This paper presents the influence of selected methodological aspects on the results of particle‐size distribution (PSD) as measured by the laser diffraction method (LDM). The investigations were carried out using the Mastersizer 2000 with Hydro MU attachment (Malvern Ltd., UK). It was found that for the investigated soils: (1) optimal speed of pump and stirrer was 2500 rpm, (2) optimal measurement time was ≈ 1 min, (3) there are two, practically equivalent methods for soil‐sample dispersion: chemical (with the use of a solution of sodium hexametaphosphate) or physical (by means of ultrasound application for 4 min at a maximum power of 35W), (4) one must not use the chemical and physical dispersing methods simultaneously, because this can lead to aggregation (not dispersion) of soil particles, (5) the Fraunhofer theory (physical models) can be used to convert scattered‐light data to PSD. In the case of the Mie theory, the best results were obtained for a refractive index (RI) in the range of 1.5–1.6 and an absorption index (AI) of 1.0. It was also found that most of the discussed parameters depend on design of the measuring device and on the type and volume of the investigated suspensions. It is necessary, therefore, to explain how the data was obtained every time and to specify the details in the methodological part of the paper.  相似文献

The University of Nebraska and AgBios, Inc. launched the AgBiosafety website (http://agbiosafety.unl.edu) with the purpose of educating the public about agricultural biotechnology risk and safety issues through science-based content. This article discusses the creation of the website, its components, and data gathered from usage statistics and a web-delivered survey. This paper presents an exploratory investigation of the application of morphological features in image analysis for varietal classification of Polish spring barley. The objective of this study was to determine the utility of morphological features for classifying individual kernels of five varieties of barley. Furthermore, this study was performed to find the best method to classify kernels of barley with the lowest error of classification. Image processing consisted of several steps: image acquisition, segmentation, external and internal image feature extraction, classification and interpretation. Each barley kernel was described using 74 morphological features. The selection was carried out using three methods based on: Fisher's coefficient, probability of error and average correlation coefficient and mutual information. Principle component analysis (PCA), linear discriminant analysis (LDA), and non-linear discriminant analysis (NDA) were used throughout this paper as the classification methods. The results confirmed that the method using morphological features may be successfully employed in image analysis for preliminary varietal identification of barley kernels. Furthermore, LDA was found to be the method which best separated different varieties of objects.  相似文献

Genetic transformation of mature trees is difficult because adult tissues are recalcitrant to Agrobacterium tumefaciens infection and transformation and because transgenic mature events are less competent for regeneration. We have shown that reinvigoration allows manipulation of the vegetative phase to increase the potential for transformation and regeneration without loss of competence for flowering and fruiting. To produce transgenic plants from clementine mandarin (Citrus clementina hort. ex Tanaka), we optimized the conditions of the source material both ex vitro and in vitro. Grafting of mature buds on juvenile rootstocks in the spring and preventing multiple bud sprouting by removing all but one bud permitted selection of vigorous first flushes for in vitro culture. Use of additional virulence genes from A. tumefaciens to increase transformation frequency and optimization of culture media and conditions to enhance explant cell competence for T-DNA integration and organogenesis resulted in efficient and reliable transgenic plant production. Transformed regenerants from explants, cultured in media without antibiotics, were identified by a screenable marker (either beta-glucuronidase or green fluorescent protein (GFP)), creating the possibility of generating transgenic clementine plants without antibiotic resistance marker genes. Stable integration of foreign genes was demonstrated by Southern blot analysis, and expression of these foreign genes was confirmed by detection of GFP fluorescence in leaves, floral organs and fruits of the transgenic plants.  相似文献

Development and phenobarbital (PB) induction of microsomal cytochrome P-450, cytochrome P-450 reductase, two epoxidation, and two O-demethylation activities were examined in chronologically timed populations of insecticide-susceptible (NAIDM) and -resistant (Rutgers) house flies. Measurements of these enzymes started with the pharate adult stage and ended 5 days following eclosion. Untreated insects of both strains had comparable reductase levels, whereas cytochrome P-450 and associated monooxygenase activities were 1.5-fold or more higher in Rutgers. Maximum induction, as well as toxicity, occurred at a lower PB concentration in NAIDM than Rutgers. The drug caused consistently higher increases in enzymes and activities within 12 hr of starting treatment in both strains. When PB was withdrawn from treated flies (both strains) 48 hr after treatment began, specific activities (product min?1 mg protein?1) in all enzymes returned to control values in 24 hr while metabolic capacity (product min?1 insect?1) achieved control values within 48 hr. The changes in turnover numbers (pmol product min?1 pmol P-450?1), in conjunction with the differences in the monooxygenation of the four substrates, suggest that PB treatment induced both a quantitative and qualitative change in NAIDM monooxygenation but only a quantitative change in Rutgers monooxygenation.  相似文献

In 1987 we conducted a sero-epidemiological survey in the Murcia Region (South-East Spain) to discover the prevalence and spread of PRCV-infection among breeding pigs and farms and determine the association between herd size and geographical zone with PRCV-infection. The Murcia Region was divided into four geographical zones and the farms classified by size in four categories. The random sample was statistically representative of both the breeding stock and farms in each geographical zone. We analysed 6,000 breeding pigs from 480 farms. The immunological techniques employed were indirect ELISA and blocking ELISA. The prevalence (P +/- IC) of PRCV-seropositive breeding pigs and infected breeding farms was 14.53 +/- 0.89% and 21.87 +/- 7.83% respectively. On 55% of the infected farms, the prevalence of seropositive breeding pigs was 60-100%. PRCV-infection appears spread throughout the four geographical zones of the Murcia Region. However, a significant association (p less than 0.01) was observed between geographical zone and the prevalence of PRCV-infection. A herd size of greater than 50 breeding pigs had a greater risk (p less than 0.01) of PRCV-infection.  相似文献

Coxiella burnetii, the causative agent of Query fever (Q fever), is among the most highly infectious zoonotic pathogens transmitted among livestock, with chronic effects challenging to veterinary and medical detection and care systems. Transmission among domestic livestock species can vary regionally due to herd management practices that determine which livestock species are raised, whether or not livestock are in contact with wildlife, and the susceptibility of these livestock to infection. To explore how different livestock management practices are associated with the risk of infection in multispecies environments, we carried out a comparative study of three types of herd management systems in the central Kenyan county of Laikipia: agro‐commercial, mixed conservancy/commercial, and smallholder ranches. We tested C. burnetii antibody seroprevalence in four common livestock species. Across all management types, the highest seroprevalence was in camels (20%), followed by goats (18%), sheep (13%), and cattle (6%). We observed a lower odds of testing seropositive for young compared to adult animals (adjusted OR = 0.44 [95% CI 0.24, 0.76]), and for males compared to females (adjusted OR = 0.52 [95% CI 0.33, 0.80]). Animals from mixed conservancy/commercial and smallholder operations had a higher odds of testing seropositive compared to animals from agro‐commercial ranches (adjusted OR = 5.17 [95% CI 2.71, 10.44] and adjusted OR = 2.21 [95% CI 1.17, 4.43] respectively). These data suggest that herd management practices might affect the transmission dynamics of C. burnetiiin arid African ecosystems like those seen in Kenya where several transmission modes are possible, risk of drought has promoted new livestock species such as camels, and multiple wildlife species may co‐occur with livestock on the landscape. Further longitudinal studies are needed to disentangle the mechanisms underlying these patterns, and further explore transmission patterns between wildlife, domestic animal, and human populations.  相似文献

Selected engineering properties of seeds of four red clover varieties were studied. The seeds of diploid and tetraploid forms were statistically different in terms of true and bulk density, porosity, volume, 1000 seed weight, equivalent diameter and yellowness index. The volume, 1000 seed weight and equivalent diameter were found to be statistically higher for tetraploid than for diploid forms. The accuracy of varietal classification based on the physical properties of seeds was found to be most satisfactory. Varieties of Bona and Jubilatka were identified with 100% accuracy based on the physical properties of seeds. Bona was the most conducive variety for the classification of red clover seeds based on four groups of engineering properties. The relationships between groups of properties described by total redundancy did not exceed 50% for any of variable pairs.  相似文献
