The aim of this work was to quantify NO, O2? and ONOO? production during heparin‐induced capacitation of cryopreserved bovine spermatozoa. A time dependent hyperbolic increase was observed for heparin‐dependent capacitation, O2 uptake, and NO production. Conversely, O2? production was increased during the first 15 min of incubation, showing a decrease from this time until 45 min. At 15 min of heparin incubation, a threefold increase in O2 consumption (5.9 ± 0.6 nmol/min × 107 cells), an enhancement in NO release (1.1 ± 0.2 nmol/min × 107 cells), and a five‐fold increase in O2? production (1.3 ± 0.07 nmol/min × 107 cells), were observed. Peroxynitrite production rate was estimated taking into account NO and O2? generation and the second‐order rate constant of the reaction between these species. To conclude, heparin‐induced capacitation of cryopreserved bovine spermatozoa activates (i) mitochondrial O2 uptake by high ADP levels due to increased energy requirements, (ii) NO production by a constitutive NOS and (iii) O2? production by a membrane‐bound NAD(P)H oxidase. The products of both enzymes are released to the extracellular space and could be involved in the process of sperm capacitation.  相似文献

Human trampling destroys seedlings and saplings without regard to species in urban park forests and the addition of deer browsing compounds the losses. The unexamined research question is: what is the effect of white deer browsing in the absence of human trampling? Radnor Lake State Natural Area, Nashville, TN, USA has been protected from off-pathway human transit since 1973 and white-tailed deer (Odocoileus virginianus Zimmerman) were restored to the Natural Area in 1980. From 1976 to 2007, a plot with tagged trees in the mesic slope forest showed the tree population for 16 species decreased, two remained stable, and one increased. The pattern of increase for sugar maple (Acer saccharum Marsh) was 57 new trees added but 46 trees were lost, which appears as a population increase from 1976 to 1996 and a decrease in 2007. In all five forest communities, the total tree stems per ha declined from 1974 to 2008. During the period 1994–2008, O. virginianus over browsing decimated the seedling population of all species and caused the total for stems per ha for saplings to become smaller than the total stems per ha for trees in each community except the ravine forest, which had the greatest loss of trees. The only consistent change in trees across the five communities when comparing 1974–2008 was the significant tree importance value increase for A. saccharum. The conflicting significant changes for major species, other than A. saccharum, across the Natural Area forest communities informs management for other urban park forests—browsing by O. virginianus results in increased numbers for trees and saplings in the communities the species are well adapted to grow and reproduce in and fewer trees and saplings in the communities with environmental conditions that are not well suited to the species.  相似文献

Testes of last-instar larvae of the tobacco budworm release five times more ecdysteroid into incubation medium (judged by radioimmunoassay) in 2.5 hours than is found in testis homogenates. Incubation of testicular components indicates that the testis sheath may be the site of ecdysteroid synthesis. Fractionation of hemalymph, testis homogenate, and incubation medium by high-performance liquid chromatography produces a distinct ecdysteroid pattern in each case. Thus, released testis ecdysteroids are probably converted to other forms for use, sequestration, or general circulation. Their functions are unknown.  相似文献

Impact of lower atmospheric carbon dioxide on tropical mountain ecosystems   总被引:1,自引:0,他引:1  
Carbon-isotope values of bulk organic matter from high-altitude lakes on Mount Kenya and Mount Elgon, East Africa, were 10 to 14 per mil higher during glacial times than they are today. Compound-specific isotope analyses of leaf waxes and algal biomarkers show that organisms possessing CO2-concentrating mechanisms, including C4 grasses and freshwater algae, were primarily responsible for this large increase. Carbon limitation due to lower ambient CO2 partial pressures had a significant impact on the distribution of forest on the tropical mountains, in addition to climate. Hence, tree line elevation should not be used to infer palaeotemperatures.  相似文献

Vineyards were surveyed for grapevine leafroll-associated viruses and their insect vectors in New York State’s Finger Lakes region in 2006–2008. Grape mealybug, Pseudococcus maritimus (Erhorn) (Hemiptera: Pseudococcidae), European Fruit Lecanium, Parthenolecanium corni (Bouche), and Cottony Maple Scale, Pulvinaria acericola (Walsh and Riley) (Hemiptera: Coccidae) were identified as vector species in this region. An increase in the incidence of Grapevine leafroll-associated virus 1 (GLRaV-1) and GLRaV-3 was observed in 8 of the 20 vineyards surveyed, which implies transmission by these insect vectors. Two of the vineyards for which a temporal increase in disease incidence was documented were then used to evaluate the efficacy of foliar applications of horticultural oil and two classes of insecticides for control of P. maritimus and for slowing virus spread over 2 years of vine protection. Delayed dormant applications of horticultural oil contributed to control of early season crawlers; however, this was not the case for control of summer populations. Applications of acetamiprid and spirotetramat achieved control in summer populations; however, spirotetramat outperformed acetamiprid in percent reduction of treated compared with control vines and in a side-by-side trial. Vines treated with spirotetramat had a lower percentage of new vines testing positive for GLRaV-1 than control vines after 2 years, while no other spray program altered the increase in incidence of GLRaV-1 or -3.  相似文献
