Protein distribution in endosperm of maize grains differing by their texture, flint or dent, and by their genotype, wild or waxy or amylose-extender, was examined by the successive use of 0.5 M NaCl, 0.5 M NaCl plus 0.6% 2-mercaptoethanol (2ME) at neutral and then alkaline pH, and 55% 2-propanol plus 0.6% 2ME as extractants. Proteins extracted in the presence of 2ME were characterized by their size polymorphism and amino acid composition. Proteins isolated with NaCl plus 2ME at neutral pH corresponded with a mixture of gamma-zein (27 kDa) and glutelin-like proteins. Proteins isolated with NaCl plus 2ME at pH 10 were a mixture of gamma-zeins (27 and 16 kDa) and beta-zeins (14 kDa). Alcohol-soluble proteins consisted of alpha-, beta-, and delta-zeins, alpha subunits being predominant. Zein quantitation was improved by weighing the nitrogen percentage of extracts by their zein content, as estimated from the data on amino acid composition. The data reported by Wolf et al. (Cereal Chem. 1975, 52, 765) were integrated to the results of this work to suggest the occurrence of an inverse correlation between amylose in starch and zeins in proteins. 相似文献

Withdrawal periods required when doses of 24,000 IU and 66,000 IU of procaine penicillin G/kg body weight were administered to yearling beef steers by intramuscular injection daily for five consecutive days were investigated. These dosages are in excess of product label recommendations, but are in the range of procaine penicillin G dosages that have been administered for the treatment of some feedlot bacterial diseases. The approved dose in Canada is 7,500 IU/kg body weight intramuscularly, once daily, with a withdrawal period of five days. Based on the tissue residue data from this study, the appropriate withdrawal period is ten days for the 24,000 IU/kg body weight dose and 21 days for the 66,000 IU/kg body weight dose when administered intramuscularly to yearling beef steers. In a related study, 18 yearling beef steers received 66,000 IU of procaine penicillin G/kg body weight administered by subcutaneous injection, an extra-label treatment in terms of both dose and route of administration, typical of current practice in some circumstances. Deposits of the drug were visible at subcutaneous injection sites up to ten days after injection, with more inflammation and hemorrhage observed than for intramuscular injections of the same dose. These results suggest that procaine penicillin G should not be administered subcutaneously at high doses; and therefore a withdrawal period was not established for subcutaneous injection. 相似文献

Eight healthy, non-pregnant, crossbred Holstein dairy cows (557-682 kg) within their first 3 months of lactation (13-21.5 kg of milk/day) were used. Cows were kept in tie stalls for the whole experiment. The 8 cows were randomly assigned to 2 (IM and SC) 4 x 4 balanced Latin square design experiments. Doses of procaine penicillin G (PPG) (300000 IU/mL) in each square were 7000, 14000, 21000 and 28000 IU/kg and were injected IM or SC once daily for 5 consecutive days. Volumes of PPG per site of injection never exceeded 20 mL. Blood was collected to determine the Cmax, Tmax, and AUC; urine and milk were also taken to measure the persistence of PPG in these fluids. Results show that serum Cmax and Tmax were only slightly affected by increasing the doses or the route of administration, whereas the AUC was linearly increased in relation to the dose injected in both modes of injection. In the urine, Cmax varied from 160 to 388 IU/mL and Tmax from 72-120 h during 5 consecutive days of PPG injection. A dose effect in Cmax was observed only for the IM route of administration and no variation (P > 0.05) was found between the IM and SC routes. Milk Cmax concentrations were only increased by the dose regimen in the IM group. At doses of 21000 and 28000 IU/kg, the IM group had a higher (P > 0.05) Cmax when compared with the SC groups. Milk PPG residues were not detectable over 96 h following the last IM injection, independently of the dose injected. However milk PPG residues were detected for up to 132 h following the last SC injection. These results show that when PPG is injected IM once daily in volumes not exceeding 20 mL/site at doses as high as 28000 IU/kg, the withdrawal period should be at least 96 h. Therefore, in the present model, there was no advantage to inject PPG by SC route to improve PPG kinetic parameters as the AUC, Cmax, or Tmax. 相似文献

The aim of this study was to evaluate the effects of different treatments for induction and synchronization of oestrus and ovulation in seasonally anovulatory mares. Fifteen mares formed the control group (C), while 26 mares were randomly assigned to three treatment groups. Group T1 (n = 11) were treated with oral altrenogest (0.044 mg/kg; Regumate®) during 11 days. Group T2 (n = 7) was intravaginally treated with 1.38 g of progesterone (CIDR®) for 11 days. In group T3 (n = 8), mares were also treated with CIDR®, but only for 8 days. All mares received PGF2α 1 day after finishing the treatment. Sonographic evaluation of follicles, pre‐ovulatory follicle size and ovulation time was recorded. Progesterone and leptin levels were analysed. Results show that pre‐ovulatory follicles were developed after the treatment in 88.5% of mares. However, the pre‐ovulatory follicle growth was dispersal, and sometimes it was detected when treatment was not finished. While in mares treated with intravaginal device, the follicle was soon detected (1.5 ± 1.2 days and 2.3 ± 2.0 days in T2 and T3 groups, respectively), in T1 group, the pre‐ovulatory follicle was detected slightly later (3.9 ± 1.6 days). The interval from the end of treatment to ovulation did not show significant differences between groups (T1 = 13.1 ± 2.5 days; T2 = 11.0 ± 3.6 days; T3 = 13.8 ± 4.3 days). The pregnancy rate was 47.4%, similar to the rate observed in group C (46.7%; p > 0.05). Initial leptin concentrations were significantly higher in mares, which restart their ovarian activity after treatments, suggesting a role in the reproduction mechanisms in mares. It could be concluded that the used treatments may be effective for oestrous induction in mares during the late phase of the seasonally anovulatory period. Furthermore, they cannot synchronize oestrus, and then, it is necessary to know the reproductive status of mares when these treatments are used for oestrous synchronization. 相似文献

Urban vegetation is shown to be unevenly distributed across cities and there is evidence of disparities in benefits provided by vegetation and of public health problems induced by urban heat islands. In order to improve vegetation cover, it remains crucial to understand the underpinning of such unevenness. In this paper, we investigate in Montreal (Canada) how the built environment, sociodemographic factors and administrative boroughs influence tree and lawn cover in public and residential land. The analysis was conducted at the dissemination area (DA) level, a Canadian census unit containing about 400–700 people. Six vegetation indicators were used as dependent variables: the proportion of a DA covered by trees/shrubs, lawn and total vegetation; the proportion of streets covered by trees/shrubs and the proportion of residential yards covered by trees/shrubs and total vegetation. Three sets of independent variables were studied: the built environment, sociodemographics and borough names. We used spatial autoregressive models to control for dependence and the spatial autoregressive term explained a large amount of variability in vegetation cover. The built-environment variables tend to have higher effects than the socio-demographic variables when predicting the three DA vegetation indicators, backyard vegetation, and to a lesser degree, street tree/shrub cover. In particular, population density is associated negatively to all indicators but positively to street tree cover. Socio-demographics are substantial in the explanation of the distribution of street trees, especially the presence of recent immigrants (negative effect) and of university degree holders (positive effect). These findings call for appropriate greening programs adapted to the local socio-demographic profile. The significance of boroughs also suggests the need for further research on the impact of within-city administrative hierarchies on the unevenness of urban vegetation. 相似文献

Winter-hardy faba bean (Vicia faba L.) from northern Europe is represented by a rather narrow gene pool. Limited selection gains for overwintering below ?25 °C have restricted the adoption of this crop. Therefore, the faba bean collection maintained by the USDA-ARS National Plant Germplasm System (NPGS) was utilized to broaden this genetic base by identifying potentially new sources of winter-hardiness using a modified mass selection scheme. From an initial source population, bulk-harvested micro-plots of 466 NPGS accessions, four different bulks were formed by harvesting seeds from plants that survived at four locations representing a range of overwintering selection environments across southeastern Washington. These four bulk populations were then mass selected for three cycles based on winter survival along with a selected group of advanced northern European populations or breeding lines at two southeastern Washington locations with divergent climates. This procedure generated breeding materials with winter-hardiness comparable to the northern European populations. We observed that Northern European populations showed slower annual gains in percent survival (<5%) than NPGS bulks (>5%). Further, the frequency of large seeded populations (>80 g × 100 seed?1) was reduced over time, suggesting an association between seed size and overwintering. The breeding materials generated by mass selection are useful for the future improvement of faba bean as a fall-sown pulse or cover crop. 相似文献
