Chromic oxide is used as an inert marker to measure apparent digestibility of feeds in insects, terrestrial, and aquatic animals. Quantitative determination of chromic oxide content in the sample requires the oxidation of water insoluble trivalent chromic oxide to its water-soluble hexavalent form. The two commonly used oxidizing agents are 70% perchloric acid or a mixture of sodium molybdate, sulfuric, and perchloric acid. Chromic oxide content of the oxidized solution is then measured against known standards either directly by spectrophotometry in the visible range at 350, 370, or 440 nm or after forming a colored complex with diphenylcarbazide (DPC) by colorimetry at 540 nm. This study compared the two methods of oxidation followed by spectrophotometry at the three wavelengths and by DPC colorimetry. DPC colorimetry gave precise results than the direct measurement of dichromate ion, irrespective of the method of oxidation used. Ash from samples oxidized by perchloric acid and quantified by DPC colorimetry gave a better measure of actual chromic oxide content as compared to the other methods tested.  相似文献

Summary Isozyme analyses of genetic diversity in Vigna unguiculata were performed to determine genetic relationships and level of genetic diversity between wild and cultivated cowpea. Thirty-four cultivated accessions of V. unguiculata, 56 wild accessions of V. unguiculata, and six accessions representing five related wild Vigna species were analyzed. Ten enzyme systems were polymorphic within Vigna unguiculata: AAT, ACO, G6PDH, DIAP, LAP, MUE, ME MDH, PRX, and SOD. Fourteen of 24 putative loci (58%) were polymorphic within wild V. unguiculata, but only one locus (4%) was polymorphic within cultivated cowpea; when five related Vigna species were examined, 21 of the 24 bands of activity showed polymorphisms (88%) adding 33 alleles to the 48 identified within V. unguiculata. In one F2 population of 68 plants (UCDVg 36 × UCDVg 21) a loose linkage was indicated between Diap-2 and G6pd-1 (2 = 15.39; p = 0.004) with an estimated distance of 36.0 cM ± 5.02 (recombination (r) = 0.31). Also in another F2 population of 38 plants (CB 88 × UCDVg 21) a loose linkage was indicated between Lap-1 and Prx (\gC2 = 9.62; p = 0.047) with an estimated distance of 39.8 cM ± 7.0 (r = 0.33). Total genetic diversity (HT) was 0.085 over all of the accessions including the one classified as V. nervosa. Within accession diversity (Hs) approached zero and between accession diversity (Dsi) was responsible for all of the genetic diversity present. Therefore the coefficient of gene differentiation (GST = DSTIIT) approached 1. Absolute gene differentiation (Dm) was 0.087. Two of the nine segregations in this study were skewed. Immunosuppressive agents were given to baby guinea pigs to develop a leukocytopenic baby guinea pig model. Azathioprine in different doses, and cyclophosamide and methylprednisolone combinations did not result in predictable leukocytopenia without significant mortality. A combination of azathioprine 20mg and cyclophosphamide 20mg administered once daily for 5 days to 42 baby guinea pigs resulted in a mean white blood cell count of 2531/mm3 ±1198 (a reduction of 3931 ± 1413 from pretreatment values). The mean counts for polymorphonuclear leukocytes and lymphocytes on day 6 were 398/mm3 and 2035/mm3, respectively. Only 7% mortality was seen in these animals. This model can be utilized to study a variety of bacterial and viral infections in “immunocompromised hosts”.  相似文献

The effects of single IV injections of sodium bicarbonate (0.5 mEq/kg of body weight, 1 mEq/kg, 2 mEq/kg, and 4 mEq/kg) on serum osmolality, serum sodium, chloride, and potassium concentrations, and venous blood gas tensions in 6 healthy cats were monitored for 180 minutes. Serum osmolality increased and remained significantly (P less than 0.05) increased for 120 minutes in cats given 4 mEq of sodium bicarbonate/kg. Serum sodium was increased significantly (P less than 0.05) for 30 minutes in cats given 4 mEq of sodium bicarbonate/kg. Serum sodium decreased and remained significantly (P less than 0.05) decreased for 120 minutes in cats given 1 g of 20% mannitol/kg, and serum osmolality was significantly (P less than 0.05) decreased at 30 and 60 minutes. Serum chloride decreased significantly (P less than 0.05) for 10 minutes in cats given 1 mEq of sodium bicarbonate/kg, and was significantly decreased for 30 minutes in cats given 2 mEq and 4 mEq of sodium bicarbonate/kg. Serum chloride decreased and remained significantly (P less than 0.05) decreased for 30 minutes in cats given 1 g of 20% mannitol/kg. Serum sodium and serum osmolality did not change significantly (P less than 0.05) in cats given 4 ml of 0.9% sodium chloride/kg. Serum potassium decreased significantly (P less than 0.05) for 10 minutes in cats given 1 mEq of sodium bicarbonate/kg, and for 120 minutes in cats given 2 mEq/kg or 4 mEq/kg. The evolution of strong organophosphorus multiresistance, suppressible by S,S,S-tributyl phosphorotrithioate (TBPT), in a California strain of Culex pipiens fatigans was examined by further selection with temephos, alone and in combination with the synergists TBPT or piperonyl butoxide (PB). Selection by temephos and temephos + PB increased resistance to higher levels. However, selection by temephos + TBPT virtually abolished TBPT-suppressible resistance while preventing the emergence of significant alternative resistance mechanisms. The phenomenon of synergism may enable the extended use of an insecticide where alternative resistance mechanisms are either absent or of low efficiency in the target population.  相似文献

: Milk samples from 285 cows in 15 dairy herds were collected for bacteriological analysis. Cows were selected on the basis of a somatic cell count (SCC) exceeding 200,000 cells per ml at the three most recent milk recordings prior to sampling. Staphylococcus aureus and Streptococcus uberis were the predominant isolates accounting for 21% (n = 61) and 19% (n = 53) of isolates, respectively. Streptococcus uberis was more frequently isolated from split-calving herds than from spring-calving herds and this difference was statistically significant (P < 0.005). Herds with suboptimal housing had a significantly greater prevalence of S. uberis than did herds where housing was adequate (P < 0.005). The isolation rates for S. aureus was significantly greater in herds where parlour hygiene was suboptimal (P < 0.05).  相似文献

Infection prevalence in a population often is estimated from grouped binary data expressed as proportions. The groups can be families, herds, flocks, farms, etc. The observed number of cases generally is assumed to have a Binomial distribution and the estimate of prevalence is then the sample proportion of cases. However, the individual binary observations might not be independent--leading to overdispersion. The goal of this paper was to demonstrate random-effects models for the estimation of infection prevalence from data which are correlated and in particular, to illustrate a nonparametric random-effects model for this purpose. The nonparametric approach is a relatively recent addition to the random-effects class of models and does not appear to have been discussed previously in the veterinary epidemiology literature. The assumptions for a logistic-regression model with a nonparametric random effect were outlined. In a demonstration of the method on data relating to Salmonella infection in Irish pig herds, the nonparametric method resulted in the classification of herds into a small number of distinct prevalence groups (i.e. low, medium and high prevalence) and also estimated the relative frequency of each prevalence category in the population. We compared the estimates from a logistic model with a nonparametric distribution for the random effects with four alternative models: a logistic-regression model with no random effects, a marginal model using a generalised estimating equation (GEE) and two methods of fitting a Normally distributed random effect (the GLIMMIX macro and the NLMIXED procedure both in SAS). Parameter estimates from random-effects models are not readily interpretable in terms of prevalences. Therefore, we outlined two methods for calculating population-averaged estimates of prevalence from random-effects models: one using numerical integration and the other using Monte Carlo simulation.  相似文献

The objectives of this study were to investigate the diversity of Escherichia coli O157:H7 isolates obtained over a 3-month period from a cattle feedlot in order to assess the relationship between environmental and faecal isolates and to determine the pattern of transmission of E. coli O157:H7 between groups of cattle. Faecal samples were obtained from cattle housed in four adjacent feedlot pens at monthly intervals, with environmental pen samples collected simultaneously. All E. coli O157:H7 isolates obtained were examined by pulsed field gel electrophoresis (PFGE), polymerase chain reaction (PCR) to detect eaeA, ehxA, stx1 and stx2 genes and antibiotic sensitivity profiling. Ten isolates were subjected to acid shock to imitate conditions in the acidic cattle abomasum and assess the effect on PFGE profiles. E. coli O157:H7 was isolated from 69 faecal samples and 26 environmental samples. All isolates (n=95) carried the genes for eaeA, ehxA and stx2 and were sensitive to all antibiotics tested. The PFGE profiles of all isolates differed by no more than two bands and clustered within 80% similarity following dendrogram analysis. Acid shock had no effect on the subsequent PFGE patterns. A total of 8.7% (6/69) of cattle were shedding E. coli O157:H7 in the first month with faecal shedding increasing to 52% (36/69) by the third month of the study. A single isolate of E. coli O157:H7 may be passed rapidly through cattle pens, with the environment acting as a significant reservoir for transmission. PFGE is a useful tool for tracking the direct and indirect transmission of E. coli O157:H7 isolates on the farm.  相似文献
