A comparative challenge study of six marine isolates representing subtypes 1-6 of salmonid alphavirus (salmon pancreas disease virus, Genus Alphavirus, Family Togaviridae) was conducted in Atlantic salmon in a fresh water cohabitation trial. Histopathological lesions typical of pancreas disease were observed with all subtypes, and virus was re-isolated from serum of cohabitant fish in each case. Using a virus neutralization (VN) test neutralizing salmonid alphavirus (SAV) subtype 1 strain F93-125, VN antibodies were detected in all challenge groups, consistent with serological cross-reactivity between these subtypes. Using real-time RT-PCR, SAV RNA was detected in heart tissue from 2 to 3 weeks post-challenge (wpc) in all cohabitant groups excluding controls. The results obtained suggested differences in the dynamics of infection between strains of SAV and potentially between subtypes. Results for SAV subtypes 1 and 3 suggested essentially synchronous infection of cohabitant fish. These two study groups also had the highest virus load in heart tissue as measured by quantitative RT-PCR and also had the most extensive histopathological changes. In contrast, results for SAV subtypes 2 and 6 strains were consistent with asynchronous infection in the cohabitant fish and were characterized by slow spread, low virus loads and mild histopathological changes. The SAV subtype 4 and 5 strains occupied an intermediate position in this regard. Despite the use of concentration procedures, it was not possible to detect SAV RNA in water samples from selected study tanks. However, testing of faeces from the SAV subtypes 1, 3 and 6 challenge groups found positive signals in each beginning at 1-3 wpc and remaining detectable for a further 2-3 weeks. Parallel testing of mucus samples found these became positive at 2-3 wpc and remained positive for a further 1-3 weeks. These results demonstrate for the first time that shedding and transmission of virus may occur by both these routes and suggest that dispersal in these matrices should be included in any disease transmission models.  相似文献

Plant-parasitic nematodes can cause serious plant diseases and adversely affect crop production. We investigated the temporal and vertical dynamics of plant-parasitic nematodes in northeast China to determine if long-term fertilizer application suppresses the population of plant-parasitic nematodes and influences their vertical distribution in the black soil region. The long-term fertilizer application lasted for 14 years, and included three treatments: pig manure combined with chemical fertilizer (MCF), chemical fertilizer (urea and ammonium phosphate, CF), and no fertilizer (NF). A 3-year corn-wheat-soybean rotation was grown on the field site, soil samples were obtained in the corn phase and nematodes were extracted, identified and counted. Six plant-parasitic nematode genera with relative abundance over 0.1% were found in this study. Heterodera was the dominant genus in all three fertilizer treatments, and its relative abundance was highest in NF (45.4%) and lowest in MCF (32.8%). Fertilizer application had a significant effect on abundance of total soil nematodes but not on either plant-parasitic nematodes or Heterodera. Significant differences in the abundance of total soil nematodes, plant-parasitic nematodes and Heterodera were observed among soil depths and interaction of fertilizer × soil depth. Total soil nematodes were mainly distributed in the 0–20 cm soil layer, and plant-parasitic nematodes and Heterodera were mostly distributed in the 10–30 cm soil layer. Fertilizer application did not show a significant effect on plant-parasitic nematodes or Heterodera at any of the sampling depths from 0 to 80 cm. The results indicated that fertilizer has no effective control on plant-parasitic nematodes when dominant genera and their respective plant hosts exist in relatively high abundance.  相似文献

From birth to day 21, rat offspring received daily injections of naltrexone at a dosage that blocked morphine-induced analgesia 24 hours a day. At 21 days, body, brain, and cerebellar weights of naltrexone-injected animals were 18, 11, and 5 percent greater than corresponding control weights. In addition, morphometric analysis of the cerebrum revealed a somatosensory cortex that was 18 percent thicker than that of the controls. The cerebellum of naltrexone-treated rats was 41 percent larger in total area and contained at least 70 percent more glial cells and 30 percent more granule neurons. Neurons derived prenatally were unaffected by drug treatment. These results show that naltrexone can stimulate body and brain growth in rats and suggest a role for the endorphin and opiate receptor system in development.  相似文献

Large particles containing nitric acid (HNO3) were observed in the 1999/2000 Arctic winter stratosphere. These in situ observations were made over a large altitude range (16 to 21 kilometers) and horizontal extent (1800 kilometers) on several airborne sampling flights during a period of several weeks. With diameters of 10 to 20 micrometers, these sedimenting particles have significant potential to denitrify the lower stratosphere. A microphysical model of nitric acid trihydrate particles is able to simulate the growth and sedimentation of these large sizes in the lower stratosphere, but the nucleation process is not yet known. Accurate modeling of the formation of these large particles is essential for understanding Arctic denitrification and predicting future Arctic ozone abundances.  相似文献

Field tests for biological control of bacterial wilt of potatoes, caused byPseudomonas solanacearum, were conducted in Florida in 1984 and 1985. Potato seedpieces of tolerant (Ontario) and susceptible (Pungo and/or Atlantic) cultivars were immersed in a suspension of the avirulent, non-bacteriocinogenic strain ofP. solanacearum, B82, prior to planting in naturally-infested soil. Relatively low populations ofP solanacearum (2 to 4 log cfu/g soil) and root-knot nematodes (0 to 33 larvae ofMeloidogyne incognita/100 cm3 soil) were present in the soil before planting. In 1984, there was approximately a 50% reduction in incidence of bacterial wilt and brown rot in B82-treated plants of cultivar Atlantic, as compared with controls. With cultivar Ontario, on the other hand, disease incidence was very low and differences between treatments were not significant. In 1985, the incidence of bacterial wilt was low in all plots. Treatment with strain B82 reduced incidence of tuber brown rot in all cultivars, but differences between treatments were not significant at P = 0.05. There was a significant correlation, however, between incidence of wilt and root-knot nematode populations in the soil. Therefore, the possibility that control of bacterial wilt in 1984 was indirect,i e, via an effect of B82 treatment on root infection byM incognita, was examined. Greenhouse tests were conducted to determine the potential effect of seedpiece treatment with strain B82 on nematode infection of cultivars Atlantic and Ontario. In two out of three trials, the B82 treatment significantly reduced incidence of root knot. These results suggest that interactions of the biocontrol agent with root-knot nematodes may determine the effectiveness of a treatment designed for bacterial wilt control in the field.  相似文献

An avirulent strain ofPseudomonas solanacearum, B82, was tested for its ability to protect the potato cultivar, Ontario, from bacterial wilt caused by virulent strains of this bacterium. Strain B82 was not antagonistic to 124 virulent strains ofP. solanacearum and was not bacteriocinogenic. When potato seedpieces were soaked for 5 hr in suspensions of B82 (108 to 109 cfu/ml), reduction in disease severity (up to 50%) was noted in some experiments. The treated plants, grown in the greenhouse or growth room for 4–5 wk, were challenged by stem inoculation, soil drenching, or root-to-root infection with virulent strains (276 or Br5) of the bacterium. Protection was not obtained consistently, however, and much of the variability could be attributed to differences in ambient temperature and light conditions in the greenhouse between experiments. In general, high ambient temperatures and/or reduced sunlight resulted in no protection. Protection appeared to depend on the ability of strain B82 to multiply in the vascular system in the crown region and to colonize the rhizosphere of treated plants. Soil temperatures above 24 C affected the ability of strain B82 to survive in the rhizosphere. The close dependence of protection on environmental conditions suggests that this method of control may not be practical under field conditions.  相似文献

Background: Previous multidrug studies have identified the value of prednisolone in treating steroid responsive meningitis-arteritis (SRMA) and the potential value of acute phase proteins (APPs) and immunoglobulin A (IgA) in diagnosis and monitoring.
Hypothesis: (1) Prednisolone monotherapy is a successful immunosuppressive modality in the treatment of SRMA; (2) protein markers are useful in identifying the potential for relapse.
Animals: Twenty client-owned dogs with SRMA presented to the University of Glasgow Small Animal Hospital between May 2006 and May 2008.
Methods: A prospective, observational study: CBC, biochemistry, and cerebrospinal fluid (CSF) analyses were performed. C-reactive protein (CRP), serum amyloid-A, α-1-acid glycoprotein, and haptoglobin (Hp) were assessed in the serum. IgA concentrations were determined in the serum and CSF.
Results: Clinical resolution of SRMA was achieved in all 20 dogs. Serum CRP concentration remained increased at remission in 16/20 dogs whereas CSF cytology was within normal limits in 20/20 dogs. Serum APPs decreased significantly on treatment ( P < .05) except Hp, which remained unaltered. Serum and CSF IgA concentrations remained increased for the duration of treatment.
Conclusions and Clinical Importance: The prednisolone regimen presented was successful in treating SRMA without the need for additional drugs. Serum APPs are of use in the diagnosis and management of SRMA, particularly in relation to identifying relapse. Serum and CSF IgA concentrations remain increased throughout disease, aiding in diagnosis but not contributing to the management of SRMA.  相似文献
