Little is known about the effectiveness of the cleaning and disinfection methods used on commercial pig farms either in Ireland or worldwide. A National Salmonella Control Programme was implemented in Ireland in August 2002 to monitor and control the infection of pigs with Salmonella species. Commercial pig herds must be categorised according to their Salmonella status as either category 1, 2 or 3, having a serological prevalence of infection with Salmonella serotypes up to 10 per cent, between 10 and 50 per cent or more than 50 per cent, respectively. The aim of this study was to assess the efficacy of washing and disinfecting finisher units on category 1 and category 3 farms in reducing or eliminating the levels of contamination. Counts of Enterobacteriaceae were used as indicators of the contamination of the environment with enteric bacteria, which could include Salmonella species. Samples were taken from the pen floors, feeders and drinkers of seven category 1 and seven category 3 farms, and Enterobacteriaceae and salmonellae were enumerated in each sample. The results suggested that intensive cleaning and disinfection was effective at reducing levels of Enterobacteriaceae on the pen floors of both categories, but that residual contamination remained on the surfaces of the feeders and drinkers on all the farms, particularly on the category 3 farms.  相似文献

The presence of apoptotic features in spermatozoa has been related to lower quality and functional impairment. Members of the poly‐ADP‐ribose polymerases (PARP) familyare involved in both DNA repair and apoptosis, playing important roles in spermatogenesis. Poly‐ADP‐ribose polymerase can be cleaved by caspases, and the presence of its cleavage product (cPARP) in spermatozoa has been related to chromatin remodelling during spermatogenesis and to the activation of apoptotic pathways. There are no reports on immunodetection of cPARP in ram spermatozoa; thus, we have tested a commercially available antibody for this purpose. cPARP was microscopically detected in the acrosomal ridge of some spermatozoa (indirect immunofluorescence). A preliminary study was carried out by flow cytometry (direct immunofluorescence, FITC). Ram semen was extended in TALP and incubated for 4 h with apoptosis inducers staurosporine (10 μm ) or betulinic acid (200 μm ). Both inducers and incubation caused a significant increase in cPARP spermatozoa (0 h, control: 21.4±3.3%, inducers: 44.3±1.4%; 4 h, control: 44.3±2.4%, inducers: 53.3±1.4%). In a second experiment, we compared the sperm fractions after density gradient separation (pellet and interface). The pellet yielded a slightly lower proportion of cPARP spermatozoa (28.5±1.2% vs 36.2±2.0% in the interface; p < 0.001), and a 12‐h incubation increased cPARP similarly in both fractions (p < 0.001). cPARP seems to be an early marker of apoptosis in ram semen, although its presence in untreated samples was weakly related to worse quality (pellet/interface). We suggest to study the relationship of PARP and cPARP levels with between‐male differences on sperm fertility.  相似文献

We tested the hypothesis that compensatory fertilization of Scots pine (Pinus sylvestris L.) stands polluted by theHarjavalta copper-nickel smelter in SW Finland alleviates thestress imposed by metal-contaminated soils. The treatments inthe experiment consisted of control, liming, slow-release mineral mixture and stand-specific fertilization, which comprised at least methylene urea and ammonium nitrate. Stresswas assessed by needle fluctuating asymmetry (FA) which increases with an increase in developmental instability. Sevenyears after the treatments, we found pronounced variation inboth FA and needle length among study sites (0.5, 2, 4 and 8 km from the smelter) and among treatments. FA increased and needlelength decreased with proximity to the pollution source. Variation in FA was independent of tree size (used as index ofgrowth rate in even-age stand); the effects of the treatmentson needle length, on the other hand, were expressed only inmedium-sized and large trees. Liming best alleviated pollutionstress, while the application of a slow-release mineral mixtureincreased FA by 30%. In pollution-stressed stands FA correlatedpositively with constitutive resin flow but was independent of volume growth. Our findings suggest that physiological stress (indicated by increase in developmental instability) and tree growth or stand productivity are not directly linked to one another, and that some increase in productivity can be achievedwithout the alleviation of pollution-induced stress.  相似文献

Two horses were diagnosed as having Mycobacterium ulcerans infections. The first was a 21-year-old Quarterhorse-cross mare living in Mallacoota (a coastal town near the border of New South Wales and Victoria, Australia) that presented with lichenification, hair-loss and oedema on a fetlock, which subsequently ulcerated, as well as a non-healing ulcer on the wither. The second horse was a 32 year-old Standardbred gelding from Nicholson, near Bairnsdale, Victoria, that had an ulcerated lesion on its caudal thigh. Histologically, there were characteristic changes seen with M. ulcerans infections in other species, including extensive necrosis without associated granulomatous inflammation. The organisms were seen in Ziehl-Neelsen-stained smears or sections of the lesions from both horses and were isolated in culture from the first horse. A definitive diagnosis was provided by real-time polymerase chain reaction targeting the M. ulcerans-specific insertion sequence, IS2404. Delayed identification of the infectious agent in the first case led to the use of suboptimal antimicrobial therapy, resulting in failure to control the infection and the horse was subsequently euthanased. The second horse was successfully treated following surgical debulking of the centre of the lesion and one session of aggressive cryosurgery. Mycobacterium ulcerans should be considered in the differential diagnosis of unexplained lichenification with oedematous and ulcerated skin lesions in horses living in regions where this organism is endemic.  相似文献

Wheat is used in shrimp feed as protein and energy sources, as well as a nutritive binder that promotes water stability. However, wheat may be contaminated with deoxynivalenol (DON or vomitoxin) when wet weather conditions prevail during the growing season. Deoxynivalenol‐contaminated wheat was added to shrimp diet to obtain DON levels of 0.2, 0.5, and 1.0 ppm. Pelleted diets were fed 3 times daily until satiation to Pacific white shrimp Liropenaeus vanna‐mei. Biological performance (live weight, weekly growth increment, feed conversion ratio, and survival) was determined biweekly for 16 wk. Histological characteristics of shrimp were determined on samples obtained after 8 and 16 wk of growth. The DON levels in ground wheat, feed mash, pellets, and freeze‐dried shrimp were determined using thin layer and high performance liquid chromatography. Deoxynivalenol levels of 0.2, 0.5, and 1.0 ppm in the diet significantly reduced shrimp body weight and/or growth rate. However, the effects of 0.2 and 0.5 ppm DON were manifested at later stages of shrimp growth, and 0.2 ppm DON significantly affected only growth rate and not body weight (P > 0.05). Feed conversion ratio and survival of shrimp fed diets containing 0.2, 0.5, and 1.0 ppm DON were not significantly different from those of shrimp fed the control diet (0.0 ppm DON). After 16 wk of growth, no DON was detected in freeze‐dried shrimp, and no consistent difference was observed in the histological organ profiles of shrimp fed diets containing various levels of DON. Because low levels of DON in the diet can significantly reduce body weight and growth rate of shrimp, feed ingredients should be monitored for DON.  相似文献

Since the arrival of humans more than half of the Hawaiian archipelago’s known endemic bird taxa have been lost. Of the taxa that remain, 31 are federally listed under the US Endangered Species Act and 17 have populations of less than 1000 individuals. Previous studies have documented a geographic disparity in recovery expenditures on listed species, but none have specifically focused on Hawaiian birds. To draw attention to this disparity with the aim to improve Hawaiian bird conservation, I summarized recovery expenditures on listed birds from 1996 to 2004 comparing mainland and Hawaiian taxa in the context of their degree of endangerment. Federal and state spending on the 95 listed bird taxa over this nine year period totaled $752,779,924. Hawaiian birds comprise a third of the listed bird taxa (n = 31), yet dedicated recovery expenditures was only $30,592,692 or 4.1% of the total spent on all listed birds. Despite similar priority ranks assigned by the US Fish and Wildlife Service, listed mainland birds received over 15 times the funding of Hawaiian birds. In general, the threats to island taxa are unlike those of mainland taxa (e.g., non-native predators), management actions are expensive, and in many cases they must be conducted in perpetuity. Because of the status of many Hawaiian birds and the threats facing them, current recovery expenditures are inadequate to prevent additional extinctions.  相似文献

A test performance study (TPS) was organized in 2018 with ten official testing laboratories to evaluate the performance of different real-time PCR tests for the detection of Clavibacter sepedonicus and/or Ralstonia solanacearum in potato tubers. Participants were sent spiked potato extracts with low (0.8–1.2 × 104 cfu mL-1), medium (1.6–2.4 × 105 cfu mL-1) and high (1.6–2.4 × 107 cfu mL-1) bacterial loads, DNA extracts thereof and heel-end cores from symptomatic potato tubers. The four real-time PCR tests in this TPS for detection of C. sepedonicus were considered fit for purpose as principal screening methods. Two real-time PCRs in this TPS were considered fit for purpose as principal screening methods for detection of R. solanacearum. A third real-time PCR missed 23% of the DNA samples from low-level R. solanacearum spikes and is considered not fit for purpose as a principal screening method. Correct identification of spiked samples was lower when DNA extraction from the spiked samples was performed by the participating laboratories, highlighting the importance of appropriate DNA extraction protocols.  相似文献

The objective of this study was to obtain beta-lactoglobulin (beta-lg) gels at very low protein concentrations using a new multistep Ca(2+)-induced cold gelation process. In the conventional cold gelation process, salt free beta-lg solutions were heated at neutral pH, cooled, and cross-linked by adding salts. In our new process, first, long linear beta-lg fibrils were formed at pH 2. Solutions of these fibrils were cooled, and subsequently, the pH was adjusted to 7 or 8. Transmission electron microscopy studies showed that the long linear fibrils formed at pH 2 were stable when the pH was adjusted to 7 or 8. In the final step, the fibrils were cross-linked using CaCl(2). Using rheological measurements, the critical percolation concentration was determined. In the new multistep cold gelation process, the critical percolation concentration was an order of magnitude lower than in the conventional cold gelation method.  相似文献

Integrated pest management (IPM) and sustainable agriculture benefit from selective and environmentally benign crop-protection products. The performance of these products, however, is threatened by resistance. If it is to be prevented from standing in the path of progress, resistance must be managed. As agricultural practices move towards IPM and sustainable crop protection, selection pressure will be concentrated on products which are compatible with these systems. Producers, researchers, government and extension workers cannot solve these complex problems alone. The key to success is collaboration. The Insecticide Resistance Action Committee (IRAC) was set up by the crop-protection industry in 1984 to promote and co-ordinate its contribution to resistance management. Today, it is collaborating with research institutes, government organisations and extension services to combat resistance. IRAC focuses on research and communication projects which reduce selection pressure, organising workshops in ‘resistance hot spots’ and providing input to draft legislation.
