Two experiments were conducted to determine theeffect of handling, short-term storage, andinitial water stress on cutting water potential (W) and rooting of loblolly pine(Pinus taeda L.) stem cuttings. First,stock plants and cuttings were measured forW at predawn (04:00 a.m.) and earlymorning (09:00 a.m.). Cuttings were thensevered, wrapped in wet paper towels, andplaced in insulated containers for 2 or 7 h atapproximately 30 °C or for 21 h in coldstorage (4 °C). Water potentials ofcuttings were measured at the end of eachstorage period. Second, effects of initialwater stress on rooting performance of cuttingswere tested by withholding water from dormant(winter) and succulent (summer) cuttings forvarying periods of time. After each dryingtreatment, W was measured on asample of cuttings and the remainder of thecuttings were transferred to a greenhouse withintermittent mist for 12 weeks.Storage of cuttings for long periods (7 to 21h) of time under low vapor pressure deficitconditions resulted in less negative waterpotentials of the cuttings. Dormant cuttingsrooted at higher percentages, even after beingexposed to lower values of W Thelower values of W in dormantcuttings could be attributed to higher ambientvapor pressure deficit during the drying phase. Results suggest that subjecting cuttings tomoderate water stress for a short period oftime does not adversely affect the rooting ofcuttings. Cutting water potentials below –1.7MPa appeared to reduce rooting of succulentcuttings and water potentials below –2.0 MPaaffected rooting in dormant cuttings.  相似文献

An analytical method was developed to quantify levels of acetaldehyde in wine samples. The method utilizes headspace solid-phase microextraction with on-fiber derivatization using O-(pentafluorobenzyl)hydroxylamine and quantification by gas chromatography with flame ionization detection. The technique showed good sensitivity and reproducibility in samples of Chardonnay, Petite Sirah, and Merlot wines containing acetaldehyde at levels below the sensory threshold (40-100 ppm). The method was used to monitor acetaldehyde concentrations during the micro-oxygenation of Merlot wine in a 141 L pilot-plant experiment and a 2400 L full-scale study. In both experiments, levels of acetaldehyde remained constant for several weeks before increasing at rates of the order of 1 ppm/day. Variations in the levels of acetaldehyde present are discussed within the context of the underlying chemical reactions.  相似文献

ABSTRACT A marked-isolate, release-recapture experiment was conducted to assess the relative contributions of seed-transmitted (released isolates) versus all other inocula to foliar and grain populations of Phaeosphaeria nodorum in winter wheat rotated with nonsusceptible crops in New York and Georgia, United States. Seed infected with two distinct groups of marked isolates of P. nodorum containing rare alleles (identified by amplified fragment length polymorphisms [AFLPs]) and balanced for mating type were planted in experimental field plots in two locations in each state. Recapture was done by isolating P. nodorum from leaves showing necrotic lesions at spring tillering and flowering stages, and mature grains from spikes showing glume blotch. Isolates from these samples were genotyped by AFLPs and categorized as released or nonreleased to infer sources of inoculum. Both infected seed and other sources of the pathogen contributed significant primary inocula to populations recovered from leaves and harvested grain. Seed-transmitted genotypes accounted for a total of 57% of all isolates recovered from inoculated plots, with a range of 15 to 90% of the populations of P. nodorum collected over the season in individual, inoculated plots at the four locations. Plants in the noninoculated control plots also became diseased and 95% or more of the isolates recovered from these plots were nonreleased genotypes. Although other potential sources of P. nodorum within and adjacent to experimental plots were not ruled out, nonreleased genotypes likely were derived from immigrant ascospores potentially from sources at a considerable distance from the plots. Our results suggest that, although reduction of seedborne inoculum of P. nodorum may delay foliar epidemics, this strategy by itself is unlikely to result in high levels of control in eastern North America because of the additional contribution from alternative sources of inoculum.  相似文献

Eastern dwarf mistletoe (Arceuthobium pusillum Peck) is a hemiparasitic angiosperm that infects white spruce (Picea glauca (Moench) Voss) and red spruce (P. rubens Sarg.) in northeastern North America. The effects of mistletoe infection differ substantially between white and red spruce, with white spruce suffering greater infection-induced mortality. In the present study, we sought to determine the role that species-specific differences in needle-scale responses to parasitism may play in the observed differences in the effect of infection on host tree health. Based on the measurements made, the most apparent effect of parasitism was a reduction in needle size distal to infections. The magnitude of this reduction was greater in white spruce than in red spruce. Eastern dwarf mistletoe was a sink for host photosynthate in red spruce and white spruce; however, there were no adjustments in needle photosynthetic capacities in either host to accommodate the added sink demands of the parasite. Needle total nonstructural carbohydrate concentrations (TNC) were also unaltered by infection. Red spruce needles had higher TNC concentrations despite having lower overall photosynthetic capacities, suggesting that red spruce may be more sink limited and therefore better able to satisfy the added sink demands of parasitic infection. However, if carbon availability limits the growth of the mistletoe, one may expect that the extent of the parasitic infection would be greater in red spruce. Yet in the field, the extent of infection is generally greater in white spruce. Taken together, these results suggest that dwarf mistletoe may not substantially perturb the carbon balance of either host spruce species and that species-specific differences in needle-scale responses to the parasite cannot explain the contrasting effects of infection on white spruce and red spruce.  相似文献

Juvenile wood (JW) of conifers is often associated with compression wood (CW), with which it is sometimes believed to be identical. To determine whether JW and CW can be distinguished metabolically, we compared gas chromatographic profiles of 25 polar metabolites from rooted cuttings of a single loblolly pine (Pinus taeda L.) clone raised in controlled environment chambers and subject to three treatments: (1) grown erect with minimal wind sway (control); (2) swayed by wind from oscillating fans; and (3) with 30-cm growth increments successively bent at an angle of 45 degrees to the vertical. Profiles were compared by principal component analysis. Substantial increases in abundances of coniferin and p-glucocoumaryl alcohol separated immature JW-forming xylem tissues of the control trees from the CW-forming xylem of the bent and swayed trees.  相似文献

BACKGROUND: Serum protein electrophoresis (SPE) has emerged as a potentially valuable diagnostic tool in avian medicine; yet, there is limited information regarding SPE in cranes. Since 2000, 20 cases of unilateral periocular or facial soft tissue swelling, blepharitis, feather loss, and ocular or nasal discharge attributed to insect bite hypersensitivity were observed in cranes from a captive breeding center. SPE may be useful for evaluating these lesions. OBJECTIVE: The aim of this study was to characterize the inflammatory response in cranes with hypersensitivity reactions using SPE. METHODS: Serum samples from 7 cranes diagnosed with hypersensitivity reactions were submitted to a diagnostic laboratory for agarose gel electrophoresis. Results were compared to those in control serum samples obtained from the same cranes during routine physical examination, when they were clinically healthy. RESULTS: Total protein and a- and g-globulin concentrations were significantly increased and albumin/globulin ratios were significantly decreased in serum samples from cranes with hypersensitivity lesions compared with control samples. CONCLUSIONS: Using SPE, we documented changes in protein fraction concentrations in cranes with clinical signs of hypersensitivity. The increase in alpha- and gamma-globulin concentrations suggested inflammation and antigenic stimulation, consistent with a Type I hypersensitivity reaction.  相似文献

DNA recognition by TAL effectors is mediated by tandem repeats, each 33 to 35 residues in length, that specify nucleotides via unique repeat-variable diresidues (RVDs). The crystal structure of PthXo1 bound to its DNA target was determined by high-throughput computational structure prediction and validated by heavy-atom derivatization. Each repeat forms a left-handed, two-helix bundle that presents an RVD-containing loop to the DNA. The repeats self-associate to form a right-handed superhelix wrapped around the DNA major groove. The first RVD residue forms a stabilizing contact with the protein backbone, while the second makes a base-specific contact to the DNA sense strand. Two degenerate amino-terminal repeats also interact with the DNA. Containing several RVDs and noncanonical associations, the structure illustrates the basis of TAL effector-DNA recognition.  相似文献

Stem-girdling treatments were applied in early spring to stimulate cone formation in two genotypes of interior Douglas-fir (Pseudotsuga menziesii var. glauca (Beissn.) Franco). After girdling treatments, male cone yield increased significantly in the next growing season. The increase was 14-fold in genotype 9137. In genotype 9550, more than 8,700 male cones were induced from each tree whereas no male cones were found in controls. Female cone yield was zero in controls and low for girdled trees in both genotypes. Multiple phytohormone-related compounds, including gibberellins (GAs), cytokinins, indole-3-acetic acid (IAA), abscisic acid (ABA) and their selected metabolites, were analyzed in developing long shoots after girdling treatments by high performance liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC?CESI?CMS/MS) in multiple reaction monitoring mode. Concentrations of GA4 were slightly higher at week 2 following girdling treatment, whereas at week 8 lower GA4 concentrations were found in girdled samples. Stem girdling did not affect concentrations of IAA and major cytokinins, such as zeatin riboside and isopentenyl adenosine. Concentrations of ABA differed two-fold between the genotypes. Although girdling treatment did not cause differences in ABA concentrations, it generally resulted in higher concentrations of ABA glucose ester. Concentration increase of 7??-hydroxy ABA by girdling was only found in genotype 9550 at week 8. Girdling caused little change in concentrations of phaseic acid in both genotypes.  相似文献
