Some Rhizobium strains synthesize a unidirectional hydrogenase system in legume nodule bacteroids; this system participates in the recycling of hydrogen that otherwise would be lost as a by-product of the nitrogen fixation process. Soybeans inoculated with Rhizobium japonicum strains that synthesized the hydrogenase system fixed significantly more nitrogen and produced greater yields than plants inoculated with strains lacking hydrogen-uptake capacity. Rhizobium strains used as inocula for legumes should have the capability to synthesize the hydrogenase system as one of their desirable characteristics. 相似文献

The malaria parasite, Plasmodium falciparum, exploits multiple ligand-receptor interactions, called invasion pathways, to invade the host erythrocyte. Strains of P. falciparum vary in their dependency on sialated red cell receptors for invasion. We show that switching from sialic acid-dependent to -independent invasion is reversible and depends on parasite ligand use. Expression of P. falciparum reticulocyte-binding like homolog 4 (PfRh4) correlates with sialic acid-independent invasion, and PfRh4 is essential for switching invasion pathways. Differential activation of PfRh4 represents a previously unknown mechanism to switch invasion pathways and provides P. falciparum with exquisite adaptability in the face of erythrocyte receptor polymorphisms and host immune responses. 相似文献

Difficulties in preparing cluster ions for spectroscopic studies have limited our understanding of intermolecular forces in charged complexes that are typical of many reactive intermediates. Here, the infrared spectrum of the charge transfer complex [Ar-N2]+, recorded in a supersonic planar plasma with a tunable diode laser spectrometer, is presented. More than 70 adjacent rovibrational transitions were measured near 2272 wave numbers and assigned to the molecular nitrogen stretching fundamental in the 2Sigma+ ground state of [Ar-N2]+. The accurate structural parameters that were determined confirm a linear structure and show that the major part of the charge is located at the argon atom. For over 30 years, Purdue University has maintained a national database of agriculture-related entrapment cases that have occurred in loose agricultural material. At present, 391 documented fatal and non-fatal entrapments from the U.S. and Canada make up the Purdue University Agricultural Entrapment Database. In order to specifically study fatal cases of entrapments in grain bins located on farms, the database was reviewed, 181 cases were identified using specific criteria, and the results were summarized. Approximately five cases per year were identified between 1966 and 1998, representing 18 states and one Canadian province. Entrapments were generally reported more often in the top corn-producing states and during the months of November, December, January, March, and June. In 24% of the cases in which the victim's age was known, the victims were younger than 16. Children and adolescents younger than 16 were more often fatally entrapped in June than in any other month. For cases in which the product was known, corn was the agent of injury in 53% of the cases and was frequently found to be out-of-condition. At the time of entrapment, victims were involved with bin unloading activities in 76% of the cases in which the activity was identified. These findings are being used to design new injury prevention strategies, including educational materials and recommendations for engineering controls that focus on primary causative factors. 相似文献

SUMMARY Thirteen Tammar wallabies (Macropus eugenii) were dosed orally with 500, 1000 or 10 000 oocysts of Toxoplasma gondii, as part of a vaccination trial. Eleven animals died of acute toxoplasmosis 9 to 15 days after challenge. The lesions were similar in all animals, consisting of foci of necrosis and inflammation in the intestines, lymphoid tissue, adrenal cortex, heart, skeletal muscle and brain, and severe generalised pulmonary congestion and oedema. Free and intracellular tachyzoites of Toxoplasma were associated with the lesions. The remaining 2 animals had shown no signs of disease when euthanased four months after challenge. Small, focal, non-suppurative inflammatory lesions were seen in brain, heart and skeletal muscle of these animals and chronic Toxoplasma infection was confirmed by mouse inoculation. 相似文献

A strain of Rhizobium japonicum used in commercial inoculants was mutagenized and screened by a rapid effectiveness assay with soybean plants. Two mutant strains nodulated the roots earlier than the wild type and also expressed greater symbiotic nitrogen-fixing activity than the wild type in the presence and absence of fixed nitrogen. In addition, one of the mutants formed more root nodules than the wild type. Plants inoculated with these strains had increased dry weights ( approximately 60 percent) and nitrogen content ( approximately 100 percent) when grown in growth chambers. 相似文献

Soil carbon dioxide (CO2) efflux is an important component of the carbon (C) cycle but the biological and physical processes involved in soil CO2 production and transport are not fully understood. To improve our knowledge, we present a new approach to measure simultaneously soil CO2 concentrations and efflux, and their respective isotopic signatures (δ13C‐CO2). To quantify soil air 13CO2 and 12CO2 concentrations, we adapted a method based on CO2 diffusion from soil pores into tubes with a highly gas‐permeable membrane wall. These tubes were placed horizontally at different depths in the soil. Air was sampled automatically from the tubes and injected through a diluting system into a tuneable diode laser absorption spectrometer. The CO2 and δ13C‐CO2 vertical profiles were thus obtained at hourly intervals. Our tests demonstrated the absence of fractionation in the membrane tubes for δ13C‐CO2. Subsequently, we set up field experiments for two forest soils, which showed that natural soil CO2 concentrations and δ13C‐CO2 were not affected significantly by the measurement system. While δ13C‐CO2 in air‐filled pores below 5 cm was constant over 3 days, we observed large diurnal variations in δ13C‐CO2 efflux. However, the average difference between the two measurements was close to ?4.4‰, which supports steady‐state diffusion over this 3‐day period. This new method seems to be a very effective way to measure the δ13C‐CO2 profile of the soil atmosphere, and demonstrates that the fractionation that occurs during diffusion is the main transport process that affects the δ13C‐CO2 of the soil CO2 efflux on a daily timescale while advection may account for within‐day variations. 相似文献
