The 90 chickens were randomly divided into control group, E.necatrix one time infection group and E.necatrix two times infection group.We used duplex PCR method to test the expression changes of HSP-90 mRNA in intestinal tissue of chickens infected with E.necatrix and researched the effects of it comprehensively and systematically.The results were showed that after 0 to 14 days the expression of HSP-90 mRNA in intestinal tissues of 14-day-old chickens infected with E.necatrix presented a upward trend though it was lower than the control chickens.These chickens were infected with E.necatrix again at 28 days, HSP-90 mRNA expressions of duodenum, jejunum and ileum decreased and then increased.However, it always showed a downward trend in appendix.When 28 days chickens were infected with high-does E.necatrix once, HSP-90 mRNA expressions of duodenum, jejunum and appendix were in a declining curve.While it was increased originally and then decreased in ileum.This study indicated that HSP-90 mRNA expressions in intestinal tissue of chickens infected with E.necatrix were inhibited at first, and the expression quantity was closely related to the degrees of intestinal tissue damage.  相似文献

近年来,为了提高母猪的繁殖效率,满足养猪业养殖高度集约化的生产要求,行业内开始大力推广批次化生产繁殖技术。目前国内都是采用提取类生殖激素孕马血清(PMSG)和孕妇尿液中的人绒毛膜促性腺激素(HCG),以及化学激素烯丙孕素进行繁殖调控。但是这些提取类产品仍存在一些缺陷,因此开发出活性好、纯度高、成本低的高品质蛋白类繁殖激素产品,进而应用于猪场批次化生产中,是提高我国乃至全球畜牧业发展效率的关键所在。  相似文献

OBJECTIVE: To compare the microstructural damage created in bone by pins with lathe-cut and rolled-on threads, and to determine the peak tip temperature and damage created by positive-profile external fixator pins with either hollow ground (HG) or trocar (T) tips during insertion. STUDY DESIGN: An acute, in vitro biomechanical evaluation. SAMPLE POPULATION: Twenty-seven canine tibiae. METHODS: Lathe-cut thread design with T point (LT-T), rolled-on thread design with T point (RT-T), and rolled-on thread design with HG point (RT-HG) pins were evaluated. Twenty pins of each type were inserted under constant drilling pressure into 12 canine tibiae (12 diaphyseal and 8 metaphyseal sites per pin type). Peak pin tip temperature, drilling energy, end-insertional pin torque, and pullout force were measured for each pin. For the histologic study, five pins of each type were inserted into cortical and cancellous sites in 15 additional tibiae. Entry and exit damage, and thread quality were assessed from 100 micron histologic sections by using computer-interfaced videomicroscopy. RESULTS: T-tipped pins reached higher tip temperature in both diaphyseal and metaphyseal bone compared with HG-tipped pins. RT-T pins had higher pullout strength (diaphyseal) and end-insertional torque compared with other combinations. No differences in drilling energy or insertional bone damage was found between the three pin types (P < .05). CONCLUSIONS: T-tipped pins mechanically outperformed HG-tipped pins. Pin tip and thread design did not significantly influence the degree of insertional bone damage. CLINICAL RELEVANCE: T-tipped pins may provide the best compromise between thermal damage and interface friction for maximizing performance of threaded external fixator pins.  相似文献

家兔骨骼标本制作过程中,漂白和粘接是非常重要的两个环节,直接影响标本质量、美观。为了找到一种良好的家兔骨骼标本漂白和粘接工艺,将脱脂处理后的8只家兔骨骼随机分成4组,分别放入5%、15%、25%和30%的过氧化氢溶液进行梯度漂白,选取最佳效果。将漂白处理后的家兔骨骼,随机分成甲、乙、丙、丁4组,将其关节按不活动关节、半活动关节、活动关节作为试验对象,分别使用骨胶黏合剂、AB胶、502胶水和热熔胶进行粘接对比。结果表明:①不同部位的骨骼所需的过氧化氢浓度和作用时间均有所差异。头骨在30%过氧化氢中漂白1.5 h;颈椎、腰椎和胸骨在25%过氧化氢中漂白1.5 h;肋骨、肱骨和肩胛骨在15%过氧化氢中漂白1.5 h;掌骨、尾椎和指骨等部位骨骼在5%过氧化氢中漂白2 h效果最佳。②骨胶黏合剂起效快,粘接紧密,强度高,定型效果好。上述试验可减少标本制作过程中对家兔骨骼的损坏,最大限度地保持其原貌,使标本更加生动、逼真。  相似文献

为探讨短花针茅(Stipa breviflora)荒漠草原建群种空间异质性在不同载畜率下的变化特点和差异,本研究以内蒙古自治区乌兰察布盟四子王旗内蒙古农牧业科学院试验基地的短花针茅种群为研究对象,采用半方差函数、分形维数和克里格差值方法进行系统研究,结果如下:短花针茅种群空间异质性随载畜率的增大呈增大趋势。轻度放牧(LG)和重度放牧(HG)处理区短花针茅种群空间分布主要受结构性因素影响,但表现结果的影响过程存在差异;中度放牧(MG)处理区短花针茅种群空间分布除结构性因素占主导地位外,放牧家畜的随机性牧食行为也占较大比重。  相似文献

AIM To study the effect of microRNA-153-3p (miR-153-3p) knock-down on oxidative injury of H9C2 cells induced by H2O2 and its specific mechanism. METHODS The oxidative stress injury of H9C2 cell model was induced by H2O2, and then the cell viability and the expression of miR-153-3p were detected by MTT assay and RT-qPCR, respectively. The effects of miR-153-3p knock-down on the H9C2 cell injury under oxidative stress were studied by RNA interference technology. The targets of miR-153-3p were identified by Western blot and dual-luciferase reporter assay. RESULTS MTT assay showed that the viability of H9C2 cells was decreased with the increase in H2O2 concentration (P<0.05). The results of RT-qPCR showed that the expression of miR-153-3p was increased with the increase in H2O2 concentration (P<0.05). Knock-down of miR-153-3p increased the viability of H9C2 cells under oxidative stress, decreased the cell apoptosis and the content of malondialdehyde (MDA), and increased the activity of superoxide dismutase (SOD). The expression of nuclear factor E2-related factor 2(Nrf2) and antioxidant response element(ARE) activity were increased with the increase in H2O2 concentration (P<0.01). TargetScan analysis and dual-luciferase reporter assay showed that Nrf2 was one of the potential target genes of miR-153-3p. The results of Western blot further showed that over-expression of miR-153-3p inhibited the expression of Nrf2 (P<0.01), while down-regulation of miR-153-3p increased the expression of Nrf2 (P<0.01). Dual interference with Nrf2 and miR-153-3p significantly reduced H9C2 cell viability, promoted the apoptosis, increased MDA content, and decreased SOD activity in the presence of H2O2 (P<0.01). CONCLUSION Inhibition of miR-153-3p expression attenuates the injury of H9C2 cells induced by H2O2 through up-regulating Nrf2/ARE signaling pathway.  相似文献
