Low levels of intracellular reactive oxygen species (ROS) are essential for normal sperm function and are produced by sperm mitochondria as a byproduct of metabolism, but in excess, ROS can cause catastrophic cellular damage and has been correlated with infertility, poor sperm motility and abnormal morphology in humans. Stallion sperm motility is fueled predominantly by oxidative phosphorylation-produced ATP, requiring high basal rates of mitochondrial function. Consequently, whether elevated ROS production by stallion sperm is an indicator of dysfunctional or highly motile cells has been debated by researchers over the last decade. The objective of this study was to evaluate the relationship between various sperm morphologies and ROS production in fresh and cooled stallion semen by employing the novel method of imaging flow cytometry for stallion semen assessment. For evaluation of fresh semen, single ejaculates (n = 5) were collected from four resident stallions at the University of California, Davis. For the evaluation of 24-h cool-stored semen, single ejaculates were collected from stallions at Texas A&M University (n = 5) and shipped to the University of California, Davis overnight for evaluation. Ejaculate volume, sperm concentration and motility parameters were recorded. Samples were co-stained for viability and ROS detection with SytoxGreen™ and dihydroethidium (DHE), respectively, and evaluated with the Amnis® ImageStream® system (Luminex Corporation). Antimycin, an electron transport chain inhibitor that triggers ROS production (1 μM), was used as a positive control for DHE, while dead cells (2× snap frozen in liquid nitrogen) served as a positive control for SytoxGreen™. Unstained samples were also evaluated as controls. Imaging flow cytometric analysis was performed with the ideas ® software (Luminex Corporation). Evaluated morphologies included abnormal head (AH), abnormal midpiece (AM), abnormal tail (AT), proximal cytoplasmic droplet (PD), or distal cytoplasmic droplet (DD), and morphologically normal (MN) cells. For fresh semen, an additional abnormality, coiled tail and midpiece (CTM) was assessed; 24-h cool-stored semen did not contain enough viable CTM cells for analysis. Only cells with obvious, single abnormalities were selected for the first portion of analysis to minimize subjectivity. Mixed effects modelling was used to evaluate the relationship between each morphologic classification and the corresponding DHE fluorescence intensity. Compared to the MN population, ROS production was significantly higher in viable cells with AH, PD and AM (p < .0001) in both fresh and cooled semen. CTM cells had significantly higher levels of ROS production compared to MN cells in fresh semen (p < .0001). There was no significant difference in ROS levels between MN cells and AT and DD cells in either fresh or cooled semen (p > .05). These results suggest that ROS generation is indicative of abnormal cell morphology and function and confirm that imaging flow cytometry is a valuable tool for the assessment of stallion semen.  相似文献

Biosecurity risks are a major threat to the profitability of the industry as well as impacting human and animal health. Livestock producers play a crucial role in biosecurity as the first to notice changes in the health or productivity of their stock and are generally responsible for implementing protective measures. However, uptake of biosecurity measures by producers is variable. We critically appraised the current literature regarding biosecurity practices in Australian livestock industries and highlight aspects that are well understood as well as those where further research or information is needed. Findings from 12 cross-sectional studies suggest that Australian producers' knowledge of biosecurity methods and importance might have a positive influence on their willingness to implement or incorporate biosecurity practices. There is moderate evidence supportive of biosecurity being well understood by livestock producers across Australia. Barriers to producers using biosecurity practices included lack of information or communication from agricultural, veterinary or government organisations. It was found that larger stock numbers were positively correlated with biosecurity implementation and that producers used veterinarians, government and industry agencies as resources for trusted information.  相似文献

Because there is no simple device capable of measuring the dry deposition rates of small particles and trace gases directly, much current activity is focused on the use of an inferential technique. In this method, measurements of atmospheric concentration (C) of selected chemical species are coupled with evaluations of appropriate deposition velocity (V d ) to yield estimates of dry deposition rate from their product. Difficulties arise concerning the ability to measure C, and especially regarding the poor knowledge of V d for many species. A multiple resistance routine for deriving deposition velocities is presented here. Current knowledge of biological processes is incorporated into a first-generation lsbig leaf’ model; formulations of resistances appropriate for describing individual leaves are combined to simulate the canopy as a whole. The canopy resistance is combined with estimates of aerodynamic and boundary-layer resistances to approximate the total resistance to transfer, from which deposition velocity is then computed. Special emphasis is given to the influence of the diurnal cycle, to the way in which the various transfer resistances can be inferred from routine data, and to the role of canopy factors (e.g., leaf area index, wetness, temperature response, and sunshade fractions).  相似文献

The discovery of D4S10, an anonymous DNA marker genetically linked to Huntington's disease (HD), introduced the capacity for limited presymptomatic diagnosis in this late-onset neurodegenerative disorder and raised the hope of cloning and characterizing the defect based on its chromosomal location. Progress on both fronts has been limited by the absence of additional DNA markers closer to the HD gene. An anonymous DNA locus, D4S43, has now been found that shows extremely tight linkage to HD. Like the disease gene, D4S43 is located in the most distal region of the chromosome 4 short arm, flanked by D4S10 and the telomere. In three extended HD kindreds, D4S43 displays no recombination with HD, placing it within 0 to 1.5 centimorgans of the genetic defect. Expansion of the D4S43 region to include 108 kilobases of cloned DNA has allowed identification of eight restriction fragment length polymorphisms and at least two independent coding segments. In the absence of crossovers, these genes must be considered candidates for the site of the HD defect, although the D4S43 restriction fragment length polymorphisms do not display linkage disequilibrium with the disease gene.  相似文献

Physicochemical, binding, and antibacterial properties of chitosans prepared without and with deproteinization (DP) process (5, 10, 15, and 30 min at 15 psi/121 degrees C) were compared. Chitosan from DP 0 min had comparable nitrogen content, lower degree of deacetylation and solubility, but higher molecular weight and viscosity than chitosans from DP 5-30 min. The latter four chitosans showed differences only in molecular weight. Deproteinization treatment resulted in slightly decreased L values and increased a and b values compared with those of DP 0 min. Chitosan from DP 0 min had comparable water and fat-binding capacity (FBC) except for chitosan from DP 15 min, which had a higher FBC but lower dye-binding capacity than those of the four chitosans from DP 5-30 min. The antibacterial activities of chitosans against seven different bacteria showed that the inhibitory effects varied with the deproteinization time and the particular bacterium.  相似文献

Objective The present study investigated the foot health of the Kaimanawa feral horse population and tested the hypotheses that horses would have a large range of foot morphology and that the incidence of foot abnormality would be significantly high. Procedures Abnormality was defined as a variation from what the two veterinarian assessors considered as optimal morphology and which was considered to impact negatively on the structure and/or function of the foot. Fifteen morphometric variables were measured on four calibrated photographic views of all four feet of 20 adult Kaimanawa feral horses. Four morphometric variables were measured from the lateromedial radiographs of the left forefoot of each horse. In addition, the study identified the incidence of gross abnormality observed on the photographs and radiographs of all 80 feet. Results There was a large variation between horses in the morphometric dimensions, indicating an inconsistent foot type. Mean hoof variables were outside the normal range recommended by veterinarians and hoof care providers; 35% of all feet had a long toe conformation and 15% had a mediolateral imbalance. Abnormalities included lateral (85% of horses) and dorsal (90% of horses) wall flares, presence of laminar rings (80% of horses) and bull-nose tip of the distal phalanx (75% of horses). Both hypotheses were therefore accepted. Conclusions The Kaimanawa feral horse population demonstrated a broad range of foot abnormalities and we propose that one reason for the questionable foot health and conformation is lack of abrasive wearing by the environment. In comparison with other feral horse populations in Australia and America there may be less pressure on the natural selection of the foot of the Kaimanawa horses by the forgiving environment of the Kaimanawa Ranges. Contrary to popular belief, the feral horse foot type should not be used as an ideal model for the domestic horse foot.  相似文献

In this study, Babesia screening was conducted in 55 rodents and 160 tick samples collected from primary forests and an oil palm plantation in Sarawak, Malaysian Borneo. PCR targeting the 18S ribosomal DNA revealed the presence of Babesia spp. DNA detected in two questing male Haemaphysalis shimoga ticks collected from the oil palm plantation. Sequence analysis revealed that both sequences were identical and had 98.6% identity to a Babesia macropus sequence obtained from Eastern grey kangaroos (Macropus giganteus) in Australia. Phylogenetic tree revealed clustering with marsupial-associated Babesia spp. in the Babesia sensu stricto clade. Whether or not H. shimoga is the competent vector and the importance of the Babesia sp. detected in this study warrants more investigation.  相似文献

Tramadol is a centrally acting analgesic with opioid and monoaminergic actions. Its clinical effects have been well characterized in humans, where it has been in use for many years, but little is known for veterinary species. This study evaluated the sedative, emetic, thiopental‐sparing and intraoperative respiratory and hemodynamic effects of tramadol in comparison to morphine for pre‐medication of dogs undergoing orthopedic surgery under halothane anesthesia. Sixteen adult, healthy, mixed breed dogs (8.0 ± 2.6 kg) were studied. Eight dogs were pre‐medicated with tramadol (1.0 mg kg‐1 IM) and the other eight with morphine (1.0 mg kg–1 IM). After 20 minutes, anesthesia was induced with thiopental and subsequently maintained with halothane in oxygen using a Bain system, with spontaneous respiration. Degree of sedation and occurrence of emesis were evaluated after pre‐anesthetic medication. Dose of thiopental necessary for tracheal intubation was compared between the two groups. Arterial blood gas analyzes were done before pre‐medication and at 60 minutes of anesthesia. Heart rate and noninvasive arterial blood pressure were recorded before pre‐medication and every 10 minutes during anesthesia. Observer was blinded of the treatment given for each dog. Tramadol produced no visible sedation and no vomiting, while morphine induced a moderate degree of sedation in all dogs and vomiting in 62% of them. Dogs pre‐medicated with tramadol required significantly more thiopental (17 ± 3.8 mg kg–1) for induction of anesthesia than those pre‐medicated with morphine (12 ± 1.8 mg kg–1). Pre‐medication with morphine was associated with significantly higher PaCO2 and lower pH at 60 minutes of anesthesia. The remaining respiratory parameters and the hemodynamic variables were similar between the two groups. In conclusion, dogs pre‐medicated with tramadol at 1 mg kg–1 IM do not become visibly sedated and require a greater amount of thiopental for induction of anesthesia than pre‐medication with morphine. As intraoperative respiratory function is better preserved with tramadol, it may be useful for pre‐medication of respiratory compromised patients.  相似文献
