In this work two cytokines were used in combination with inactivated bacteria (bacterin) to test the bovine conjuctival immune response to the pathogen Moraxella bovis, the causative agent of Infectious bovine keratoconjunctivitis (IBK). Treatments using the bacterin vaccine combined with interleukin-2 and interferon-α as adjuvants (Group A), the bacterin vaccine only (Group B), and controls without treatment (Group C), were applied by ocular spraying to evaluate the local immune response in the corneal structure of cattle experimentally infected with M. bovis. Six weeks after infection, 14 out of a total of 34 animals presented different corneal lesions; 9 corresponding to the control group C, 4 to the group B and only one to the group A. According to the clinical manifestations, a numeric score was calculated. Control animals presented the highest score value (12 points), followed by group B (7.5 points) and group A (1 point). These results suggest that the addition of cytokines to M. bovis treatments can reduce not only eye injuries caused by IBK but also the number of diseased animals. 相似文献

Processed organic agricultural byproducts such as liquid organic fertilizer (LOF) are promising alternatives that can improve crop productivity while reducing mineral fertilizer use and improving sustainability. The effects on beneficial soil organisms, such as arbuscular mycorrhizal (AM) fungi and phosphate solubilizing microorganisms (PSM), caused by LOF spraying on shoots is poorly understood. Therefore, we evaluated how AM colonization and soil PSM are affected by LOF spraying on common beans (Phaseolus vulgaris L.) grown in a greenhouse. The LOF was obtained by anaerobically fermenting a mixture of fresh water, cattle manure, bovine milk, sugarcane molasses and mineral salts. LOF was applied twice on the plant shoots. Fresh and dry mass of root and shoot, P content, P accumulation in the shoots and AMF spore counts in the soil were not LOF dose dependent. However, we found that a 5% LOF application stimulated AM colonization and led to dose-dependence for calcium and aluminium phosphate solubilizing bacteria and fungi. These results show that LOF sprayed on shoots causes responses on soil communities. We therefore endorse the use of LOF in low input agriculture. 相似文献

Characterization of fecundity genes offers the opportunity to improve production efficiency, and the consequent increase in litter size in livestock industry, through utilizing them in breeding programs. The main objective of this study was to detect the BMPR‐IB, BMP15 and GDF9 gene mutations and to investigate whether these mutations are associated with litter size in Egyptian sheep breeds. To achieve this goal, 73 adult ewes representing Barki (n = 33) and Rahmani (n = 40) breeds were used. Polymerase chain reaction–restriction fragment length polymorphism (PCR‐RFLP) screening approach was used to detect the presence of FecB, FecXG and FecXI mutations in the two selected breeds. Results of this study showed that the three different candidate gene mutations, namely FecB, FecXG and FecXI are not present among these selected populations of the Egyptian breeds. Further studies regarding other mutations and/or other genes, which may influence ovulation rate, should be carried out to determine the type and mode of inheritance of such genes in Egyptian sheep breeds. 相似文献

The objective of this study was to investigate (i) the effect of two different ovum pick-up (OPU) schemes (once vs twice weekly aspirations) on oocyte recovery rate, quality and subsequent in vitro embryo development, (ii) the influence of days post-partum on oocyte recovery and (iii) possible differences in OPU results from two different herds. In group A, OPU was performed twice weekly in two Holstein Friesian (HF) and three Danish Red and White (DRW) cows from a private herd. In the research herd, two groups of eight HF cows were investigated: group B (OPU once weekly) and group C (OPU twice weekly). The collected oocytes were subsequently submitted to in vitro embryo production. More oocytes were recovered from the private herd when compared with the research herd. In the research herd, the twice weekly scheme aspirated more oocytes than the once weekly scheme. The quality of the retrieved oocytes was significantly different between groups B and C but not between groups A and C, and HF cows yielded higher quality oocytes than DRW cows (p = 0.029). Oocytes from group C showed higher level of embryonic development than group B oocytes. No differences in blastocyst rates were observed between groups A and C. Session affected the number of retrieved oocytes and subsequent developmental rates, with these being lower in the first compared with the last sessions. Finally, there was no significant effect of days post-partum in the number and quality of the retrieved oocytes, likely because of the small group size and high variation between sessions. 相似文献

Pollution is a subject of worldwide concern and discussion and various areas of science are seeking to integrate their knowledge in an attempt to improve understanding of the harmful effects of modern human activities. Thus, the objective of the current study was to assess the potential for using the horse as a bio-indicator of environmental pollution. Blood and serum samples from 120 horses from locations with different degrees of urbanization and industrialization and the feeds used for these animals were analyzed to determine their heavy metal (Cd, Pb, Ni, Cu, and Zn) content. The data obtained were compared. The results did not show increases in heavy metal concentration in the blood and serum of the horses with the intensification of urbanization and industrialization around the farms. Only Cd, as determined in the blood of the animals, showed a certain potential for using horses as bio-indicators of environmental pollution. The data for sex, age, and chemical composition of the foodstuffs supplied to the animals were not determinants in explaining the variation in the concentrations of heavy metals observed. 相似文献

Soon after ovulation, the newly formed corpus luteum (CL) starts secreting progesterone (P(4)), necessary for implantation. The CL, an ovarian transient endocrine organ, undergoes growth and regression throughout its life span. The objective of this study was to evaluate if caspase-3 mediates cell death in the equine cyclic luteal structures and relate it to luteal endocrine function. Blood and luteal tissue were collected during the breeding season after slaughter from 38 randomly assigned cycling mares. Luteal tissues were classified as corpora haemorrhagica (CH; n = 7); mid luteal phase corpora lutea (Mid-CL; n = 17); late or regressing corpora lutea (Late-CL; n = 9) and corpora albicans (CA; n = 5). Plasma P(4) concentration, determined by radioimmunoassay, showed a significant increase from CH to Mid-CL (p < 0.001), followed by a decrease to Late-CL (p < 0.001) and CA (p < 0.001). Caspase-3 processing and poly (ADP) ribose polymerase (PARP) degradation were assessed by western blotting. Active caspase-3 was twofold increased in Mid-CL, Late-CL and CA as compared with CH (p < 0.05). Immunocytochemistry also showed a significant increase in caspase-3 expression in large luteal cells in all structures when compared with CH (p < 0.05). Consistently, the endogenous caspase-3 substrate, PARP, was markedly degraded from CH to CA (p < 0.05). In fact, the ratio of full-length to degraded PARP showed a significant decrease from CH to Mid-CL, Late-CL and CA (p < 0.05). Finally, the decrease in P(4) from Mid- to Late-CL coincided with no further increases in apoptosis. In conclusion, these results suggest that the effector caspase-3 of apoptosis, might play an important role during luteal tissue involution in the mare, even though its relationship with P(4) remains to be elucidated. 相似文献

Bagaza virus (BAGV) is a mosquito-borne flavivirus belonging to the Ntaya serocomplex. In 2010, a disease outbreak was reported in Cádiz (Southern Spain) affecting game birds (red-legged partridges and common pheasants). In this work, red-legged partridges were inoculated experimentally with infectious BAGV isolated from this outbreak in order to make a complete clinical and analytical assessment of the disease caused by the pathogen in this species. Viral load (by real-time RT-PCR) in blood, oral and cloacal swabs, and feathers, and neutralizing antibody titres (by VNT) were measured. In order to determine direct contact transmission, non-inoculated partridges were caged together with the inoculated ones. To assess infectiousness in other species, house sparrows and mice were also inoculated with the virus. All the inoculated partridges were clinically affected, and 30% of them died. All the infected individuals lost weight, with larger losses being recorded in females. Conversely, no mortality or disease symptoms were observed in the sparrows or mice. Remarkably, all the contact partridges acquired the infection by direct (non-vectored) transmission. This study confirms that the red-legged partridge is a susceptible host for BAGV infection, and that this pathogen is transmitted by direct contact. Long-lasting viral loads detected in calami of immature feathers demonstrate that feather sampling could be a useful strategy in active surveillance programs for early detection of BAGV.

Eucalyptus plantations have become increasingly common in Latin America. However, because Eucalyptus is an exotic species, its presence has raised concerns about changes in the environment, especially to soil properties. The objective of this study was to investigate possible changes in selected soil enzyme activity after several years of Eucalyptus cultivation. Soil samples were collected from four locations: a native forest (Atlantic Forest) used as a reference for the original soil conditions and three E. grandis plantations aged 2, 3 and 5 years, established in 2008, 2007 and 2005, respectively. The native vegetation had been removed and the soil graded and ploughed to establish these plantations. We evaluated soil enzymatic activities (β‐glucosidase, acid phosphatase, dehydrogenase, urease and arylsulfatase) at each location. The activity of β‐glucosidase, phosphatase, dehydrogenase and urease was improved after 5 years, whereas arylsulphatase was impacted negatively. The multivariate analysis showed that the majority of enzyme activities reached the values observed in native forest after the third year of reforestation. The activity of β‐glucosidase was crucial in differentiating the area with 2 years of reforestation from the native forest. The removal of native vegetation in order to establish commercial plantations raises concerns about the real impacts of this practice on the soil. An outbreak of trypanosomiasis by Trypanosoma vivax is reported in the semiarid of Paraíba, Northeastern Brazil from May to August 2002. Sixty-four cows out of 130 were affected; 11 died and the other recovered after treatment with diminazene aceturate. Affected animals had fever, anemia, weight loss, hypoglycemia, increased serum levels of aspartate aminotransferase and, in nine cows, nervous signs. All cows with nervous signs died; six of them recovered after treatment, but the disease relapsed. Six cows aborted and one delivered a calf that died immediately after parturition. Thirty-two out of 100 calves were affected and five died. Nervous signs were not observed in the calves. Gross lesions were thickening of the meninges, enlarged lymph nodes and prominent white pulp of the spleen. The main histological lesion was meningoencephalitis and malacia in the brain of cows with nervous signs. No antibodies against trypanosomes were found in 33 blood samples collected before the outbreak in the affected farm and in 29 samples collected at the same time in two other neighbor farms. Until January 2003, all 89 animals tested had antibodies against T. vivax, suggesting the occurrence of sub clinical infections in cattle without clinical signs. Only two out of 85 serum samples collected on April 2004 were positive for T. vivax antibodies. Data obtained suggested that the semiarid region is non-endemic for trypanosomiasis and that disease occurred due to introduction of the parasite in a susceptible population after an apparent rise in the Tabanus spp. population. 相似文献
