature > 41 degrees C) was noted in 13 of 22 immobilizations. Arterial pH and PaO2 increased significantly from 10 to 20 min postrecumbency, but acidemia (pH < 7.3) was present throughout immobilization periods for all deer. There were ketamine dosage-dependent increases in respiratory components of this acidemia compared with that associated with carfentanil/xylazine alone. Possible hypoxemia was present at both sampling times for both groups, while hypercapnea (PaCO2 > 60 mm Hg) was present for the high-ketamine group only. Reversal times for naltrexone and yohimbine were rapid (mean 2.9 +/- 0.7 min for low ketamine and 3.3 +/- 0.8 min for high ketamine), with no evidence of renarcotization. Although the addition of ketamine to carfentanil/xylazine caused faster inductions and improved induction qualities, it also produced an increased incidence of hyperthermia, acidemia, hypoxemia, and hypercapnea. Supplemental oxygen and close monitoring of body temperature is recommended when using this immobilization regimen.  相似文献

The objectives of the present study were to investigate the bacteria accompanying hydrosalpinx of the buffalo cow and investigate the correlation between bacterial infection of the uterus and hydrosalpinx. Buffalo cows’ reproductive tracts were collected from Mosul abattoir. A total 385 uterine samples were examined of which 25 were having hydrosalpinx. Swabs for bacteriology, fluid for cytology and biopsies for histopathology were collected from the hydrosalpinx and the uterus from each samples included in this study. Results of this study indicated high prevalence of hydrosalpinx (6.5%) including unilateral (n = 19; 76%) and bilateral (n = 6; 24%) hydrosalpinx. Although 16 samples (64%) of the hydrosalpinx samples had no bacterial growth, the most prevalent bacteria recovered from hydrosalpinx were Corynebacterium hemolyticum and Actinomyces bovis, 42.8% and 28.6%, respectively. The most prevalent bacteria in the uterus were Archanobacterium pyogenes (18.5%), Staphylococcus aureus (14.8%), and Listeria monocytogenes (11.0%). Higher rates of leukocytes infiltration (p < 0.01) were observed in the uterine discharge than hydrosalpinx. A significant (p < 0.01) increase in lymphocytes was found in uterine discharge. Microscopic examination of the hydrosalpinx showed mucosal atrophy and dilatation of oviductal lumen without any signs of inflammation. It could be concluded that there is no correlation between bacteria isolated from uterus and hydrosalpinx. No association was found between bacteriological cultures and hydrosalpinx. Inflammation of the uterine tissue could be extended to utero‐tubal junction producing local inflammation resulting in fibrosis and tubal obstruction. The obstruction in the lumen of the oviducts resulted in accumulation of fluid.  相似文献

Footrot disease due to Nectria haematococca (anamorph Fusarium solani f.sp. pisi) is an economically important disease of peas globally. However, our ability to predict accurately the likelihood of footrot infections is limited because there is no method to determine inoculum density prior to planting. In this research, a PCR-based assay was developed to quantify the pea pathogenicity gene (PEP3), exclusive to highly pathogenic forms of N. haematococca, from DNA extracted from agricultural field soils. The applicability of using quantitative PCR (qPCR) to measure this gene in soil was validated, and the relationship between PEP3 gene numbers and footrot disease was also studied. Results showed that the quantitative assay is both efficient and specific; amplification efficiency of the Q-PCR assay for the PEP3 gene was 92%. Gene copy numbers were shown to vary significantly (P = 0.01) between fields, and were positively correlated to the number of spores of pathogenic N. haematococca, and to footrot disease. PEP3 numbers of up to 100 g−1 soil constituted a threshold number for infection—potentially capable of causing economically significant pea footrot disease. The density of virulent N. haematococca in soil fields capable of causing footrot disease could be determined with a high degree of accuracy, with this assay. It offers the opportunity for prediction of pea footrot infections in agricultural soils prior to cultivation.  相似文献
