Farinography and mixography are two commonly used procedures for evaluating dough properties. These procedures, however, cannot separate hydration and energy input during dough development, both of which are critically important for understanding fundamental rheological properties of dough. A rheometer and laser scanning confocal microscopy (LSCM) were used to study the relationship between rheological properties and microstructural characteristics of developed (by farinograph with both shear and extensional deformations), of partially developed (by rheometer with either shear or extensional deformation), and of nondeveloped (no deformation) dough samples of wheat flours. Rheological data revealed that developed dough had the highest G* (most elastic or strong), followed by doughs partially developed with extensional deformation, and then shear deformation, and finally by nondeveloped dough. The LSCM z‐sectioning (scanning of different layers of the sample) and the analysis of amount of protein matrix showed that developed dough had the most protein matrix and nondeveloped dough had the least protein matrix. It also showed that the higher the G*, the greater the protein network. Moreover, the type of deformation appeared to contribute to the development of protein matrix and further increase the dough strength. In this study, a combination of shear and extensional deformations by farinograph produced the most protein matrix and the strongest dough, followed by extensional deformation, shear deformation, and then no deformation.  相似文献

The principle of mixer viscometry was utilized to determine the average shear rate in the mixing system (impeller‐cup combination) of the Rapid Visco Analyser (RVA). A relationship between the impeller Reynolds number and the power number was established with Newtonian standards. Using the matching viscosity technique and non‐Newtonian fluids consisting of various aqueous solutions of guar gum and methylcellulose, the average value of the mixer viscometer constant (k′) was 20.1/rev over speeds of 1.0–3.5 rev/sec (60–210 rpm). Hence, the average shear rate in the RVA can be estimated as 20.1 multiplied by the angular velocity given in revolutions per second. Results show the RVA system has very good potential for characterizing the rheological properties of various non‐Newtonian fluid foods.  相似文献

Okara is the residue left after soymilk or tofu production. In North America, okara is used either as animal feed, fertilizer, or landfill. The purpose of this study was to use wet okara to produce and enrich extruded cereal products and to study the effects of extrusion on the dietary fiber and isoflavone contents. Wet okara was combined with soft wheat flour to produce two different formulations (33.3 and 40% okara) and extruded using four combinations of two screw configurations and two temperature profiles. Various physicochemical properties, dietary fiber by enzymatic-gravimetric method, and isoflavone content by HPLC were analyzed. The radial expansion ratio decreased as fiber content increased. On the other hand, both bulk density and breaking strength increased as fiber content increased. Combining okara with soft wheat flour resulted in increased protein, dietary fiber, and isoflavone contents compared with soft wheat flour alone. Extrusion of the formulations resulted in decreased insoluble fiber (≤25.5%) and increased soluble fiber (≤150%) contents of extrudates. Extrusion decreased the total detectable isoflavones (≤20%) and altered the distribution of the six detected isoflavones.  相似文献

Twin‐screw extrusion of wheat flour and the effects on the flour proteins were studied using flour samples containing 9, 20, and 30% protein. Vital gluten containing 70% protein was used to achieve the flour protein levels. The three flour samples were extruded with a twin‐screw extruder at a combination of processing parameters (exit die temperatures of 120, 140, and 160°C, and screw speeds of 240, 320, and 400 rpm). Increasing extruder exit die temperatures resulted in increased sulfhydryl content of the 9 and 20% protein content flour samples, but appeared to have little or no effect on the 30% protein content flour sample. Similarly, disulfide content decreased, albeit disproportionately, following the same trend. Both sulfhydryl and disulfide contents of extruded samples were lower than those of the nonextruded samples and could imply denaturation of protein, aggregation through intermolecular disulfide bonds, or oxidation during extrusion processing. Total cysteine content of extruded samples decreased by ≈16% relative to nonextruded samples, but otherwise remained almost unchanged among all extruded samples. The loss of total cysteine in extruded samples could represent the generation of hydrogen sulfide, volatile organic compounds, or flavor compounds during extrusion. SDS‐PAGE analysis of total proteins showed a shift from the higher to lower molecular weight regions for certain protein bands. Both depolymerization and protein aggregation occurred at higher shear forces during extrusion.  相似文献

Clostridium autoethanogenum protein (CAP) is a single‐cell protein derived from ethanol production and may have potential to become a substitute for fishmeal in aquafeeds. A 70‐day feeding trial was conducted with black sea bream (Acanthopagrus schlegelii) (mean initial weight 6.03 ± 0.09 g) to evaluate partial replacement of fishmeal with CAP in diets. Seven isonitrogenous and isoenergetic diets were formulated with graded levels of CAP (0, 4.85, 9.70, 14.55, 19.40, 38.80 and 58.20%) to replace fishmeal. The inclusion of CAP at all dietary levels tested did not significantly affect the growth performance (p > .05). Fish fed the CAP58.20% diet showed a significantly lower feeding rate, with significantly higher protein efficiency ratio and feed efficiency ratio compared with fish fed the other diets (p < .05). No statistical differences were found in dorsal muscle and whole‐body compositions. Total superoxide dismutase in serum of fish fed CAP58.20% diet was significantly lower compared with that of the control. Malondialdehyde, catalase, total antioxidant capacity and digestive enzyme activities revealed no significant differences among dietary treatments. Phosphorus retention efficiency significantly increased, and phosphorus discharge showed a downward trend with increasing CAP inclusion levels. In conclusion, the results indicated that CAP is a safe and effective alternative protein source, which can replace fishmeal in the diet of black sea bream up to 58.20%, without adverse effects on growth performance, antioxidation and digestive enzyme activity. This study has shown the potential of converting industrial waste into a high protein feed ingredient for aquafeeds.  相似文献

In coral mariculture, growth is one of the most common ways to track success. However, numerous methods of monitoring coral growth make comparative studies challenging. A literature review of 39 studies from 1982 to 2017 indicated that the most predominant non‐invasive methods used were linear and areal measurement and these were evaluated for their accuracy using nursery‐reared corals. The monthly change in linear and areal growth rates of six coral species (= 215), Pocillopora acuta, Hydnophora rigida, Merulina ampliata, Podabacia crustacea, Echinopora lamellosa and Platygyra sinensis were measured via photogrammetry. We tested whether the planar area of coral colonies can be estimated using three geometric formulas of linear measurements. Based on the literature review, the six types of measurement methods were namely linear, volume, weight, area, count of polyps/branches and calcification, in decreasing order of application. Our results showed that the change in area calculated by geometric mean diameter (GMD) formulas provided the most accurate estimation among the three formulas and was strongly correlated with planar area (R2 ≥ .60; p < .05) for all coral species, except E. lamellosa and Pl. sinensis. However, our findings suggest that it is not ideal to use geometric formulas to estimate the change in area. Instead, we posit that areal photogrammetry represents the simplest yet accurate non‐invasive method for rapid monitoring of extensive areas of corals in situ. Lastly, we discuss the recommendations and limitations for areal photogrammetry.  相似文献

This paper examines the influences of Malaysia's Federation Land Development Authority (FELDA) scheme in fostering inclusive rural development. Based on the model of holistic inclusive development, the paper investigates the performance of FELDA from four perspectives: social development, industrialisation, modernisation and basic needs. The main findings of the study indicate that, to a large extent, the scheme has successfully stimulated both the social and economic development of the community. Nonetheless, establishing a strong trust, social cohesion and rapport between the public authorities and community remain the main challenges in determining the success of this state‐led agenda. Moreover, keeping traditional values in the modern system will be the key principle for the sustainability of the programme if plans are made to adopt the scheme in other regions.  相似文献

An LC-MS-based metabolomics approach was used to characterise the variation in secondary metabolite production due to changes in the salt content of the growth media as well as across different growth periods (incubation times). We used metabolomics as a tool to investigate the production of rifamycins (antibiotics) and other secondary metabolites in the obligate marine actinobacterial species Salinispora arenicola, isolated from Great Barrier Reef (GBR) sponges, at two defined salt concentrations and over three different incubation periods. The results indicated that a 14 day incubation period is optimal for the maximum production of rifamycin B, whereas rifamycin S and W achieve their maximum concentration at 29 days. A “chemical profile” link between the days of incubation and the salt concentration of the growth medium was shown to exist and reliably represents a critical point for selection of growth medium and harvest time.  相似文献
