ABSTRACT Even though Botrytis cinerea, the causal agent of gray mold, is a highly variable fungus with strains displaying very different degrees of virulence toward one given host plant species, no study has yet shown any correlation between the lack of aggressiveness of one given strain and its ability to stimulate a defense response from its host. Strains of B. cinerea collected from different host plant species were screened for their pathogenicity on grapevine to select two strains with similar morphological characteristics but different levels of virulence. In grapevine leaves, the less aggressive strain, T4, enhanced the accumulation of many defense products including secondary metabolites and the pathogenesis-related proteins, chitinase and beta-1,3-glucanase. Interestingly, secondary metabolites were formed in cells around a small group of dead cells. When compared with T4, the more aggressive strain, T8, had larger necrotic spots, no secondary metabolite biosynthesis, and accumulations of chitinases and beta-1,3-glucanases that were more delayed, yet only slightly weaker. The culture fluids of both strains mimicked the differential effect of each isolate in stimulating chitinase activity when infiltrated into grapevine leaves.  相似文献

Canine hemangiosarcoma (HSA) is an aggressive cancer of endothelial cells with short survival times. Understanding the genomic landscape of HSA may aid in developing therapeutic strategies for dogs and may also inform therapies for the rare and aggressive human cancer angiosarcoma. The objectives of this study were to build a framework for leveraging real-world genomic and clinical data that could provide the foundation for precision medicine in veterinary oncology, and to determine the relationships between genomic and clinical features in canine splenic HSA. One hundred and nine dogs with primary splenic HSA treated by splenectomy that had tumour sequencing via the FidoCure® Precision Medicine Platform targeted sequencing panel were enrolled. Patient signalment, weight, metastasis at diagnosis and overall survival time were retrospectively evaluated. The incidence of genomic alterations in individual genes and their relationship to patient variables including outcome were assessed. Somatic mutations in TP53 (n = 44), NRAS (n = 20) and PIK3CA (n = 19) were most common. Survival was associated with presence of metastases at diagnosis and germline variants in SETD2 and NOTCH1. Age at diagnosis was associated with somatic NRAS mutations and breed. TP53 and PIK3CA somatic mutations were found in larger dogs, while germline SETD2 variants were found in smaller dogs. We identified both somatic mutations and germline variants associated with clinical variables including age, breed and overall survival. These genetic changes may be useful prognostic factors and provide insight into the genomic landscape of hemangiosarcoma.  相似文献

Monitoring of foliage nutrients in forest plantations is used as a method to identify nutritional problems and recommend ameliorations. A requirement is the development of nutrient bench marks and these are usually a single concentration based on the level at which symptoms appear. The critical level is defined as 90% of optimum growth below which it is expected that growth responses to applied nutrients would be obtained. The phosphorus critical level at different ages in radiata pine has been developed showing a decline with age from 0.18% P at 2 years to 0.12% after 15 years. A typical standard level applied is 0.14% P which would underestimate young stands and overestimate older stands. Routine assessments in plantations show patterns of decline in P over time, on some soils they generally remain above the critical levels, others below, and some move into deficiency with time.  相似文献

Mapping weed densities within crops has conventionally been achieved either by detailed ecological monitoring or by field walking, both of which are time‐consuming and expensive. Recent advances have resulted in increased interest in using Unmanned Aerial Systems (UAS ) to map fields, aiming to reduce labour costs and increase the spatial extent of coverage. However, adoption of this technology ideally requires that mapping can be undertaken automatically and without the need for extensive ground‐truthing. This approach has not been validated at large scale using UAS ‐derived imagery in combination with extensive ground‐truth data. We tested the capability of UAS for mapping a grass weed, Alopecurus myosuroides , in wheat crops. We addressed two questions: (i) can imagery accurately measure densities of weeds within fields and (ii) can aerial imagery of a field be used to estimate the densities of weeds based on statistical models developed in other locations? We recorded aerial imagery from 26 fields using a UAS . Images were generated using both RGB and Rmod (Rmod 670–750 nm) spectral bands. Ground‐truth data on weed densities were collected simultaneously with the aerial imagery. We combined these data to produce statistical models that (i) correlated ground‐truth weed densities with image intensity and (ii) forecast weed densities in other fields. We show that weed densities correlated with image intensity, particularly Rmod image data. However, results were mixed in terms of out of sample prediction from field‐to‐field. We highlight the difficulties with transferring models and we discuss the challenges for automated weed mapping using UAS technology.  相似文献

The pharmacokinetics of ketoprofen were determined after an intravenous (i.v.) and intramuscular (i.m.) dose of 2.0 mg/kg body weight in five camels (Camelus dromedarius) using gas chromatography/mass spectrometry (GC/MS). The data obtained (median and range) following i.v. administration was as follows: the elimination half-life (t(1/2beta)) was 4.16 (2.65-4.29) h, the steady state volume of distribution (Vss) was 130.2 (103.4-165.3) mL/kg, volume of distribution (area method) (Vd(area)) was 321.5 (211.4-371.0) mL/kg, total body clearance (Cl) was 1.00 (0.88-1.08) mL/min x kg and renal clearance was 0.01 (0.003-0.033) mL/min x kg. Following i.m. administration, the drug was rapidly absorbed with peak serum concentration of 12.2 (4.80-14.4) microg/mL at 1.50 (1.00-2.00) h. The systemic availability of ketoprofen was complete. The apparent half-life was 3.28 (2.56-4.14) h. A hydroxylated metabolite of ketoprofen was identified by (GC/MS) under electron impact (EI) and chemical ionization (CI) scan modes. The detection times for ketoprofen and hydroxy ketoprofen in urine after an intravenous (i.v.) dose of 3.0 mg/kg body weight was 24.00 and 70.00 h, respectively. Serum protein binding of ketoprofen at 20 microg/mL was extensive; (99.1+/-0.15%).  相似文献

A multitracer study of a small aquifer in northern Switzerland indicates that the atmosphere in central Europe cooled by at least 5 degreesC during the last glacial period. The relation between oxygen isotope ratios (delta18O) and recharge temperatures reconstructed for this period is similar to the present-day one if a shift in the delta18O value of the oceans during the ice age is taken into account. This similarity suggests that the present-day delta18O-temperature relation can be used to reconstruct paleoclimate conditions in northern Switzerland. A gap in calculated groundwater age between about 17,000 and 25,000 years before the present indicates that during the last glacial maximum, local groundwater recharge was prevented by overlying glaciers.  相似文献

BACKGROUND: Tracking and census baiting are two techniques that are commonly advocated for monitoring the size of mouse populations. However, currently these techniques are only able to provide an index of population size, rather than an assessment of absolute numbers. In this study the authors tested the reliability of both tracking (footprints left on tiles of fixed size) and census baiting as indices of population size, and sought to calibrate levels of activity and bait consumption under both semi‐natural and field conditions (inside farm buildings). RESULTS: Under semi‐natural conditions, census baiting produced more satisfactory population estimates than those derived from tracking activity. An initial field trial established that the optimum bait point density for this technique was 1 point per 2 m. Subsequent field trials demonstrated that the bait census technique offers a way to estimate the approximate size of stable populations of mice (population size = (mean daily bait consumption ‐ 36.3)/2.46). CONCLUSION: The results to date are sufficiently encouraging to support the use of this cost‐effective approach to monitoring mouse numbers in the type of habitats investigated in this study. © Crown copyright 2008. Cocoa butter is the major commercial product from seeds of Theobroma cacao (cacao). However, most cacao breeding programs have only considered as selection criteria total seed yield per plant and disease resistance. Progress in cocoa butter yield might be possible if variability for fat content exists and its associations with the other production components are established. The average fat content in dry unfermented seeds for 490 accessions of the Centro de Pesquisa do Cacau germplasm collection in Ilhéus, Bahia, Brazil was 53.2%, ranging from 45.4% in CC 57 to 60.3% in NA 312. Fat content averaged over series indicated that genotypes originally collected in the Upper Amazonian region had higher fat levels, while the Trinitario-Criollo and Bahian genotypes tended to have lower fat content. When considering total fat yield plant-1 an opposite trend arose with the series with low fat content having the highest total fat yield. The correlation between fat content and fat yield plant-1 was not significant, while there was a significant negative correlation between dry seed yield plant-1 and fat content. Despite the significant negative association between fat content and dry seed yield plant-1, the correlation coefficient was rather low, and genotypes could be identified with both high fat content and dry seed yield. A significant pollen effect on fat content was observed based on a complete diallel crossing scheme between three genotypes with high and three with low levels. The average fat content values in progenies were similar to the average of the parents, and the segregation range transgressed the distribution of the parents.
