An alternative use of solid organic and inorganic residues as fertilizers from a Kraft pulp industry was studied. Residues of inorganic nature, such as ashes, fly‐ashes, dregs, grits, as well those rich in organic matter, primary sludge and brown stock rejects, were examined for plant growth enhancement. These residues, all alkaline in nature, used in different concentrations together with soil, bark, organic soil or mixed with a nutrient solution, were tested on the growth of Monterey pine (Pinus radiata), Eucalyptus globulus, rice (Oryza sativa. cv. ‘Diamante’), and duckweed (Lemna minor) under greenhouse and in‐vitro conditions, respectively. Responses varied according to plant species, type, and waste content in combination with substrate. For Monterey pine, substrates including ash, fly‐ash, and dregs promoted growth; in Eucalyptus seedlings dregs and fly‐ash were also beneficial. Primary sludge and ash were favorable for rice growth. Duckweed increased frond number and plant biomass when grown in water containing fly‐ash and primary sludge extracts, combined with nutrient salts.  相似文献

The effect of a granular formulation of sodium azide, applied annually to pine nursery beds at rates of 0, 67.2, and 134.5 kg a. i. ha?1 under water seal or plastic seal, on soil fungal populations was determined over a 3-year period. Populations of fungi in the soil decreased following application of the sodium azide each year; the greatest decrease occurred at the highest rate of application. Populations of fungi in soil treated with the azide generally remained lower than in the controls throughout each of the 3 years; however, the population disparity between treated and control plots decreased in magnitude with each succeeding year. Populations of Trichoderma spp., in plots treated with 134.5 kg sodium azide ha?1, increased 2 weeks after treatment each year, and the population peaks increased in magnitude each year. In addition the effect of sodium azide (technical grade > 99%) at concentrations of 0, 2, 5, 10, 20 and 50 μg ml?1 in potato dextrose agar and blackstrap molasses agar media was determined in vitro for 14 isolates of Trichoderma harzianum. Growth and sporulation differed among the isolates and between the two media tested. Generally, the azide temporarily inhibited growth of the fungi, but the majority of the isolates were able to grow on either medium containing 50 μg sodium azide ml?1, although sporulation was more profuse on the molasses than on the potato dextrose agar medium.  相似文献

Levamisole, morantel, fenbendazole, or ivermectin was administered at 2-week intervals from May 1 through Sept 14, 1985, to breeding ewes (20 ewes/drug) infected with various gastrointestinal nematodes. All ewes had fewer gastrointestinal nematode eggs per gram of feces (epg) after 2 treatments, compared with pretreatment epg counts. Ewes administered ivermectin continued to have a low mean epg (0 to 3) throughout the study. The mean epg counts of ewes treated with levamisole increased from 3 to 483 during the study. This increase was similar to that of ewes treated with morantel (7 to 485 epg). The mean epg count of fenbendazole-treated ewes increased from 4 to 192 during the study. By the end of the study, the mean epg counts when expressed as a percentage of the pretreatment epg counts were 4% (ivermectin), 249% (fenbendazole), 627% (levamisole), and 630% (morantel). With the exception of the ivermectin-treated ewes, the epg count increased almost linearly in the ewes after the 2nd anthelmintic treatment. These data indicate that the gastrointestinal nematodes (including Haemonchus contortus) may have developed more resistance to levamisole and morantel than to fenbendazole. On the basis of the epg counts, resistance to ivermectin did not develop during the 4.5-month treatment period. The percentage of ewes shedding eggs after 2, 4, and 6 anthelmintic treatments was lowest for ewes treated with ivermectin (20%) and was similar (40%) for ewes treated with 1 of the other 3 anthelmintics. Isolates of bovine herpesvirus-1 (BHV-1) recovered from tissue explants of trigeminal ganglia of clinically healthy cattle were studied in vitro and in an animal model, and their characteristics were compared with those of vaccine and field strains of BHV-1. The isolates could be distinguished by their plaque size on cell monolayers, but were not significantly different in their thermal inactivation profiles at 48 C. Temperature-sensitive mutants were not found among the isolates when they were grown at 41 C. Selected isolates had different pathogenicity when inoculated in young rabbits.  相似文献

Most animals adapt readily to increased supplies of carbohydrate in the intestinal lumen by increasing enzymes for degradation and increasing glucose transporter activity. However, the extent of upregulation of Na+-dependent glucose cotransporter 1 (SGLT1) activity and content in response to increased delivery of carbohydrate to the small intestinal lumen of ruminants is unclear. Therefore, an experiment was conducted to determine the effect of glucose and starch hydrolysate on the activity and abundance of SGLT1 in the small intestine of steers. In a randomized complete block design, 40 crossbred beef steers (243+/-2 kg BW) were fed 0.163 Mcal of ME/(kg BW0.75(d; W), 0.215 Mcal of ME/(kg BW0.75 x d; 2M), or 0.163 Mcal ME/(kg BW0.75 x d) and infused for 35 d into the rumen (R) or abomasum (A) with 12.6 g/(kg BW0.75 x d) of starch hydrolysate (S) or into the abomasum with 14.4 g/(kg BW0.75 x d) of glucose (G). Steers were slaughtered, and brush-border membrane vesicles were prepared from the small intestinal samples obtained from five equidistant sites along the intestine. Maltase activity in vesicles and homogenates differed with intestinal sampling site (quadratic, P < 0.001). Steers on the AG treatment yielded a greater intestinal maltase activity (38 nmol glucose x mg protein(-1) x min(-1)) compared with the AS, RS, W, or 2M treatments (34, 26, 23, and 23 nmol glucose x mg protein(-1) x min(-1) respectively [SEM = 3; P = 0.02]). Sodium-dependent glucose uptake averaged 18.4+/-3.94 pmol glucose/(mg protein x s) and was not affected by treatment, but uptake decreased distally along the intestine (P < 0.001). There was no effect of treatment on SGLT1 protein abundance, but SGLT1 protein abundance increased linearly from the duodenum to the ileum (P = 0.05). The inverse relationship between glucose uptake and SGLT1 abundance suggests that the regulation of brush border Na+-dependent glucose transport capacity is complex, involving factors other than the presence of luminal carbohydrate.  相似文献

Sampling an intact sequence of oceanic crust through lavas, dikes, and gabbros is necessary to advance the understanding of the formation and evolution of crust formed at mid-ocean ridges, but it has been an elusive goal of scientific ocean drilling for decades. Recent drilling in the eastern Pacific Ocean in Hole 1256D reached gabbro within seismic layer 2, 1157 meters into crust formed at a superfast spreading rate. The gabbros are the crystallized melt lenses that formed beneath a mid-ocean ridge. The depth at which gabbro was reached confirms predictions extrapolated from seismic experiments at modern mid-ocean ridges: Melt lenses occur at shallower depths at faster spreading rates. The gabbros intrude metamorphosed sheeted dikes and have compositions similar to the overlying lavas, precluding formation of the cumulate lower oceanic crust from melt lenses so far penetrated by Hole 1256D.  相似文献

The aim of the present study is to highlight the advantages of real-time quantitative PCR intended to aid in the diagnosis and monitoring of canine leishmaniosis. Diagnosis of canine leishmaniosis is extremely challenging, especially in endemic areas, due to the diverse and non-specific clinical manifestations, and due to the high seroprevalence rate in sub-clinical dogs. Veterinarian clinicians are usually confronted with cases that are compatible with the disease, and with several diagnostic tests, sometimes with contradictory results. We have developed a new TaqMan assay, targeting the kinetoplast, applied to 44 samples of bone marrow aspirate or peripheral blood. The dynamic range of detection of Leishmania DNA was established in 7 logs and the limit of detection is 0.001 parasites in the PCR reaction. At the time of diagnosis parasitemia ranges from less than 1 to 10(7)parasites/ml. The ability to quantify the parasite burden allowed: (i) to elucidate the status of positive dogs by conventional PCR, although larger studies are necessary to clarify the dividing line between infection and disease, (ii) to estimate the kinetics of the parasite load and the different response to the treatment in a follow-up and (iii) to validate blood as less invasive sample for qPCR. The continuous data provided by real-time qPCR could solve the dilemma for the clinician managing cases of canine leishmaniosis by differentiating between Leishmania-infected dogs or dogs with active disease of leishmaniosis.  相似文献
