The 1994 eruption of Rabaul, in Papua New Guinea, involved a small plinian eruption at Vulcan and a vulcanian eruption on the opposite side of the caldera at Tavurvur. Vulcan's ash leachates indicate seawater interaction that is consistent with earlier observations of low sulfur dioxide emissions and the presence of ice crystals in the initial plinian eruption cloud. In contrast, Tavurvur ash leachates indicate no seawater interaction, and later sulfur dioxide emissions remained high despite low-level eruptive activity. Silicic melt inclusions indicate that the andesitic melt contained about 2 weight percent water and negligible carbon dioxide. Mafic melt inclusions in Tavurvur ash have water and carbon dioxide contents that vary systematically over the course of the eruption. The mafic melt inclusions suggest that a mafic dike intruded from below the silicic chamber and provide further evidence that mafic intrusions drive caldera unrest. 相似文献

Obesity is a serious international health problem that increases the risk of several common diseases. The genetic factors predisposing to obesity are poorly understood. A genome-wide search for type 2 diabetes-susceptibility genes identified a common variant in the FTO (fat mass and obesity associated) gene that predisposes to diabetes through an effect on body mass index (BMI). An additive association of the variant with BMI was replicated in 13 cohorts with 38,759 participants. The 16% of adults who are homozygous for the risk allele weighed about 3 kilograms more and had 1.67-fold increased odds of obesity when compared with those not inheriting a risk allele. This association was observed from age 7 years upward and reflects a specific increase in fat mass. 相似文献

Manganese (Mn) is a trace element present in forages and cereals, and its concentration depends on soil status. Manganese deficiency in cattle, goats and ewes not only impairs oestrous cycle but reduces calf birth weight. The achievement of the first oestrus is delayed, and more attempts are necessary to obtain a successful conception. This study was conducted to investigate the effect of the availability of supplemental Mn during IVM on DNA damage of cumulus cells and total glutathione (GSH) content in oocytes and cumulus cells. The effect of supplementary Mn during IVM on subsequent embryo development was also studied. The results reported here indicate (i) DNA damage in cumulus cells decreased with 0, 2, 5 and 6 ng/ml Mn supplementation during IVM (p < 0.05). (ii) Intracellular GSH‐GSSG content increased (p < 0.01) with different Mn concentrations in oocytes and cumulus cells. Also, cumulus cell number per cumulus oocyte‐complexes (COC) did not differ either before or after IVM. (iii) Addition of Mn to maturation medium resulted in similar cleavage rates (p > 0.05) at 0, 2, 5 and 6 ng/ml Mn. However, subsequent embryo development to blastocyst stage was significantly higher (p < 0.01) in oocytes matured with 5 and 6 ng/ml Mn. (iv) There was also an increase (p < 0.05) in mean cell number per blastocyst obtained from oocytes matured with 5 and 6 ng/ml respect to zero Mn (IVM alone) and 2 ng/ml Mn. This study provides evidence that optimal embryo development to the blastocyst stage was partially dependent on the presence of Mn during IVM. Moreover, the availability of Mn during oocyte maturation ensures ‘normal’ intracellular GSH content in COCs and protects DNA integrity of cumulus cells. 相似文献

Concentrations of prolactin (PRL), LH, testosterone (T), TSH and thyroxine (T4 ) were determined before and at 20, 120 and 180 min after a single iv injection of thyrotropin-releasing hormone (TRH) in eight Beagles, eight Fox Terriers, six Labrador Retrievers and five Great Danes that were normospermic. Mean basal PRL concentrations were lower in the Fox Terriers compared with the Great Danes (p < 0.05). Mean LH concentrations were higher in the Fox Terriers than in the Beagles, and T was lower in the Fox Terriers at some times but not others (p < 0.05). Thyroid Stimulating Hormone (TSH) concentrations did not differ among breeds, while mean basal T4 values were lower in Fox Terriers compared with Labrador Retrievers and Great Danes (p < 0.05). Stimulation of T4 secretion 120 and 180 min after iv TRH injection was most pronounced in the Beagles and less in the Fox Terriers (p < 0.05). The results of the present study indicate that potential breed differences in circulating concentrations of PRL, LH, T, TSH and T4 in male dogs with apparently normal fertility can be encountered, but further studies are needed to determine whether the observed differences are typical features of these breeds, reflect subsets of dogs within breeds, or are in part because of possible uncontrolled parameters such as sample timing, ambient photoperiod, housing conditions or diet. 相似文献

Increasing genetic variation beyond natural variation is an important aim in plant breeding. In the past 70 years, random mutagenesis by irradiation or by chemicals has created numerous mutants which have been frequently used in breeding. However, their application is hampered by the mutational load due to many background mutations. In the past 10 years, new techniques for site‐directed mutagenesis have been introduced to plant breeding which are commonly referred to as “genome editing.” Among these, the CRISPR/Cas9 system turned out to be the most efficient and easy to apply. DNA is cleaved by a nuclease precisely at a target site where a mutation is likely to be beneficial. The DNA is healed by the cellular repair system either by error‐prone non‐homologous end joining or by homologous recombination, by which small DNA fragments can be inserted at the target site. In this review, we describe the application of targeted mutagenesis to crop plants and the modification of agronomically important traits, which could have direct impacts on plant breeding. 相似文献

Central Greenland ice cores provide evidence of abrupt changes in climate over the past 100,000 years. Many of these changes have also been identified in sedimentary and geochemical signatures in deep-sea sediment cores from the North Atlantic, confirming the link between millennial-scale climate variability and ocean thermohaline circulation. It is shown here that two of the most prominent North Atlantic events-the rapid warming that marks the end of the last glacial period and the Bolling/Allerod-Younger Dryas oscillation-are also recorded in an ice core from Taylor Dome, in the western Ross Sea sector of Antarctica. This result contrasts with evidence from ice cores in other regions of Antarctica, which show an asynchronous response between the Northern and Southern Hemispheres. 相似文献

The aim of this study was to determine the optimum conditions for vitrifying in vitro
produced day 7 porcine embryos using different vitrification devices and blastocoele collapse methods. Firstly
embryos were collapsed by micro-pipetting, needle puncture and sucrose with and without conducting
vitrification. In the next experiment, non-collapsed embryos were vitrified in an open device using either
superfine open-pulled straws (SOPS) or the CryoLoopTM system, or vitrified in a closed device using
either the CryoTipTM or Cryo BioTM’s high security vitrification system (HSV). The
post-thaw survival of embryos vitrified in the open devices did not differ significantly (SOPS: 37.3%;
CryoLoopTM: 37.3%) nor did the post-thaw survival of embryos vitrified in the closed devices
(CryoTip™: 38.5%; HSV: 42.5%). The re-expansion rate of embryos that were collapsed via micro-pipetting
(76.0%) did not differ from those that were punctured (75.0%) or collapsed via sucrose (79.6%) when
vitrification was not performed. However, embryos collapsed via sucrose solutions (24.5%) and needle puncture
(16.0%) prior to vitrification were significantly less likely to survive vitrification than the control
(non-collapsed) embryos (53.6%, P < 0.05). The findings show that both open and closed vitrification
devices were equally effective for the vitrification of porcine blastocysts. Collapsing blastocysts prior to
vitrification did not improve survival, which is inconsistent with the findings of studies in other species.
This may be due to the extremely sensitive nature of porcine embryos, and/or the invasiveness of the
