To date Cryptosporidium muris has been identified by microscopy and genotyping in cats in two studies. We report morphological and genetic evidence of a mixed C. muris and C. felis infection in a cat and provide the first histological, immunohistochemical, in situ hybridisation and genetic confirmation of a C. muris infection in the stomach of a cat. The cat suffered persistent diarrhoea after the initial consultation, which remained unresolved, despite several medical interventions. Further studies are required to determine the range, prevalence and clinical impact of Cryptosporidium species infecting cats. 相似文献

Theileria parva antigens recognized by cytotoxic T lymphocytes (CTLs) are prime vaccine candidates against East Coast fever in cattle. A strategy for enhancing induction of parasite-specific T cell responses by increasing recruitment and activation of dendritic cells (DCs) at the immunization site by administration of bovine Flt3L and GM-CSF prior to inoculation with DNA vaccine constructs and MVA boost was evaluated. Analysis of immune responses showed induction of significant T. parva-specific proliferation, and IFN-γ-secreting CD4(+) and CD8(+) T cell responses in immunized cattle. However, antigen-specific CTLs were not detected. Following lethal challenge, 5/12 immunized cattle survived by day 21, whereas all the negative controls had to be euthanized due to severe disease, indicating a protective effect of the vaccine (p<0.05). The study demonstrated the potential of this technology to elicit significant MHC class II and class I restricted IFN-γ-secreting CD4(+) and CD8(+) T cells to defined vaccine candidate antigens in a natural host, but also underscores the need to improve strategies for eliciting protective CTL responses. 相似文献

Chalkbrood (Ascosphaera apis) and stonebrood (Aspergillus flavus) are well known fungal brood diseases of honeybees (Apis mellifera), but they have hardly been systematically studied because the difficulty of rearing larvae in vitro has precluded controlled experimentation. Chalkbrood is a chronic honeybee-specific disease that can persist in colonies for years, reducing both brood and honey production, whereas stonebrood is a rare facultative pathogen that also affects hosts other than honeybees and can likely survive outside insect hosts. Hive infection trials have indicated that accidental drops in comb temperature increase the prevalence of chalkbrood, but it has remained unclear whether virulence is directly temperature-dependent. We used a newly established in vitro rearing technique for honeybee larvae to test whether there are systematic temperature effects on mortality induced by controlled infections, and whether such effects differed between the two fungal pathogens. We found that increasing spore dosage at infection had a more dramatic effect on mortality from stonebrood compared to chalkbrood. In addition, a 24h cooling period after inoculation increased larval mortality from chalkbrood infection, whereas such a cooling period decreased mortality after stonebrood infection. These results raise interesting questions about honeybee defenses against obligate and facultative pathogens and about the extent to which stress factors in the host (dis)favor pathogens with lesser degrees of specialization. 相似文献

This study investigated the storage of nitrogen (N) and phosphorus (P) in the biomass, bed sediments and water column of representative reaches of a sub-tropical river, the upper Brisbane River (UBR), Queensland, Australia, and contrasted instream storage with total wet season exports. In reaches which contained accumulated fine sediments, more than 87% of total P and between 50% and 92% of total N were stored in the surface sediments. The lower proportion of N in sediment at some sites was attributed to substantial differences in the N/P ratios of sediments and macrophytes. At one site, the riverbed was dominated by cobbles and boulders and total nutrient stocks were comparatively low and dominated by the biomass. In reaches with a narrow channel and intact riparian cover, biomass N and P were stored predominately in leaf litter, while in wider unshaded reaches, macrophytes dominated. Total instream storage in the mid to lower reaches of the UBR was ??50.9 T for N and ??18.1 T for P. This was considerably higher than total wet season N (??15.6 T) and P (??2.7 T) exports from the UBR. The first flow event in the river after a prolonged period of no flow resulted in the export of free-floating, emergent species Azolla. The estimated biomass of Azolla in the mid to lower reaches of the river was equivalent to approximately 24% and 9% of the total N and P flux, indicating that this may be a significant, previously unaccounted for, source at peak flow. 相似文献

The Diep River is a major freshwater ecosystem in the Western Cape, South Africa. Although it is surrounded by many sources of metal pollution, the actual metal levels in this river system are unknown. Wetland plants are known to accumulate metals and may possibly be used as biomonitors of metal contamination in a river system. One such species, the sedge Bolboschoenus maritimus, is found in abundance along the banks of this river. The aim of this study was to investigate and monitor the degree of metal contamination in the water and sediments of the lower Diep River, as well as to study the seasonal accumulation and distribution of metals in B. maritimus, and the use thereof as biomonitor species. Two sampling sites were used: one site above the wetland section of the river, receiving runoff mainly from agricultural lands (site 1), and one site close to the river mouth (site 2), exposed to several possible pollution sources. Water, sediment, and plant (root, leaf and stem) samples were collected seasonally for 1 year and analyzed for Al, Zn, Cu, and Fe. There was greater bioaccumulation of metals by plants at site 2, due to greater bioavailability of metals. B. maritimus was shown to be a root accumulator of metals. Seasonal fluctuations in root, stem, and leaf metal concentrations did not follow seasonal sediment concentration patterns. However, using B. maritimus as test species did provide valuable additional information to sediment and water analyses. More extensive research is needed to conclude whether this species is an effective biomonitor in the lower Diep River environment. 相似文献

The enzymatic hydrolysis of brewers' spent grain (BSG) has been investigated through treatment with commercial carbohydrases and proteases. Resultant residues were then chemically fractionated and delignified. Enzymatic treatments released 25-30% of the BSG mass and yielded precursors suitable for subsequent conversion to potentially value-added products. Controlled chemical fractionation selectively solubilized arabinoxylan but with no differences apparent due to prior enzyme treatment. The loss of non-polysaccharide components during alkali treatment suggests the presence of a high proportion of alkali-soluble lignin. Further delignification of the alkali-insoluble residues and further chemical fractionation released the remaining hemicellulose, to yield a residue which was >90% cellulose. Further knowledge of the properties and interaction between BSG polymers will facilitate an improved enzyme-assisted total deconstruction of BSG and hence the exploitation of its biomass. 相似文献

A rapid and comprehensive qualitative method has been developed to characterize the different classes of polyphenols, such as anthocyanins, flavonols, phenolic acids, and flavanols/proanthocyanidins, in grape products. The detection was achieved by two runs with the same LC gradient in different MS ionization modes and mobile phase modifiers (positive ionization mode and 0.4% trifluoroacetic acid for anthocyanins and flavonols; negative ionization mode and 0.1% formic acid for phenolic acids and flavanols). From an analysis of the MS and UV data and in comparison with the authenticated standards, a total of 53 compounds were identified, including 33 anthocyanins, 12 flavonols, 4 phenolic acids, and 4 flavanols/proanthocyanidins. With the method developed, a survey was then conducted to qualitatively assess the composition of polyphenols among 29 different grape products including original grape, grape juice, grape wine, and grape-derived dietary supplements, and their chemical profiles were systematically compared. Rat‐associated zoonoses transmitted through faeces or urine are of particular concern for public health because environmental exposure in homes and businesses may be frequent and undetected. To identify times and locations with greater public health risks from rats, we investigated whether rat characteristics, environmental features, socioeconomic factors, or season could predict rat infection risk across diverse urban neighbourhoods. In partnership with a pest management company, we sampled rats in 13 community areas along an income gradient in Chicago, a large city where concern about rats has increased in recent years. We collected kidneys for Leptospira spp. testing and colon contents for aerobic bacteria such as Salmonella spp. and Escherichia coli. Of 202 sampled rats, 5% carried Leptospira spp. and 22% carried E. coli. Rats were significantly more likely to carry Leptospira spp. on blocks with more standing water complaints in higher‐income neighbourhoods (OR = 6.74, 95% CI: 1.54–29.39). Rats were significantly more likely to carry E. coli on blocks with more food vendors (OR = 9.94, 2.27–43.50) particularly in low‐income neighbourhoods (OR = 0.26, 0.09–0.82) and in the spring (OR = 15.96, 2.90–88.62). We detected a high diversity of E. coli serovars but none contained major virulence factors. These associations between environmental features related to sanitation and infection risk in rats support transmission through water for Leptospira spp. and faecal–oral transmission for E. coli. We also found opposing relationships between zoonotic infection risk and income for these two pathogens. Thus, our results highlight the importance of sanitation for predicting zoonotic disease risks and including diverse urban areas in pathogen surveillance to mitigate public health risks from rats. 相似文献
