Using genomic and mass spectrometry-based proteomic methods, we evaluated gene expression, identified key activities, and examined partitioning of metabolic functions in a natural acid mine drainage (AMD) microbial biofilm community. We detected 2033 proteins from the five most abundant species in the biofilm, including 48% of the predicted proteins from the dominant biofilm organism, Leptospirillum group II. Proteins involved in protein refolding and response to oxidative stress appeared to be highly expressed, which suggests that damage to biomolecules is a key challenge for survival. We validated and estimated the relative abundance and cellular localization of 357 unique and 215 conserved novel proteins and determined that one abundant novel protein is a cytochrome central to iron oxidation and AMD formation.  相似文献

Many studies have evaluated the adequacy of alternate ingredient diets for Atlantic salmon, Salmo salar, mainly with focus on fish performance and health; however, comprehensive analysis of fillet quality is lacking, particularly for salmon fed these diets in recirculation aquaculture systems (RAS). To this end, a study was conducted comparing fillet quality and processing attributes of postsmolt Atlantic salmon fed a fishmeal‐free diet (FMF) versus a standard fishmeal‐based diet, in replicate RAS. Mean weight of Atlantic salmon fed both diets was 1.72 kg following the 6‐mo trial and survival was >99%. Diet did not affect (P > 0.05) processing and fillet yields, whole‐body proximate composition(fat, moisture, protein), fillet proximate composition, cook yield, fillet texture, color, or omega‐3 fatty acid fillet content, including eicosapentaenoic acid and docosahexaenoic acid levels. Whole‐body ash content was greater in salmon fed the FMF diet. The FMF diet resulted in a wild fish‐in to farmed fish‐out ratio of 0:1 per Monterey Bay Aquarium's Seafood Watch criteria due to its fishmeal‐free status and use of lipids from fishery byproduct. Overall, fillet quality and processing attributes were generally unaffected when feeding a diet devoid of fishmeal to postsmolt Atlantic salmon cultured in RAS. Recent reports have shown that some of the immunological aspects of Q fever, a rickettsiosis caused by Coxiella burnetii, could be related to self-antigen responses. The aim of this study was to determine the specificity of the autoantibody response of patients with acute and chronic Coxiella infections. Smooth muscle and cardiac muscle-specific autoantibodies were observed in significant percentages in acutely or chronically affected Q fever patients when compared to healthy volunteers. Moreover, the incidence of cardiac muscle-specific autoantibody was significantly higher among chronically ill patients compared to acutely ill patients. Moreover, a band of 50 kD of a HeLa extract was detected in most of the sera of individuals with chronic infections and previous sequence analysis suggests that this antigen presents a high degree of homology with the human actin elongation factor 1 alpha. Further research would be necessary to confirm if antibodies to human cytoskeletal proteins could be of clinical importance in chronically infected Q fever patients.  相似文献

Fucoidans are complex polysaccharides derived from brown seaweeds which consist of considerable proportions of L-fucose and other monosaccharides, and sulphated ester residues. The search for novel and natural bioproduct drugs (due to toxicity issues associated with chemotherapeutics) has led to the extensive study of fucoidan due to reports of it having several bioactive characteristics. Among other fucoidan bioactivities, antidiabetic and anticancer properties have received the most research attention in the past decade. However, the elucidation of the fucoidan structure and its biological activity is still vague. In addition, research has suggested that there is a link between diabetes and cancer; however, limited data exist where dual chemotherapeutic efforts are elucidated. This review provides an overview of glucose metabolism, which is the central process involved in the progression of both diseases. We also highlight potential therapeutic targets and show the relevance of fucoidan and its derivatives as a candidate for both cancer and diabetes therapy.  相似文献

Grain yield monitoring is an integral tool in the Precision Agriculture management system. When used in conjunction with a satellite-based navigation system, it provides spatial information on output variability, output response to managed inputs and is used to identify limiting resources in the crop production process. Accurately matching measured yield quantities with spatial units within a field is therefore important. At present, a simple linear time shift is employed by all commercial monitoring systems to account for the delay between GPS recorded positions and subsequent yield measurements. This study examines the internal process of grain transport to the sensor by monitoring the flow of strategically coloured grain. The flow is shown to be significantly influenced by mixing induced by threshing and auger transport processes. In contrast to the common assumption that grain moves as a spatially related cohort through to the sensor, the results suggest that a diffusion process is more realistic. A parametric model for the diffusion process is provided which suggests that from each individual yield measurement a maximum 20% of the mass could be assigned to a single spatial unit of the size that is typically allocated. The results imply that for further analyses, the inconclusive spatial origin and artificially smoothed quantities of instantaneous yield measurements should be considered.  相似文献

Because of its requirement for signaling by multiple cytokines, Janus kinase 3 (JAK3) is an excellent target for clinical immunosuppression. We report the development of a specific, orally active inhibitor of JAK3, CP-690,550, that significantly prolonged survival in a murine model of heart transplantation and in cynomolgus monkeys receiving kidney transplants. CP-690,550 treatment was not associated with hypertension, hyperlipidemia, or lymphoproliferative disease. On the basis of these preclinical results, we believe JAK3 blockade by CP-690,550 has potential for therapeutically desirable immunosuppression in human organ transplantation and in other clinical settings.  相似文献

Abstract— Different amounts of dietary phospholipid were tested for their effect on the digestibility of total neutral lipids in the diet of the prawn Penaeus monodon . A purified source of the phospholipid, L-α-phosphatidylcholine, was included at 0. 5, 10, 20 or 40 g/kg in a purified diet containing 80 g/kg olive oil as the predominant neutral lipid source. The apparent digestibility of the neutral lipid (ADNL) in these diets was determined by comparing concentrations of the digestibility marker, cholestane in the feed and faeces of the prawns. ADNL (%) increased curvilinearly with increasing dietary phospholipid content (X, %) as described by the equation: ADNL = 80.4 + 9.08X -1.705X2 (P < 0.05; R2 = 0.71); the asymptote of 92.5% was achieved with a phosphatidylcholine value of 27 g/kg. These results demonstrate that phosphatidylcholine significantly increases the digestibility of neutral lipids by penaeid prawns. However, olive oil was used as the predominant neutral lipid source to examine the effects with a relatively poorly digestible lipid source. The effects of phosphatidylcholine on more readily digested lipid sources such as fish oils, may not be as great, but still be sufficient to improve the utilisation of these nutrients.  相似文献
