Eighty-eight yearling beef steers (308 +/- 1.4 kg) were used in two separate trials to determine the protein-sparing value of the N added to wheat straw during the ammoniation process and to determine the effects of supplementing ammoniated straw diets with energy and ruminal escape protein. In Exp. 1, steers were fed untreated straw (US) with either 0, 150, or 500 g of soybean meal (SBM) for 88 d. The addition of SBM to US diets increased (P less than .01) straw intake and average daily gains (ADG), indicating that N was limiting. When ammoniated straw (AS) was substituted for US, the N in the AS was used as efficiently as 500 g of SBM for growth. In Exp. 2, steers had ad libitum access to AS with three levels of supplemental corn (0, 1.23, or 2.45 kg DM.animal-1.d-1) either with or without .41 kg DM of corn gluten meal (CGM) added. Straw intake decreased (P less than .01) as the amount of corn in the diet was increased, but ADG increased (P less than .01) with the addition of corn. Straw consumption was not altered by the addition of CGM, but ADG was increased (P less than .01) by an average .35 kg by CGM. Rumen and blood N components indicated that the N from AS was contributing to the ruminal N pool and that CGM was compensating for microbial protein deficiencies postruminally.  相似文献

Acetylene reduction techniques are frequently utilized to estimate legume nodule nitrogenase activity levels. However, the known symbiotic nitrogen fixation reactions have no equivalent for the rapid permeastic transport of C2H4 reduced by nitrogenase of rhyzobial cells through the cortex tissues with the subsequent volatile excretion that is essential for GC quantitation procedures. The objective of this study was to determine interrelationships of nitrogenase (C2H2 reduction) with associated cytosol enzyme components from morphologically homologous nodules of Madison hairy vetch (Vicia villosa, Roth) at anthesis as influenced by soil potassium levels. The vetch plants were grown in a siliceous thermic Psammentic Paleustalf, Eufaula, and inoculated with Rhizobium leguminosarum Frank, ATCC 10314.

Highly significant enhancement of nitrogenase activity progressed from quadratic to linear with increased soil K levels in time‐course samplings at 30, 60, 90 and 120 min. incubations at 27C. Means as C2H4 μmole g‐1 fresh nodule wt. were 25.1, 38.8, 50.1 and 92.2 for 0, 100, 200, and 300 mg K/kg soil, respectively.

Activity levels of four cytosol enzymes, aspartate aminotransferase (AST), glutamate dehydrogenase (GDH) glutamine synthetase (GS) and glutamate synthase (GOGAT), increased significantly with increased K soil levels. These are requisite to enzymatic pathways for fixed N ammonia biotransformations with subsequent xylem translocation from the legume nodule. The transaminase (AST) and ligase (GS) were dominant at all K levels with GS increasing linearly to six fold levels over the check treatment. Cytosol composition of total ureides and αKG increased significantly with increased soil K. levels. Cytosol Ca and Mg increases were not significant but highly significant increased K content with reciprocal decreased Na resulted from increased soil K levels. Multiple regression for the most reliable response surface equation within a general linear model with R2 = 60.3% was: Nitrogenase (C2H2 reduction) = 2.84 nod. wt. + 1.05 GS + 8.08 αKG + 0.11 ureide, CV = 16.2%. Practical application of these data include need for more than single time‐course C2H4 determinations from one culture incubation in order to reliably estimate C2H2 reduction capabilities of legume nodules. Adequate levels of available soil potassium were necessary for sustained high nltrogenase activity levels.  相似文献

Six 3‐year‐old goats (three males and three females) weighing 60.0 ± 18 kg (mean ± SD) were used to investigate the effect of medetomidine (MED; 20 µg kg?1 IV) and its antagonism with atipamezole (ATI; 100 µg kg?1 IV) on physiologic responses (heart rate (HR; beats minute?1), respiratory rate (RR; breaths minute?1), electrocardiogram (ECG), rectal temperature (T; °C), blood pressure (oscillometric; mm Hg), sedation (SED), posture (REC), analgesia (ALG), and stress‐related hormonal and metabolic responses (epinephrine and norepinephrine (high performance liquid chromatography with electrochemical detection), cortisol (COR; µg dL?1; radioimmunoassay), glucose (GLU; mg mL?1; enzymatic colorimetric assay), and free fatty acids (modified enzymatic colorimetric assay)); each goat received ATI or SAL in random order separated by 1 week. Jugular catheters were placed for drug administration and blood sampling (10–12 mL sample?1) using a lidocaine skin block (20 mg) 2 hours prior to beginning of each trial; during this trial, goats breathed room air. Physiologic parameters were measured, SED, REC, and ALG were scored, and blood samples were collected from jugular catheters at baseline (time = ?30 minutes), 5 minutes post‐MED administration (time = ?25 minutes), 25 minute post‐MED administration and immediately prior to antagonism (time = 0 minute), and at 5, 30, 60, and 120 minutes after administering ATI or SAL. ALG was tested by clamping the withers and metacarpus with hoof testers fitted with a force transducer to measure applied isometric force (lb) (a technique used previously in goats to evaluate analgesia). Continuous variables were analyzed by Repeated Measures analysis of variance (anova ); categorical data were analyzed using a Friedman Repeated Measures anova on ranks. A p‐value of <0.05 was considered significant. If a significant difference was found, a Dunnett's pair‐wise comparison of means was conducted. Differences between ATI and SAL were examined at 5, 30, 60, and 120 minutes using a paired t‐test with a Bonferroni correction. Administration of MED resulted in a decrease in T (38.7 ± 0.3 to 34.5 ± 0.4 °C), HR (78 ± 19 to 55 ± 9), and RR (31 ± 12 to 14 ± 5) over time; an increase in mean arterial blood pressure (90 ± 19 to 132 ± 23), COR (0.254 ± 0.125 to 4.327 ± 1.233), and GLU (82.0 ± 13.2 to 255.9 ± 38.9); and changes in SED (alert to marked sedation), REC (standing to recumbent), and ALG (metacarpus = 5 ± 2 to 14 ± 0; withers = 3 ± 2 to 14 ± 0). GLU was 62–70% higher at 60 and 120 minutes and COR was 336% higher after SAL than after ATI at 120 minutes; at 30, 60, and 120 minutes, T was 4–10% higher after ATI than SAL. There were no other significant differences. REC, SED, and ALG were antagonized after ATI. ATI did not antagonize the effect of MED on HR, RR, or MAP, but stabilized T and antagonized the increase in GLU and COR.  相似文献

Grassland birds have undergone substantial population declines throughout much of their historic ranges in North America. Most of the remaining grassland bird habitat is restricted to rangelands managed for livestock production, so grazing management has strong implications for grassland bird conservation efforts. We conducted 1 830 point-count surveys at 305 sites during 2016–2017 to evaluate the relative effects of three livestock grazing systems on the abundance and community composition of grassland birds in a northern mixed-grass prairie ecosystem of eastern Montana, United States. Our objectives were to 1) evaluate effects of grazing management on abundance and community composition of grassland obligate birds, focusing specifically on grazing systems, stocking rates, and interactions with rangeland productivity; 2) evaluate the importance of local vegetation characteristics for grassland birds within grazing systems; and 3) assess the effectiveness of rest-rotation grazing to create patch-heterogeneity in rangeland vegetation through the alteration of structural components and the response of grassland birds to these treatments. Overall, we found inconsistent responses in abundances of grassland birds relative to livestock grazing systems and no discernable differences among grazing systems relative to community composition. However, local abundances were often driven by interactions between grazing system and rangeland production potential, suggesting the effects of livestock grazing management were generally mediated by rangeland productivity. In addition, associations between avian abundance and grazing management parameters (e.g., stocking rate) were species specific. Ubiquitous guidelines for livestock grazing systems may be inappropriate for grassland bird conservation efforts in the northern mixed-grass prairie, and high stocking rates may negatively impact populations of dense-grass obligate grassland birds in this region.  相似文献
