In vitro and in vivo experiments with Sprague-Dawley rats showed that the three organophosphate insecticides tested (see below) depressed endogenous corticosterone synthesis and blocked corticosteroidogenesis in response to ACTH and cAMP stimulation of a suspension of adrenal cells. Pregnenolone stimulation of adrenal cells was not inhibited at the insecticide concentrations which blocked the ACTH and cAMP stimulation of corticosteroidogenesis. It was concluded that the insecticides act beyond the site of action of ACTH and at or beyond the level of cAMP metabolism and prior to the metabolism of pregnenolone.Insecticides tested were: dichlorvos (O,O-dimethyl-O,2,2-dichlorovinyl phosphate), Dursban (phosphorothioic acid: O,O-diethyl O-3,5,6-trichloro-2-pyridyl ester), and Diazinon (phosphorothioic acid: O,O-diethyl O-(2-isopropyl-6-methyl-4-pyrimidinyl ester).  相似文献

Context

Winter soundscapes are likely different from soundscapes in other seasons considering wildlife vocalizations (biophony) decrease, wind events (geophony) increase, and winter vehicle noise (technophony) occurs. The temporal variation and spatial relationships of soundscape components to the landscape in winter have not been quantified and described until now.

Objectives

Our objectives were to determine the temporal and spatial variation and acoustic–environmental relationships of a winter soundscape in south-central Alaska.

Methods

We recorded ambient sounds at 62 locations throughout Kenai National Wildlife Refuge (December 2011–April 2012). We calculated the normalized power spectral density in 59,597 recordings and used machine learning to determine acoustic–environmental relationships and produce spatial models of soundscape components.

Results

Geophony was the most prevalent component (84 %) followed by technophony (15 %), and biophony (1 %). Geophony occurred primarily at night, varied little by month, and was strongly associated with lakes. Technophony and biophony had similar temporal variation, peaking in April. Technophony occurred closer to urban areas and at locations with high snowmobile activity. Biophony occurred closer to rivers and was inversely related to snowmobile activity. Over 75 % of sample sites had >1 recordings of airplane or snowmobile noise, mainly in remote areas.

Conclusions

The soundscape displayed distinct patterns across 24-h and monthly timeframes. These patterns were strongly associated with land cover variables which demonstrate discrete acoustic–environmental relationships exhibiting distinct spatial patterns in the landscape. Despite the predominance of geophony, the presence of technophony in this winter soundscape may have significant negative effects to wildlife and wilderness quality.

Although one study showed lower adiponectin concentrations in obese dogs, other recent studies indicate that adiponectin might not be decreased in obese dogs, raising the possibility that the physiology of adiponectin is different in dogs than in humans. The aim of this study was to investigate possible causes of the discrepancy between the two largest studies to date that assessed the association between adiposity and adiponectin concentration in dogs, including the validity of the assay, laboratory error, and the effects of breed, sex, and neuter status on the relationship between adiposity and adiponectin concentrations. Adiponectin concentrations measured with a previously validated adiponectin ELISA were compared with those estimated by Western blotting analysis of reduced and denatured plasma samples. The possibility of laboratory error and the effect of EDTA anticoagulant and aprotinin were tested. Adiponectin concentration was measured by ELISA in 20 lean dogs (10 male and 10 female, 5 neutered in each sex). There was close correlation between adiponectin concentrations measured by ELISA and those estimated by Western blotting analysis (r = 0.90; P < 0.001). There was no substantial effect of EDTA, aprotinin, or laboratory error on the results. There was confounding by neuter status of the relationship between adiposity and adiponectin concentrations, but adiponectin concentrations were not significantly lower in male than in female lean dogs (females, 36 mg/L; males, 26 mg/L; P > 0.20) and were not significantly lower in intact than in neutered lean male dogs (intact, 28 mg/L; neutered, 23 mg/L; P = 0.49). We conclude that the adiponectin ELISA previously validated for use in dogs appears to be suitable for determination of canine adiponectin concentrations and that testosterone does not appear to have a strong effect on plasma adiponectin concentrations in dogs. OBJECTIVE-To compare early socialization practices between litters of breeders registered with the Canine Control Council (CCC) and litters of nonregistered breeders advertising puppies for sale in a local newspaper. DESIGN-Retrospective cohort study. Animals-80 litters of purebred and mixed-breed dogs from registered (n = 40) and non-registered (40) breeders. PROCEDURES-Registered breeders were randomly selected from the CCC website, and nonregistered breeders were randomly selected from a weekly advertising newspaper. The litter sold most recently by each breeder was then enrolled in the study. Information pertaining to socialization practices for each litter was obtained through a questionnaire administered over the telephone. RESULTS-Registered breeders generally had more breeding bitches and had more litters than did nonregistered breeders. Litters of registered breeders were more likely to have been socialized with adult dogs, people of different appearances, and various environmental stimuli, compared with litters of nonregistered breeders. Litters from registered breeders were also much less likely to have been the result of an unplanned pregnancy. CONCLUSIONS AND CLINICAL RELEVANCE-Among those breeders represented, litters of registered breeders received more socialization experience, compared with litters of nonregistered breeders. People purchasing puppies from nonregistered breeders should focus on socializing their puppies between the time of purchase and 14 weeks of age. Additional research is required to determine whether puppies from nonregistered breeders are at increased risk of behavioral problems and are therefore more likely to be relinquished to animal shelters or euthanized, relative to puppies from registered breeders.  相似文献

The objective of this study was to determine the effects of various methods of sperm pre‐treatment on male pronuclear (MPN) formation and subsequent development of ovine embryos derived from in vitro‐matured oocytes and intracytoplasmic sperm injection (ICSI). The effect of treatment of injected oocytes with dithiothreitol (DTT) on embryo development was also assessed. In Exp. 1, the injected oocytes with non‐treated sperm were activated with three different procedures. The cleavage and blastocyst rates in those activated with DTT was lower (p < 0.05) than those activated with either ionomycin (Io) + 6‐dimethylaminopurine (6‐DMAP) or DTT + I + 6‐DMAP. In Exp. 2, the effects of sperm pre‐incubated with DTT, sodium dodecyl sulphate (SDS) or DTT + SDS as well as two‐time frozen/thawed sperm (without cryoprotectant) on MPN formation and oocyte activation were examined. The non‐treated sperm served as controls. The MPN formation in DTT + SDS group was higher (p < 0.05) than other groups except for freeze–thaw group. No difference in the rate of activated ICSI oocytes was observed among groups. In Exp. 3, the effect of pre‐treatment of sperm on subsequent development of ICSI embryos and blastocyst cell numbers were examined. The rates of cleavage and blastocyst formation as well as the blastocyst cell numbers were similar among the pre‐treated and control groups. In conclusion, pre‐treatment of sperm with DTT + SDS positively affected MPN formation, although the subsequent development capacity of the resulting embryos remained limited. Moreover, DTT was not effective on oocyte activation compared with Io + 6‐DMAP after ICSI.  相似文献

The concentrations of trace elements in fish roes and the effect of processing conditions (karasumi-like or karashi mentaiko) were investigated in six commercial fish species from New Zealand. The studied elements were As, Cd, Cr, Cu, Hg, Pb, and Zn, and the roes were from the following species: chinook salmon ( Oncorhynchus tshawytscha), hoki ( Macruronus novaezelandiae), southern blue whiting ( Micromesistius australis), hake ( Merluccius australis), blue warehou ( Seriolella brama), and barracouta ( Thyrsites atun). The concentrations of As, Cd, Cr, Hg, and Pb in the roes were lower than literature values for fish muscles. Only Zn in barracouta roe and Cu in salmon roe and their products were relatively higher than the generally accepted levels in fish muscles and could be of safety concern. Hence, the consumption of barracouta and salmon roes among certain parts of the population needs to be monitored and assessed. Dry salting (karasumi-like) processing increased ( P < 0.001) the concentrations of the studied trace elements while salting fermentation (karashi mentaiko) processing tended to decrease the levels of trace elements. Fermentation may be a useful process to decrease the level of toxic trace elements.  相似文献
