In this work, in vitro fermentation of alternansucrase raffinose-derived oligosaccharides, previously fractionated according to their degree of polymerization (DP; from DP4 to DP10), was carried out using small-scale pH-controlled batch cultures at 37 °C under anaerobic conditions with human feces. Bifidogenic activity of oligosaccharides with DP4-6 similar to that of lactulose was observed; however, in general, a significant growth of lactic acid bacteria Bacteroides , Atopobium cluster, and Clostridium histolyticum group was not shown during incubation. Acetic acid was the main short chain fatty acid (SCFA) produced during the fermentation process; the highest levels of this acid were shown by alternansucrase raffinose acceptor pentasaccharides at 10 h (63.11 mM) and heptasaccharides at 24 h (54.71 mM). No significant differences between the gas volume produced by the mixture of raffinose-based oligosaccharides (DP5-DP10) and inulin after 24 h of incubation were detected, whereas lower gas volume was generated by DP4 oligosaccharides. These findings indicate that novel raffinose-derived oligosaccharides (DP4-DP10) could be a new source of prebiotic carbohydrates.  相似文献

New ecosystems are being actively mined for new bioactive compounds. Because of the large amount of unexplored biodiversity, bacteria from marine environments are especially promising. Further, host-associated microbes are of special interest because of their low toxicity and compatibility with host health. Here, we identified and characterized biosynthetic gene clusters encoding antimicrobial compounds in host-associated enterococci recovered from fecal samples of wild marine animals remote from human-affected ecosystems. Putative biosynthetic gene clusters in the genomes of 22 Enterococcus strains of marine origin were predicted using antiSMASH5 and Bagel4 bioinformatic software. At least one gene cluster encoding a putative bioactive compound precursor was identified in each genome. Collectively, 73 putative antimicrobial compounds were identified, including 61 bacteriocins (83.56%), 10 terpenes (13.70%), and 2 (2.74%) related to putative nonribosomal peptides (NRPs). Two of the species studied, Enterococcus avium and Enterococcus mundtti, are rare causes of human disease and were found to lack any known pathogenic determinants but yet possessed bacteriocin biosynthetic genes, suggesting possible additional utility as probiotics. Wild marine animal-associated enterococci from human-remote ecosystems provide a potentially rich source for new antimicrobial compounds of therapeutic and industrial value and potential probiotic application.  相似文献

In classical enzymology, intermediates and transition states in a catalytic mechanism are usually inferred from a series of biochemical experiments. Here, we derive an enzyme mechanism from true atomic-resolution x-ray structures of reaction intermediates. Two ultra-high resolution structures of wild-type and mutant d-2-deoxyribose-5-phosphate (DRP) aldolase complexes with DRP at 1.05 and 1.10 angstroms unambiguously identify the postulated covalent carbinolamine and Schiff base intermediates in the aldolase mechanism. In combination with site-directed mutagenesis and (1)H nuclear magnetic resonance, we can now propose how the heretofore elusive C-2 proton abstraction step and the overall stereochemical course are accomplished. A proton relay system appears to activate a conserved active-site water that functions as the critical mediator for proton transfer.  相似文献

Initial images of Venus's south pole by the Venus Express mission have shown the presence of a bright, highly variable vortex, similar to that at the planet's north pole. Using high-resolution infrared measurements of polar winds from the Venus Express Visible and Infrared Thermal Imaging Spectrometer (VIRTIS) instrument, we show the vortex to have a constantly varying internal structure, with a center of rotation displaced from the geographic south pole by ~3 degrees of latitude and that drifts around the pole with a period of 5 to 10 Earth days. This is indicative of a nonsymmetric and varying precession of the polar atmospheric circulation with respect to the planetary axis.  相似文献

Juveniles of the Caribbean scallops Argopecten nucleus and Nodipecten nodosus were suspended in pearl nets at densities of 25%, 30% and 50% coverage of the bottom of the net (BC) during days. Additionally, adults of A. nucleus (>35 mm) were placed in lantern nets at densities of 40%, 50% and 65% BC during days. Stocking density affected the growth of animals, with higher growth rates obtained at the lower densities. Although the survival of the two scallops was not influenced at the densities studied, the 15-day net changes, and culture in water of <28 °C produced better survival of A. nucleus and N. nodosus respectively. Predators of the families Cymatiidae, Grapsidae, Portunidae, Majidae and Xanthidae were found in the culture nets of juvenile A. nucleus , but during the adult phase they were absent. Predators exhibited a dense-independent behaviour in relation to scallops. There was not any association between predator frequency and survival of A. nucleus .  相似文献

Five hormonal treatments with human chorionic gonadotropin (hCG) were tested for the induction of maturation and spermiation in male farmed eels. The main aim was to optimize previously used hormonal treatments to achieve shorter induction treatments, longer spermiation periods and/or higher sperm quality. Fish treated for just 3 weeks (treatment E) or until the onset of spermiation (treatment C) showed the worst results, while the treatment consisting of weekly administration of 1.5 IU hCG g?1 fish (treatment A) induced the highest percentage of spermiating males, the highest number of sperm samples and sperm volumes and densities similar to the rest of the treatments (B: half hormone dosage, or D: biweekly administration). Evaluation of the sperm quality was performed by computer‐assisted sperm analysis (CASA), considering the percentage of total motile spermatozoa, the percentage of fast and medium‐velocity spermatozoa, as well as different motility parameters. Sperm samples from A‐D groups showed between 44% and 54% motile spermatozoa, and between 10% and 15% fast spermatozoa, while samples from E‐treated males showed 0% motile cells. No significant differences were found in the spermatozoa straight line velocity (VSL), curvilinear velocity (VCL) or the angular velocity (VAP), neither spermatozoa beating cross frequency (BCF) between A–D groups.  相似文献

This study aimed to analyse the effects of different water temperatures on the growth, survival and blood physiological parameters of Lophiosilurus alexandri. Juveniles measuring 12.6 ± 0.5 cm and weighing 30.6 ± 3.6 g were subjected to four water temperature regimes: 23, 26, 29 and 32°C with four replicates. The animals were fed twice daily with formulated diet. The experiment lasted for 35 days. Survival was 100% at all water temperatures, and the best temperature for growth was estimated to be 27.8°C using a quadratic regression model. The estimated temperatures for the best feed conversion and highest feed consumption rates were 26.2 and 28.8°C respectively. The fat visceral‐somatic index was highest at 32°C. The highest haemoglobin and haematocrit values were 29.4 and 32.1°C respectively. For plasma protein and cholesterol, the lowest values estimated were 23.9 and 24.1°C respectively. Glucose and liver nitrogen content increased with rising temperature. Higher values of total ammonia in the water were observed at higher temperatures. No effects of temperature were seen on the amounts of muscle dry matter, nitrogen, energy content and triglycerides or on the hepatosomatic index. Water temperature has an important role in the developmental and blood physiological parameters of juvenile L. alexandri, and the quadratic regression model showed that the appropriate temperature for growth is between 27 and 28°C.  相似文献

The diatom Chaetoceros muelleri was grown in outdoor mass cultures under the winter, spring and summer conditions of Bahia Kino, Sonora, NW Mexico. The solar irradiance in winter was close to 60% of that available in spring and summer, but the cell concentrations and the harvestable biomass were one order of magnitude higher in spring and summer than in winter. There was no difference between the biomass harvested after 2 and 3 days in winter and summer, whereas in spring it was higher after 3 days. The protein content was significantly lower in winter, and carbohydrates and lipids were higher in winter and spring. The number of cells and the amount of harvestable biomass of outdoor cultures of C. muelleri depend on the temperature prevailing in each season, which causes significant differences in growth rates and in biochemical composition.  相似文献

Negative refraction in metamaterials has generated great excitement in the scientific community. Although negative refraction has been realized in microwave and infrared by using metamaterials and by using two-dimensional waveguide structures, creation of a bulk metamaterial showing negative refraction at visible frequency has not been successful, mainly because of the significant resonance losses and fabrication difficulties. We report bulk metamaterials made of nanowires that show such negative refraction for all incident angles in the visible region. Moreover, the negative refraction occurs far from any resonance, resulting in a low-loss and a broad-band propagation at visible frequencies. These remarkable properties can substantially affect applications such as imaging, three-dimensional light manipulation, and optical communication.  相似文献
