OBJECTIVE: To compare function of cultured cryopreserved stallion spermatozoa in a modified Tyrode's medium (TM), with or without bovine serum albumin (BSA), or in uterine tube (oviduct) epithelial cell (OEC) coculture in TM, with or without BSA. SAMPLE POPULATION: Cryopreserved spermatozoa from 6 proven stallions and OEC from bovine reproductive tracts in follicular phase. PROCEDURE: Thawed spermatozoa were cultured in TM, with or without BSA, or cocultured with OEC monolayers in TM, with or without BSA. Percentages of capacitated and acrosome-reacted spermatozoa were measured at 5 hours for TM cultures. Spermatozoal survival and motility characteristics were observed over time for all culture methods. Number of spermatozoa attaching to OEC were compared for cocultures. RESULTS: Use of TM without BSA altered spermatozoal function in cell-free medium culture and OEC coculture. A higher percentage of spermatozoa were acrosome reacted in TM with BSA, although percentages of capacitated spermatozoa did not differ. Spermatozoa survived longer and maintained superior motion in TM culture without BSA and in OEC cocultures. More spermatozoa were able to attach to OEC in TM without BSA. CONCLUSIONS: Incubation of cryopreserved spermatozoa in media with BSA resulted in rapid decrease in percentage of intact, motile spermatozoa and limited their ability to interact with OEC. CLINICAL RELEVANCE: Current culture media used for assisted reproduction techniques in horses do not provide functionally capacitated spermatozoa. A meta-analysis was performed with the aim to identify factors with a relevant influence on the expression of clinical postweaning multisystemic wasting syndrome (PMWS) under experimental conditions. Data from 44 studies were included in the analysis. Several variables were studied: number of pigs in the experiment, intake of colostrum, serological status against porcine circovirus type 2 (PCV2), strain of PCV2 used for inoculation, the route and dose of inoculation, and use of potential triggering factors (such as co-infections, vaccinations, or immunomodulator products). Multiple correspondence analysis and log-linear regression methods were used to establish the relationships between the studied variables and the number of PCV2 infected pigs that developed PMWS. Based on the results of the meta-analysis, the most successful animal experiment aimed to develop PMWS should include: (1) colostrum-deprived pigs, (2) age of inoculation below 3 weeks, (3) high doses of PCV2 inoculum, (4) PCV2 strain from genotype 1, and (5) co-infection with another swine pathogen as a triggering factor. 相似文献

Streptococcus uberis is an important environmental pathogen that causes subclinical and clinical mastitis in lactating and nonlactating cows throughout the world. S. uberis adhesion molecule (SUAM) was identified recently by our laboratory and we hypothesize that SUAM is a potential virulence factor involved in the pathogenesis of S. uberis mastitis. The first objective of the present study was to clone and sequence the SUAM gene (sua) from S. uberis UT888. The second objective was to determine the prevalence of sua in strains of S. uberis isolated from geographically diverse locations. The 20 amino acid N-terminal sequence of purified SUAM was utilized to identify a single open reading frame (ORF) in the S. uberis O140J (ATCC BAA-854) genome database. Three sets of primers were identified from this sequence for amplification of sub-fragments and the complete gene encoding SUAM. Restriction fragment analysis of the largest polymerase chain reaction (PCR) product confirmed the desired fragment had been amplified. This 2970bp PCR fragment was cloned into plasmid pCR-XL-TOPO and sequenced. The S. uberis UT888 sua sequence (NCBI Accession no. DQ232760) was 99% similar to the S. uberis O140J database sequence. The three pairs of PCR primers were used in a subsequent experiment to identify sua in 12 strains of S. uberis isolated in milk from dairy cows with mastitis in Tennessee (n=6), Colorado (n=1), Washington (n=1), New Zealand (n=1) and from the American Type Culture Collection (n=3). Primer pairs yielded the expected 2970, 2639 and 2362bp PCR fragments in all strains evaluated. In conclusion, we cloned and sequenced sua, which codes for the first described S. uberis adhesin, SUAM. sua was detected in all strains of S. uberis evaluated suggesting that it is conserved. 相似文献

Significant changes have occurred in the well‐established partnership between fisheries managers and geneticists over the last 50 years. It is therefore timely to review and recalibrate the ways in which genetic technologies can assist the fishing industry to maintain productive and sustainable harvests. Our objective is to contribute to the mutual understanding of all stakeholders in the genetics–management partnership. Genetic technologies that are relevant to fisheries management are grouped into eleven themes, which are described in plain language for a non‐specialist audience. The role that the genetic information plays in fisheries management is explained, along with an assessment of the challenges and barriers that may be preventing the uptake of the information into the fisheries management process. The compelling conclusion is that genetics offers a diverse collection of versatile and useful tools for informing fisheries managers about issues that have a biological basis. Presently, mainstream use of genetic tools focuses on a narrow set of fisheries management issues, but the diversity of genetic tools and the novel issues they can address indicates that uptake will grow, particularly as communication between geneticists and end‐users improves. 相似文献

Bacterial blight (BB) incited by Xanthomonas citri subsp. malvacearum (Xcm) is an important bacterial disease occurring in all cotton growing areas throughout the world, including parts of Western Kenya, that are characterized by a hot and humid climate. The disease causes seedling blight, angular leaf spot, boll rot and black arm on petioles and branches leading to a loss of fruiting branches with yield losses of up to 35 %. Fifty- one Kenyan cotton genotypes (Gossypium hirsutum L.) were established in the two counties of Siaya and Busia famous for their cotton cultivation in the western region. The BB symptoms caused by natural Xcm infection in the field were scored for each cotton accession. In addition artificial inoculation with the same strains was done to confirm the status under controlled greenhouse conditions. The results of BB disease scoring revealed that some accessions (e.g. T 24A and T 24B) possess a reasonable level of resistance. However, most of the Kenyan genotypes surveyed showed medium to severe symptoms of BB, including KSA 81 M, the only commercially grown cultivar in the region of Western Kenya. Overall, 71 % of the genotypes showed susceptibility and 29 % were classified as either resistant or moderately resistant. There is therefore need to improve the local commercial genotypes by introducing new genetic resources with a more durable BB resistance to ensure a successful revitalization of the Kenyan cotton sector. 相似文献

Genetic disease resistance is widely assumed, and occasionally proven, to cause host yield or fitness penalties due to inappropriate activation of defence response mechanisms or diversion of resources to surplus preformed defences. The study of resistance gene trade‐offs has so far been restricted to biotrophic pathogens. In some Pleosporales necrotrophic interactions, quantitative resistance is positively associated with insensitivity to effectors. Host lines that differ in sensitivity can easily be identified amongst current cultivars and advanced breeding lines. Large wheat cultivar trials were used to test whether lines sensitive or insensitive to three necrotrophic effectors from Pyrenophora tritici‐repentis and Parastagonospora nodorum differed in yield when subjected to natural disease and stress pressures in the West Australian wheat belt. There was no significant yield penalty associated with insensitivity to the fungal effectors ToxA, SnTox1 and SnTox3. Some yield gains were associated with insensitivity and some of these gains could be attributed to increased disease resistance. It is concluded that insensitivity to these effectors does not render such plants more vulnerable to any relevant biotic or abiotic stress present in these trials. These results suggest that the elimination of sensitivity alleles for necrotrophic effectors is a safe and facile strategy for improving disease resistance whilst maintaining or improving other desirable traits. 相似文献

Rust fungi are major pathogens of many annual and perennial crops. Crop protection is largely based on genetic and chemical control. Fungicide resistance is a significant issue that has affected many crop pathogens. Some pathogens have rapidly developed resistance and hence are regarded as high‐risk species. Rust fungi have been classified as being low risk, in spite of sharing many relevant features with high‐risk pathogens. An examination of the evidence suggests that rust fungi may be wrongly classified as low risk. Of the nine classes of fungicide to which resistance has developed, six are inactive against rusts. The three remaining classes are quinone outside inhibitors (QoIs), demethylation inhibitors (DMIs) and succinate dehydrogenase inhibitors (SDHIs). QoIs have been protected by a recently discovered intron that renders resistant mutants unviable. Low levels of resistance have developed to DMIs, but with limited field significance. Older SDHI fungicides were inactive against rusts. Some of the SDHIs introduced since 2003 are active against rusts, so it may be that insufficient time has elapsed for resistance to develop, especially as SDHIs are generally sold in mixtures with other actives. It would therefore seem prudent to increase the level of vigilance for possible cases of resistance to established and new fungicides in rusts. © 2014 Society of Chemical Industry 相似文献

Which plant species can be found on rice field bunds and what are the prevailing life forms? Which plant communities occur and what are the main environmental drivers and phytogeographic patterns shaping these communities? How do species diversity and composition differ between bunds and paddies? To answer these questions, 133 vegetation relevés using the Braun-Blanquet method were collected in lowlands and uplands of Vietnam and the Philippines between 2012 and 2015. Soil samples were collected and farmers were interviewed. Properties of soil, climate, and geography were assessed, further structural parameters, landscape heterogeneity, seasonality, management and intensity of cultivation. Hierarchical UPGMA cluster analysis and NMDS ordinations were performed to visualize variation in plant community composition and the determinants. We found 302 vascular plant species, of which 94 species are red listed by the IUCN (under category “Least concern”). Therophytes and helophytes are the prevailing life forms. Six clusters of plant communities were classified, and temperature, soil acidity, land use intensity and nutrient availability were identified (according to relevance) as explanatory variables. Bunds revealed higher species richness than paddies and bunds in mountain areas were more species-rich than those in the lowlands. We conclude that the composition of bund communities provides valuable information on environmental and biogeographical conditions of the local rice landscapes, and that it is probably the best applicable and most reliable indicator of management intensity. 相似文献

Amazonian palms are prime candidates for sustainable management of non-timber forest products and yet useful species are often harvested destructively. This paper examines a promising management initiative for one of the most useful Amazonian palms – Mauritia flexuosa – in a rural community of northeastern Peru. Based on data from household surveys (n = 57), filmed in-depth interviews, focus groups and participant observation, we identify the factors that influence: (1) the adoption of a locally developed climbing device for wild-harvesting of aguaje palm fruit; (2) the continued practice by some households of palm felling for harvesting fruit; and, (3) investment by households in planting and cultivation of aguaje palm. Our findings identify key conditions for palm management and point to the particular importance of the adoption of palm-climbing devices, not only for reducing wild palm felling but also for stimulating broader community-level conservation as well as efforts to cultivate the palm. 相似文献

The recent taxonomic classification of beech in Europe considers existence of one species (Fagus sylvatica L.) with two subspecies: F. sylvatica ssp. sylvatica and F. sylvatica ssp. orientalis. Four beech populations growing on the Greek part of the Rodopi Mountains were studied using morphological traits as well
as DNA molecular markers (AFLPs and chloroplast DNA SSR). The aim of the study was to describe the variation patterns of beech
in the Rodopi Mountains and to test the hypothesis of possible introgression between the beech subspecies’ sylvatica and orientalis in this area. Both morphological traits and gene markers revealed a possible influence of F. orientalis on the east side of Rodopi and at the low elevations, while characters resembling F. sylvatica were observed mainly on the western part of the mountains and in higher altitudes. There was a clinal increase of genetic
diversity from the west to the east, reaching a level firstly reported for beech populations. These results can be explained
either by the existence of a main refugial area for beech during the last glaciation or by the occurrence of a recent hybridization
among the subspecies, which were spatially isolated during the last glaciation and came into reproductive contact during their
