Soil erosion has long been recognized as a major environmental problem in the Three Gorges Region (TGR) where slope farming is commonly practiced but the local topography is hilly. In consideration of the poor socioeconomic position of local farmers, low cost hedgerows had been introduced as a soil conservation measure to the TGR in the late 1980s. A collaborative research programme was initiated by the Chinese University of Hong Kong, the Chinese Academy of Science, the Huazhong Agricultural University, and the Bureau of Soil and Water Conservation of Zigui County to study the potential of adopting hedgerows in the TGR. Six experimental plots (10 m × 2 m, gradient = 25°) were constructed at Zigui County, Hubei Province to study effects of hedgerows on erosion, nutrient loss and crop productivity. Results indicated that there were significant relationships between rainfall and runoff, and rainfall and soil loss, respectively. Conventional slope farming could not be considered a sustainable agricultural practice because it resulted in severe erosion and low crop yield. Hedgerows per se seemed not to be effective in reducing soil loss and boosting crop productivity, but performances could be greatly improved when they combined with the use of fertilizers. Current farming and fertilization practices, however, generally did not meet N demands of crops. Results and findings of this paper will contribute towards a technical reference for the promotion and adoption of hedgerows in the TGR.  相似文献

Aqueous biphasic electrophoresis system (ABES) incorporates electric fields into the biphasic system to separate the target biomolecules from crude feedstock. Ionic liquid (IL) is regarded as an excellent candidate as the phase-forming components for ABES because of the great electrical conductivity, which can promote the electromigration of biomolecules in ABES, and thereby enhances the separation efficiency of the target biomolecules from crude feedstock. The application of electric fields to the conventional biphasic system expedites the phase settling time of the biphasic system, which eases the subsequent scaling-up steps and reduces the overall processing time of the recovery process. Alkyl sulphate-based IL is a green and economical halide-free surfactant when compared to the other halide-containing IL. The feasibility of halide-free IL-based ABES to recover Kytococcus sedentarius TWHK01 keratinase was studied. Optimum partition coefficient (Ke = 7.53 ± 0.35) and yield (YT = 80.36% ± 0.71) were recorded with IL-ABES comprised of 15.0% (w/w) [EMIM][ESO4], 20.0% (w/w) sodium carbonate and 15% (w/w) crude feedstock. Selectivity (S) of 5.75 ± 0.27 was obtained with the IL-ABES operated at operation time of 5 min with 10 V voltage supplied. Halide-free IL is proven to be a potential phase-forming component of IL-ABES for large-scale recovery of keratinase.  相似文献

Lanthanide nanomaterials are considered a less toxic alternative to quantum dots for bioimaging applications. This study evaluated the cytotoxicity of terbium (Tb)-doped gadolinium oxide (Gd(2)O(3)) and dysprosium oxide (Dy(2)O(3)) nanoparticles exposed to human (BEAS-2B) and mouse (L929) cell lines at a concentration range of 200-2000?μg/ml for 48 h. Two assay methods were utilized-WST-8 assay (colorimetric) based on mitochondrial metabolic activity and Pico-Green assay (fluorescence), which measures total DNA content. The authors' data showed that Tb-doped Gd(2)O(3) nanoparticles were consistently more toxic than Tb-doped Dy(2)O(3) nanoparticles. However, exposure to these nanomaterials caused a decrease in proliferation rate for both cell lines rather than a net loss of viable cells after 48 h of exposure. Additionally, there was some degree of discrepancy observed with the two assay methods. For the mouse L929 cell line, the WST-8 assay yielded consistently lower proliferation rates compared to the Pico-Green assay, whereas the opposite trend was observed for the human BEAS-2B cell line. This could arise because of the differential effects of these nanoparticles on the metabolism of L929 and BEAS-2B cells, which in turn may translate to differences in their postexposure proliferation rates. Hence, the Pico-Green assay could have an advantage over the WST-8 assay because it is not skewed by the differential effects of nanomaterials on cellular metabolism.  相似文献

The stability of the aphid transmission phenotype in seven field isolates of cucumber mosaic virus (CMV) was studied, using aphids Aphis gossypii and Myzus persicae . Field isolates, obtained from four vegetable crops, were propagated in squash and Nicotiana glutinosa , and passaged by either aphid transmission or mechanical transfer. All seven isolates were transmissible by both aphids and this aphid transmission phenotype was stable after 20–24 mechanical passages. Upon further mechanical passaging, one of the seven isolates, CMV-2 A1-MT 60x, lost its transmissibility by Myzus persicae but was still transmissible by Aphis gossypii , although at a reduced rate. Isolates maintained by both aphid transmission and mechanical transfer were transmitted more efficiently by Aphis gossypii than by Myzus persicae . A comparison of the RNA profiles showed no major differences among the CMV isolates before and after mechanical passage.  相似文献

Inaccurate or incomplete diagnosis of the root causes of overfishing can lead to misguided and ineffective fisheries policies and programmes. The “Malthusian overfishing narrative” suggests that overfishing is driven by too many fishers chasing too few fish and that fishing effort grows proportionately to human population growth, requiring policy interventions that reduce fisher access, the number of fishers, or the human population. By neglecting other drivers of overfishing that may be more directly related to fishing pressure and provide more tangible policy levers for achieving fisheries sustainability, Malthusian overfishing relegates blame to regions of the world with high population growth rates, while consumers, corporations and political systems responsible for these other mediating drivers remain unexamined. While social–ecological systems literature has provided alternatives to the Malthusian paradigm, its focus on institutions and organized social units often fails to address fundamental issues of power and politics that have inhibited the design and implementation of effective fisheries policy. Here, we apply a political ecology lens to unpack Malthusian overfishing and, relying upon insights derived from the social sciences, reconstruct the narrative incorporating four exemplar mediating drivers: technology and innovation, resource demand and distribution, marginalization and equity, and governance and management. We argue that a more nuanced understanding of such factors will lead to effective and equitable fisheries policies and programmes, by identifying a suite of policy levers designed to address the root causes of overfishing in diverse contexts.  相似文献

The microalgae Isochrysis galbana and Chaetoceros muelleri were cultured with seaweed extract (SWE) from six different seaweeds and each SWE was used either as a supplement or as an alternative media to conventional f/2 medium. Microalgae cultured with only the f/2 media acted as the control treatment. I. galbana and C. muelleri were then fed to Penaeus indicus larvae, either singly or in combination, for 8 days to evaluate its nutritional quality. Shrimp larvae fed on microalgae cultured in f/2 media supplemented with SWE from Ulva lactuca or Enteromorpha intestinalis moulted faster (P < 0.05) to the second mysis stage compared with the control. Survival, total length and growth rates were better in shrimp larvae fed on microalgae previously cultured in media supplemented with SWE. In general, shrimp larvae fed microalgae cultured entirely in SWE performed equally well compared with the control. Shrimp larvae fed on mixed microalgae (I. galbana + C. muelleri) showed significantly better growth, survival and development compared with larvae on either single microalgal diet. It was concluded that microalgae cultured with SWE could be used as a low‐cost alternative media in producing live food for the hatchery production of shrimp larvae.  相似文献

The effect of the acute-phase response (APR) on the activity of the hepatic drug-metabolizing system (DMS) and on the binding of phenylbutazone to plasma proteins was investigated in the horse. An APR was induced by intramuscular injections of Freund's complete adjuvant in five horses and, five days later, these horses together with five clinically normal horses were shot and the right ventral lobe of each liver removed. The hepatic microsomal fractions from the liver samples ere isolated and significantly lower (p<0.01) concentrations of cytochromes P450 and b5 and activities of aniline-p-hydroxylase and aminopyrine N-demethylase (43%, 55%, 45% and 30%, respectively) were measured in the livers from the adjuvant-inflamed horses, compared to the controls. Phenylbutazone (PBZ) was administered intravenously (4.4 mg/kg) to a further four horses and plasma protein binding was measured by ultracentrifugation. Five weeks later, these horses were injected with Freund's complete adjuvant and the intravenous administration of PBZ (4.4 mg/kg) was repeated. Inflammation induced a significant increase (p<0.01) in the unbound fraction of PBZ (5.2±0.5 as against 1.4±0.6%). These results suggest that the APR depresses the hepatic DMS and reduces the binding of PBZ to plasma proteins.  相似文献

Albumin, globulin, glutelin and prolamin fractions were isolated from an Australian rice variety (cv. Langi) and characterised by yield, protein content and molecular weight profile using both capillary electrophoresis (SDS-CE) and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The influence of pre-extraction enzymatic hydrolysis of starch and heating to 70 °C was also investigated, as was the extraction of the glutelin fraction without prior removal of the albumin and globulin fractions. Pre-extraction treatment affected mainly the albumin fraction, increasing dry matter yield but reducing protein content. SDS-CE was able to separate the protein fractions over a wider molecular weight range than SDS-PAGE, and the peaks from SDS-CE showed slightly higher molecular weight compared to equivalent bands from SDS-PAGE. The glutelin fraction extracted without prior removal of albumin and globulin fractions had different characteristics compared to those obtained by conventional extraction methods. Pre-extraction hydrolysis of starch did not significantly affect the emulsifying, foaming and gelling properties of extracted protein. Although rice glutelin had poor solubility, emulsifying and foaming properties in aqueous systems, it had good gelling properties which could be important for food applications.  相似文献
