Tufa is a carbonate sediment contains inorganic and organic substances such as algae, microorganism and invertebrate. Microbial diversity of tufa found in Taroko National Park was investigated using 16S rRNA cloning and fluorescent in situ hybridization (FISH). Eleven 16S rRNA phylotypes and 37 genus and group of bacteria were identified. Of total 381 clones isolated, proteobacteria occupied 25-30% whereas cyanobacteria dominated 16-28% in total microbial population in the three sites. Acidobacteria, agricultural soil bacterium, verrucomicrobia and firmicutes were, generally, distributed in the three sampling sites. Among the three sampling sites, Baiyang walkway is found to be the most diverse site in its tufa microbial composition, indicated by species richness plot and FISH.  相似文献

In support of the efforts to combat smuggling, as well as illegal sale and distribution of cigarettes, an analytical approach for the characterization of tobacco has been proposed and evaluated. It involves aqueous extraction of the filler tobaccos followed by direct analysis of the extracts by electrospray ionization-ion trap mass spectrometry (ESI-ITMS) in the negative mode. Typically, the deprotonated ions, [M - H](-), of organic acids (malic, citric, caffeic, quinic acid) and polyphenols (chlorogenic acid, rutin, scopoletin) were detected. MS/MS spectra of the ion at m/z 191, which is the [M - H](-) of quinic acid, citric acid, and scopoletin, and a fragment ion of chlorogenic acid were acquired. Significant differences in the MS and MS/MS spectra were observed between counterfeit samples and the corresponding authentic brand name cigarettes. Analysis of 25 commercial cigarettes showed that straight Virginia blends were readily distinguished from the blended products containing different tobacco types (Virginia, burley, and Oriental). The former exhibited consistently higher relative abundances of m/z 353 (chlorogenic acid) to m/z 133 (malic acid) in the MS spectra (0.9-1.2 vs 0.4-0.6) and higher intensity ratios of m/z 176 (scopoletin) to m/z 173 (0.4-0.8 vs 0.1-0.3) and of m/z 127 (quinic acid) to m/z 173 (0.7-1.0 vs 0.3-0.5) in the MS/MS spectra. Evidence is presented to demonstrate that the spectral differences were related not only to the tobacco type (Virginia, burley and Oriental) but also to the tobacco part (stem, lamina) used in the manufacture of the cigarettes.  相似文献

13C NMR with magic angle spinning (MAS) has been employed to investigate the cell walls of potatoes and Chinese water chestnuts over a range of hydration levels. Both single-pulse excitation (SPEMAS) and cross-polarization (CPMAS) experiments were carried out. Hydration led to a substantial increase in signal intensities of galactan and galacturonan in the SPEMAS spectra and a decrease in line width, implying mobilization in the backbone and side chains of pectin. In CPMAS spectra of both samples, noncellulose components showed signal loss as hydration increased. However, the signals of some galacturonan in the 3(1) helix configuration remained in the spectra even when the water content was as high as 110%. Cellulose was unaffected. It is concluded that the pectic polysaccharides experience a distribution of molecular conformations and mobility, whereas cellulose remained as typical rigid solid.  相似文献

Many species of conservation concern are in decline due to threats from multiple sources. To quantify the conservation requirements of these species we need robust estimates of the impact of each threat on the rate of population decline. However, for the vast majority of species this information is lacking. Here we demonstrate the application of integrated population modelling as a means of deriving robust estimates of the impact of multiple threats for a rapidly declining koala (Phascolarctos cinereus) population in South-east Queensland, Australia. Integrated population modelling provides a basis for reducing uncertainty and bias by formally integrating information from multiple data sources into a single model. We quantify mortality rates due to threats from dog attacks, vehicle collisions and disease and the extent to which each of these mortality rates would need to be reduced, or how much habitat would need to be restored, to stop the population declining. We show that the integrated population modelling approach substantially reduces uncertainty. We also show that recovery actions that only address single threats would need to reduce those threats to implausibly low levels to recover the population. This indicates that strategies for simultaneously tackling multiple threats are necessary; a situation that is likely to be true for many of the world’s threatened species. This study provides an important framework for quantifying the conservation requirements of species undergoing declines due to multiple threats.  相似文献

Near-infrared (NIR) spectroscopic methods for measuring degradation products, including total polar materials (TPMs) and free fatty acids (FFAs), in soy-based frying oil used for frying various foods have been successfully developed. Calibration models were developed using forward stepwise multiple linear regression (FSMLR) and partial least-squares (PLS) regression techniques and then tested with an independent set of validation samples. The results show that the quality of oil used for frying different foods can be measured with a single model. First-derivative treatments improved results for TPM measurement. In addition, PLS models gave better prediction results than FSMLR models. For PLS models, the best correlations (r) between the NIR-predicted data and the chemical method data for TPMs and FFAs in oils were 0.995 and 0.981, respectively. For FSMLR models, the best r values for TPMs and FFAs in oils were 0.993 and 0.963, respectively.  相似文献

Purpose

Biochar has long been proposed for amending agricultural soils to increase soil-water retention capacity and therefore promotes crop growth. Recent studies revealed the potential use of biochar-amended soil in landfill final covers to promote methane oxidation and odor reduction. However, the effects of biochar application ratio, compaction water content (CWC), and degree of compaction (DOC) on soil-water retention characteristics of biochar-amended clay (BAC) at high soil suction (dry condition) are not well understood. The present study aims to overcome this knowledge gap.

Materials and methods

Soil suction was induced using vapor equilibrium technique by a temperature- and humidity-controlled chamber, and the water desorption (drying) and adsorption (wetting) water retention curves (WRCs) of compacted pure kaolin clay and peanut shell BAC with different biochar application ratios (0, 5, and 20 %, w/w), DOCs (80, 90, and 100 %), and CWCs (30 and 35 %) were measured. The correlations between these factors and the gravimetric water content were analyzed by three-way ANOVA followed by the Tukey HSD test. The soil micro-structure was studied by scanning electronic microscope with energy-dispersive X-ray spectroscopy.

Results and discussion

Measured WRCs of BAC suggest that the soil-water retention capacity at high suction range (48.49–124.56 MPa) was in general increased, upon biochar application. The BAC compacted with CWC of 35 % at low (80 %) and high (100 %) DOCs for the 5 % BAC were increased by 7.30 and 9.77 %, when compared with clay, while the increases of 20 % BAC were 39.89 and 59.20 %, respectively. This is attributed to the embedded effects of clay particles in biochar pores, which reduce the total pore space of BAC. The soil-water retention capacity of BAC was also increased with CWC and decreased with DOC. The results of three-way ANOVA analysis show that the effects of DOC and biochar ratio on soil gravimetric water content was significant (p?<?0.05) only at 48.49 MPa on drying path. For other induced suctions, only effects of CWC were significant (p?<?0.05).

Conclusions

Biochar application increases soil-water retention capacity of the BAC at high soil suction (48.49–124.56 MPa) (dry condition) at both low (80 %) and high DOC (100 %). The soil-water retention capacity of 20 % BAC was much higher than that of 5 % BAC. BAC is a potential alternative landfill final cover soil with a higher soil-water retention capacity to be used in dry areas or regions with a long period of evaporation event.

Nondeveloped, partially developed with shear and extensional deformations, and developed doughs represent different stages of dough development. To understand the relationship between gluten proteins and dough rheology, this study used disulfide‐sulfhydryl analyses, gel filtration chromatography, SDS‐PAGE, acid polyacrylamide gel electrophoresis (A‐PAGE), and densitometry to examine proteins in the four types of doughs mentioned. Free sulfhydryl content was the lowest in native flour and nondeveloped dough, and the highest in partially developed doughs, while a reverse trend was observed for disulfide content. For each flour sample, the protein elution profile from gel filtration chromatography shifted with the level of dough development. With respect to the smallest sized molecules, native flour had the most, followed by nondeveloped, partially developed, and then developed doughs. SDS‐PAGE and A‐PAGE exhibited similar protein patterns among the same chromatographed protein fractions of each native flour and its different doughs. Densitometric data showed that the amount of high molecular weight (HMW) glutenins increased and the amounts of low molecular weight (LMW) glutenins, gliadins, and albumins/globulins decreased with progressive stages of dough development. In conjunction with previously published results, indications are that the increase in the size and the amount of HMW glutenins is related to the strength of dough and the amount of protein matrix present in the dough.  相似文献

Spatially resolved fluorescence resonance energy transfer (FRET) measured by fluorescence lifetime imaging microscopy (FLIM), provides a method for tracing the catalytic activity of fluorescently tagged proteins inside live cell cultures and enables determination of the functional state of proteins in fixed cells and tissues. Here, a dynamic marker of protein kinase Calpha (PKCalpha) activation is identified and exploited. Activation of PKCalpha is detected through the binding of fluorescently tagged phosphorylation site-specific antibodies; the consequent FRET is measured through the donor fluorophore on PKCalpha by FLIM. This approach enabled the imaging of PKCalpha activation in live and fixed cultured cells and was also applied to pathological samples.  相似文献

Serum dopamine beta-hydroxylase activity, a useful biochemical index of peripheral sympathetic nervous activity, was measured in rats treated with Delta(9)-tetrahydrocannabinol or ethanol or both substances. After 7 days of treatment with either substance, serum dopamine beta-hydroxylase activity decreased significantly. Combined treatment with both agents enhanced the effects of each given alone. In rats subjected to immobilization stress, treatment with Delta(9)- tetrahydrocannabinol appeared to potentiate the stress-induced increase in serum enzyme activity. Treatment with ethanol, with or without Delta(9)-tetrahydrocannabinol, effectively blocked this increase in enzyme activity. These results show that both substances have significant effects on the sympathetic nervous system which are critically influenced by environmental setting.  相似文献

Properties of electroless copper-plated polyester fabric mainly depend on the plating bath constituents/conditions. The nickel serves to catalyze the copper deposition when hypophosphite is used as a reducing agent. In this study, the effects of deposition parameters including additive NiSO4 concentration and pH on microstructure and properties of the electroless copper plating on polyester fabric using hypophosphite as a reducing agent were investigated. The results show that at a higher NiSO4 concentration, the copper content present in the coating decreases whereas the nickel content increases slightly. On the other hand, the copper content present in the coating increases, whereas the nickel content and phosphorus decreases with respect to the rise of pH. The morphology of the copper deposits show that the particle size increase with respect to the rise of NiSO4 concentration and pH. The XRD patterns indicate that the copper-plated polyester fabrics are crystalline. In addition, there is a decrease in the surface resistance and an increase in the electromagnetic interference (EMI) shielding effectiveness (SE) with respect to the rise of Ni2+ concentration and pH of the solution as a result of gaining a greater weight in the deposits. The results suggest that the copper-plated polyester fabrics have a great potential application as an EMI shielding material.  相似文献
