Enzyme changes in neurons and glia during barbiturate sleep   总被引:1,自引:0,他引:1  
During barbiturate sleep of rabbits, the succinoxidase activity in isolated neurons and glia from the caudal part of the reticular formation was lower than that during physiological sleep. No rhythmical, inverse enzyme changes were detected in barbiturate sleep in the neuron-glia unit, such as were found in physiological sleep.  相似文献

Some non-starch polysaccharides (NSP) promote the intestinal abundance of pathogenic Escherichia coli in pigs. However, their impact on pathogenic clostridia in the porcine intestinal tract is unknown. The present study aimed to determine the abundance of pathogens belonging to Clostridium clusters I and XI in faeces of growing pigs fed four semi-purified diets containing 5% of purified NSP differing in viscosity and fermentability (low-fermentable low-viscous cellulose; low-fermentable high-viscous carboxymethylcellulose; high-fermentable low-viscous oat β-glucan; and high-fermentable high-viscous oat β-glucan). For quantification of clostridial pathogens, toxin genes were determined using quantitative PCR. Clostridium perfringens α-toxin was detected in faeces of pigs fed low-fermentable cellulose and carboxymethylcellulose diets but was generally below detection limit for the two high-fermentable oat β-glucans. Clostridium botulinum toxin C2 was higher (P<0.05) by 0.7 to 0.9 log units in faeces of pigs fed the low-fermentable low-viscous cellulose diet than of pigs fed the other three diets. Clostridium difficile toxin B and Clostridium sordellii phospholipase C were not detected in pig faeces indicating that pigs were not colonized by these clostridial species belonging to Clostridium cluster XI. In conclusion, high-fermentable β-glucans decreased the faecal abundance of C. perfringens and, together with low-fermentable high-viscous CMC, that of C. botulinum in growing pigs when compared to low-fermentable low-viscous CEL. Besides fermentability, structural characteristics and indirect effects of NSP on nutrient flow into the large intestine may have played a role for the abundance of pathogenic clostridial species.  相似文献

Field experiments in Europe have shown that Chinese cultivars of winter oilseed rape ( Brassica napus ) are very susceptible to the pathogen Leptosphaeria maculans (cause of phoma stem canker). Climatic and agronomic conditions in China are suitable for L. maculans since the closely related but less damaging pathogen L. biglobosa occurs on the winter and spring oilseed rape crops there. Major gene resistance to L. maculans is not durable; when introduced into commercial oilseed rape cultivars it is rapidly rendered ineffective by changes in the pathogen population. The threat to Chinese oilseed rape production from L. maculans is illustrated by the way in which L. maculans has spread into other areas of the world where previously only L. biglobosa was present, such as Canada and Poland. Models were developed to describe the spread (in space and time) of L. maculans across Alberta province, Canada, based on survey data collected over a 15-year period. These models were used to estimate the potential spread of L. maculans across the Yangtze river oilseed rape growing areas of China and its associated costs. Short-term strategies to prevent occurrence of severe phoma stem canker epidemics in China include training of extension workers to recognise symptoms of the disease and use of PCR-based diagnostics to detect the pathogen on imported seed. Long-term strategies include the introduction of durable resistance to L. maculans into Chinese oilseed rape cultivars as a component of an integrated disease management programme. The costs of such strategies in relation to costs of a phoma stem canker epidemic are discussed.  相似文献

Effects of clear-cut harvesting on ground vegetation plant species diversity and their cover are investigated at two Norway spruce sites in southern Norway, differing in climate and topography. Experimental plots at these two sites were either harvested conventionally (stem-only harvesting) or whole trees including crowns, twigs and branches were removed (whole-tree harvesting), leaving residue piles on the ground for some months. We compare the number of plant species in different groups and their cover sums before and after harvesting, and between the different treatments, using non-parametric statistical tests. An overall loss of ground vegetation biodiversity is induced by harvesting and there is a shift in cover of dominant species, with negative effects for bryophytes and dwarf shrubs and an increase of graminoid cover. Differences between the two harvesting methods at both sites were mainly due to the residue piles assembled during whole-tree harvesting and the physical damage made during the harvesting of residues in these piles. The presence of the residue piles had a clear negative impact on both species numbers and cover. Pile residue harvesting on unfrozen and snow-free soil caused more damage to the forest floor in the steep terrain at the western site compared to the eastern site.  相似文献

Classical swine fever (CSF) is a severe multi-systemic disease that can affect both domestic pigs and wild boar. Past outbreaks in European wild boar involved high-virulent CSF virus (CSFV) strains and were mostly self-limiting. In these cases, morbidity and mortality rates were high in the affected regions. In contrast, endemic infections have been observed in several European wild boar populations in recent decades. Morbidity and mortality rates were much lower despite the fact that outbreaks were still detected via diseased or fallen animals. The virus strains involved were mostly classified as genotype 2.3 strains of moderate virulence causing age-dependent disease outcomes. The mechanisms leading to the establishment and perpetuation of endemicity are still not fully understood, but the factor "moderate virulence" seems to be of considerable importance. In this study, we aim to clarify whether the perception of declined 'CSF severity' could hypothetically reflect the adaptation of an initially high-virulent virus or whether this might be better explained as a misinterpretation of observations. A mechanistic eco-epidemiological model was employed to follow up a highly virulent strain of CSFV introduced into large connected wild boar populations. In the model, the virulence of the CSF virus is represented by case mortality and life expectancy after lethal infection. Allowing for small stochastic variation, these two characteristics of the virus are passed on with every new simulated infection that occurs. Model analysis revealed a decrease from high to moderate case mortality within a few years of simulated perpetuation of the virus. The resulting mortality corresponded to the level where the population average of the infectious period and the basic reproduction number of the disease were maximal. This shift in virulence was sufficient to prolong virus circulation considerably beyond the epidemic phase of the simulated outbreaks. Alternative mechanistic explanations for the decrease in disease severity in a CSF-affected wild boar population were evaluated in the light of the simulation experiments and the available epidemiological or virological evidence. In conclusion, the current virus isolates of subgroup 2.3 might be the ideally adapted variants of the CSF virus for long-term perpetuation in wildlife and indeed may have evolved (once) during past outbreaks in large populations. A repeated perception of a declining severity of disease pattern during the course of a CSF outbreak, however, favours the explanation based on monitoring and detection biases rather than repeated observation of selection against highly virulent virus during the time of virus perpetuation.  相似文献

Flavor of beef has been recognized as an equal quality trait to tenderness and lately high correlations between beef flavor and overall liking of beef have been demonstrated. In this review, we evaluate the effect of breed, sex, age, feeding, growth rate, slaughter method and aging on beef flavor. A common determinator for these factors is their influence on intramuscular fat (IMF) where more IMF increase the overall flavor liking. Feeding can, however, affect the flavor of beef beyond that of the amount of IMF. In several studies, pasture or pasture-based silage lead to a beef flavor characterized as fishy, gamey and grassy in contrast to concentrate-fed beef cattle. A geographical difference in flavor preferences of high-quality beef exists, most clearly illustrated among US consumers who prefer beef from concentrate-fed cattle, whereas European consumers tend to prefer beef from cattle fed a mixed diet on pasture and concentrate.  相似文献

This study was carried out to evaluate the usefulness of a pre‐maturation step in improving the coordination between cytoplasmic and nuclear maturation of horse compact cumulus oocytes by the addition of roscovitine (ROSC). Oocytes were collected by scraping and pre‐cultured for 18 h in a maturation medium TCM199 supplemented with pyruvate, LH, FSH, insulin growth factor (IGF), epidermal growth factor (EGF), insulin, transferrin and selenium (IVM‐ROSC) or in a simple medium (M199‐ROSC). After pre‐maturation, oocytes from both the groups were in part denuded and fixed‐stained and in part in vitro matured to assess the kinetic of in vitro maturation (IVM). The nuclear progression and the cytoskeletal organization of microfilaments and cortical granules (CG) of treated and untreated oocytes were assessed by fluorescent probes. Oocytes immediately fixed after recovery and oocytes pre‐cultured in M199‐ROSC for 18 h did not show metaphase II (MII) plates, whereas in IVM‐ROSC group, 6/69 oocytes (8.7%) showed MII plates. After inhibition, during maturation kinetics at 11, 18 and 29 h, maturation rate of M199‐ROSC group progressively increased and at 29 h of IVM, reached the maturation rate of control group (13/66, 19.7% vs 31/125, 24.8%). No statistically significant differences in cytoplasmic maturation were found. The number of MII plates after 29 h of IVM, was significantly higher (p < 0.05) in IVM‐ROSC group (34/90) compared with M199‐ROSC (13/66) and control groups (31/125) as well as the number of oocytes with microfilaments and CG distributed in cortical region (25/34 vs 3/13 and 7/31 respectively). Our results showed that pre‐culturing in the presence of Roscovitine in a fully supplemented maturation medium containing gonadotropins and growth factors partially suppressed the meiotic maturation, but established a more suitable environment for improving cytoplasmic maturation of horse compact cumulus oocytes as defined by microfilaments and CG configuration.  相似文献
