Gastroendoscopic examinations were conducted on 75 Thoroughbred foals aged two to 85 days on seven breeding farms in England and Ireland. The foals showed no signs of gastric disease. There was no significant difference between lesion prevalence in foals in England (16 of 28 foals; 57 per cent) or Ireland (22 of 47 foals; 47 per cent). Neither was there any sex predilection (18 of 36 males; 20 of 39 females). Lesions were most prevalent in foals under 10 days old (8 of 9) and least prevalent in foals older than 70 days (3 of 10). Lesions occurred most frequently in the squamous mucosa immediately adjacent to the margo plicatus along the greater curvature (34 foals), whereas lesions in the squamous fundus, the glandular fundus, and the lesser curvature were observed in 11, six and three foals, respectively. Lesions were not observed in the squamous mucosa surrounding the cardia. Gastric lesions were more prevalent in foals with a previous disorder than in those without (68 vs 43 per cent) and particularly with a history of recent or concurrent diarrhoea (9 of 14).  相似文献

Sitka spruce (Picea sitchensis (Bong.) Carr.) seedlings were supplied with solutions containing nitrogen (N) at 0.1 x or 2 x the optimum rate (low-N and high-N supply, respectively) and grown either outside in a control plot or inside open-top chambers and exposed to ambient (355 &mgr;mol mol(-1)) or elevated (700 &mgr;mol mol(-1)) CO(2) concentration ([CO(2)]). Gas exchange measurements, chlorophyll determinations and nutrient analysis were made on current-year (< 1-year-old) shoots of the upper whorl after the seedlings had been growing in the [CO(2)] treatments for 17 months and the nutrient treatments for 6 months. Total seedling biomass and biomass allocation were assessed at the end of the experiment. Nutrient treatment had a significant effect on the light response curves, irrespective of [CO(2)] or chamber treatment; seedlings supplied with high-N rates had higher net photosynthetic rates than seedlings supplied with low-N rates. The degree of photosynthetic stimulation in response to elevated [CO(2)] was larger in seedlings receiving high-N rates than in seedlings receiving low-N rates. Light-saturated net photosynthesis of seedlings grown and measured in elevated [CO(2)] was 26% higher than that of seedlings grown and measured in ambient [CO(2)]. There was no significant effect of [CO(2)] or chamber treatment on the CO(2) response curves of seedlings receiving High-N supply rates. In contrast, analysis of the CO(2) response curves of seedlings receiving Low-N supply rates showed acclimation to elevated [CO(2)]. Both maximum rate of carboxylation (V(cmax)) and maximum electron transport capacity (J(max)) were lower and J(max)/V(cmax) higher in seedlings in the elevated [CO(2)] treatment. There was no effect of elevated [CO(2)] on stomatal conductance, although it was highly dependent on foliar [N], ranging from ~60 mmol m(-2) s(-1) at ~1.5 g N m(-2) to 200 mmol m(-2) s(-1) at ~5 g N m(-2). In the high-N and low-N treatments, foliar N concentration was 10 and 28% lower in seedlings grown in elevated [CO(2)] than in seedlings grown in ambient [CO(2)], respectively. There was no [CO(2)] effect on foliar phosphorus concentration ([P]). Chlorophyll concentration increased with increasing N supply in all treatments. There was no significant effect of elevated [CO(2)] on specific leaf area. Chlorophyll concentration expressed either on an area or dry mass basis for a given foliar [N] was higher in seedlings grown in elevated [CO(2)] than in seedings grown in ambient [CO(2)]. Elevated [CO(2)] increased total biomass accumulation by 37% in seedlings in the high-N treatment but had no effect in seedlings in the low-N treatment. There was a proportionally bigger allocation of biomass to roots of seedlings in the elevated [CO(2)] + low-N supply rate treatment compared with seedlings in other treatments. This resulted in a reduction in aboveground biomass compared with corresponding seedlings grown in ambient [CO(2)].  相似文献

Gross and histologic postmortem studies were performed on the hind feet of feedlot cattle that had, or were free from, lesions of toe tip necrosis. The hind feet of feedlot cattle were collected by 3 veterinary feedlot practices in southern Alberta, Canada. Three studies of these feet were conducted: i) prediction of disease based on the presence or absence of apical white line separation, ii) gross assessment of the distribution and severity of lesions within affected claws, and iii) microscopic evaluation of the distal phalanx and surrounding soft tissues of affected claws. Prediction of toe tip necrosis based on the presence of apical white line separation was statistically significant (P < 0.0001). This, combined with a pattern of lesions indicative of an ascending infection of the distal phalanx and the absence of other lesions, suggests that the pathogenesis involves bacterial infection originating from the most distal aspect of the toe, at the apical white line.  相似文献

A retrospective study of the demographics of the Canadian dairy industry was conducted using data derived from Statistics Canada’s Census of Agriculture from 1991 to 2011. This longitudinal study shows that Canada’s dairy industry has undergone considerable consolidation. From 1991 to 2011, the number of dairy producers and dairy farms decreased by 48.9% and 61.9%, respectively. Furthermore, this trend in consolidation is likely to continue; nearly half (45.8%) of all dairy producers in Canada were ≥ 50 y of age in 2011. Not only will most of these producers be retired by 2021, but younger producers will also exit the industry for other reasons. These findings mirror what is occurring in Canada’s cow-calf industry, underscoring that supply-managed sectors experience demographic consolidation similar to non-supply managed sectors. These substantial changes will have ramifications for the veterinary profession. There will be fewer but larger farms, and the services and knowledge demanded of veterinarians will change accordingly, which has implications for food animal veterinary education.  相似文献

Background

There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells (MSCs) have been isolated and characterized from various species, but are poorly characterized in goats.

Results

Goat MSCs isolated from bone marrow (BM-MSCs) and adipose tissue (ASCs) have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency (CFE), yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection.

Conclusions

Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

This paper investigates the occurrence and distribution of the lignan metabolites enterodiol (END) and enterolactone (ENL) and the isoflavone daidzein (DAID) in rat tissues by use of liquid chromatography-electrospray ionization mass spectrometry (LC-ESI/MSn) following a variety of dietary regimes. Furthermore, we examined the dose-response and distribution of END and ENL in liver, testes, prostate, and lung, and we investigated the effects of competition between lignans and isoflavones on metabolite distribution. In liver, testes, prostate, and lung tissue, dose-related increases in END concentration were observed. In the testes, coadministration of 60 mg/kg secoisolariciresinol diglycoside (SDG) with 60 mg/kg isoflavones produced alterations in the resulting metabolite profile, causing increased END concentration and decreased DAID concentration. Results indicate lignan accumulation in tissues occurs, and coadministration of lignans with isoflavones affects the metabolite profile, with effects dependent on tissue type.  相似文献

There is substantial interest in how mortality rates affect animal populations, but mechanisms explaining when and under what circumstances particular causes of death incur demographic responses are far from clear. In theory, small or expanding populations should experience additive mortality from anthropogenic causes of death, but whether such effects are homogenous across a population or expressed only in certain high-risk individuals is open for debate. We used competing risks models to analyze mortality patterns among radio-collared wolves (Canis lupus, n = 711) from three populations in northwestern United States (1982-2004), and evaluated the degree to which anthropogenic mortality was additive vs. compensatory to natural demographic processes. Almost 80% (n = 320) of wolves dying of known fates were killed by anthropogenic causes (legal control, illegal killing, harvest in Canada, vehicle collision), and additive effects of anthropogenic mortality were most pronounced in northwestern Montana where wolf exposure to humans and livestock was high compared to either the Greater Yellowstone Area or central Idaho, where anthropogenic risk was lower. In contrast, risk from natural hazards was lower in northwestern Montana than in the other areas, implying some degree of compensatory mortality from anthropogenic risk. Animals recruited to the study following human-wolf conflict had markedly higher anthropogenic risk than those recruited for standard monitoring purposes, and juvenile wolves as well as dispersers, succumbed to higher anthropogenic risk. Multivariate models revealed that increasing wolf population density promoted higher anthropogenic risk and reduced natural risk, indicating that partially-compensatory effects of anthropogenic mortality actually became increasingly additive with population density. The observed compensatory mortality and hazard heterogeneity in our study implies that demographic responses to mortality risk may be complex and more subtle than previously thought; the density-dependent effect of anthropogenic mortality portends a stabilizing influence of humans on recovering wolf populations. We conclude that future assessment of the role of anthropogenic mortality should include individual-based hazard estimation as a complement to traditional population-level approaches.  相似文献

Soil-dwelling insect herbivores are significant pests in many managed ecosystems. Because eggs and larvae are difficult to observe, mathematical models have been developed to predict life-cycle events occurring in the soil. To date, these models have incorporated very little empirical information about how soil and drought conditions interact to shape these processes. This study investigated how soil temperature (10, 15, 20 and 25 °C), water content (0.02 (air dried), 0.10 and 0.25 g g?1) and pH (5, 7 and 9) interactively affected egg hatching and early larval lifespan of the clover root weevil (Sitona lepidus Gyllenhal, Coleoptera: Curculionidae). Eggs developed over 3.5 times faster at 25 °C compared with 10 °C (hatching after 40.1 and 11.5 days, respectively). The effect of drought on S. lepidus eggs was investigated by exposing eggs to drought conditions before wetting the soil (2–12 days later) at four temperatures. No eggs hatched in dry soil, suggesting that S. lepidus eggs require water to remain viable. Eggs hatched significantly sooner in slightly acidic soil (pH 5) compared with soils with higher pH values. There was also a significant interaction between soil temperature, pH and soil water content. Egg viability was significantly reduced by exposure to drought. When exposed to 2–6 days of drought, egg viability was 80–100% at all temperatures but fell to 50% after 12 days exposure at 10 °C and did not hatch at all at 20 °C and above. Drought exposure also increased hatching time of viable eggs. The effects of soil conditions on unfed larvae were less influential, except for soil temperature which significantly reduced larval longevity by 57% when reared at 25 °C compared with 10 °C (4.1 and 9.7 days, respectively). The effects of soil conditions on S. lepidus eggs and larvae are discussed in the context of global climate change and how such empirically based information could be useful for refining existing mathematical models of these processes.  相似文献
