Disease in animals is a well-known inhibitor of growth and reproduction. Earlier studies were initiated to determine the effects of endotoxin on pituitary hormone secretion. These studies found that in sheep, growth hormone (GH) concentration was elevated, whereas insulin-like growth factor-I (IGF-I) was inhibited, as was luteinizing hormone (LH). Examination of the site of action of endotoxin in sheep determined that somatotropes expressed the endotoxin receptor (CD14) and that both endotoxin and interleukin-Iβ activated GH secretion directly from the pituitary. In the face of elevated GH, there is a reduction of IGF-I in all species examined. As GH cannot activate IGF-I release during disease, there appears to be a downregulation of GH signalling at the liver, perhaps related to altered nitration of Janus kinase (JAK). In contrast to GH downregulation, LH release is inhibited at the level of the hypothalamus. New insights have been gained in determining the mechanisms by which disease perturbs growth and reproduction, particularly with regard to nitration of critical control pathways, with this perhaps serving as a novel mechanism central to lipopolysaccharide suppression of all signalling pathways. This pathway-based analysis is critical to the developing novel strategies to reverse the detrimental effect of disease on animal production.  相似文献

Pharmacokinetics and pharmacodynamics of alfaxalone was performed in mallard ducks (Anas platyrhynchos) after single bolus injections of 10 mg/kg administered intramuscularly (IM; n = 10) or intravenously (IV; n = 10), in a randomized cross‐over design with a washout period between doses. Mean (±SD) Cmax following IM injection was 1.6 (±0.8) µg/ml with Tmax at 15.0 (±10.5) min. Area under the curve (AUC) was 84.66 and 104.58 min*mg/ml following IV and IM administration, respectively. Volume of distribution (VD) after IV dose was 3.0 L/kg. The mean plasma clearance after 10 mg/kg IV was 139.5 (±67.9) ml min?1 kg?1. Elimination half‐lives (mean [±SD]) were 15.0 and 16.1 (±3.0) min following IV and IM administration, respectively. Mean bioavailability at 10 mg/kg IM was 108.6%. None of the ducks achieved a sufficient anesthetic depth for invasive procedures, such as surgery, to be performed. Heart and respiratory rates measured after administration remained stable, but many ducks were hyperexcitable during recovery. Based on sedation levels and duration, alfaxalone administered at dosages of 10 mg/kg IV or IM in mallard ducks does not induce clinically acceptable anesthesia.  相似文献

Lettuce leaves co-inoculated withSclerotinia sclerotiorum andConiothyrium minitans and controls were placed on, or buried in, soil for a period of two weeks to study development and survival ofC. minitans. OnS. sclerotiorum-infected leaves on the soil surface, the number of colonies ofC. minitans recovered was about 40% of the number of pycnidiospores applied. When buried in the soil there was a reduction to about 2% of the spores applied. WhenC. minitans was applied on healthy lettuce leaves, which were subsequently placed on or in soil, the recovery was about 4%. It is argued that these figures indicate multiplication ofC. minitans onS. sclerotiorum-infected lettuce leaves on the soil, and good survival in all other cases.  相似文献

Susceptibilities to metabolites of tiamulin (TIA) and enrofloxacin (ENR) were tested using selected bacteria with previously defined minimal inhibitory concentrations (MIC). The TIA metabolites tested were: N-deethyl-tiamulin (DTIA), 2beta-hydroxy-tiamulin (2beta-HTIA) and 8alpha-hydroxy-tiamulin (8alpha-HTIA), and the ENR metabolites were: ciprofloxacin (CIP) and enrofloxacin N-oxide (ENR-N). Bacteria, all of porcine origin, were selected as representatives of bacterial infections (Staphylococcus hyicus and Actinobacillus pleuropneumoniae), zoonotic bacteria (Campylobacter coli) and indicator bacteria (Escherichia coli and enterococci). Furthermore the effects of these compounds were tested on the microbial community of active sludge to test any negative effect on colony forming units (CFU). DTIA had a potency of 12.5-50% of the potency of TIA. 2beta-HTIA and 8alpha-HTIA had potencies less than 1% of the potency of TIA. ENR-N had a potency of 0.75-1.5% of the potency of ENR, while CIP and ENR had similar potencies. Results obtained here indicate that CIP and DTIA could contribute to the selective pressure for upholding antimicrobial resistant bacteria in animals under ENR or TIA treatment. The most potent metabolites CIP and DTIA showed considerable potencies against activated sludge bacteria compared to the parent compounds. EC(50) (microg/ml) for ENR, CIP, TIA and DTIA were 0.018 [95% CI: 0.028-0.149], 0.064 [95% CI: 0.007-0.046], 6.0 [95% CI: 3.6-9.8], and 9.7 [95% CI: 5.8-16.3], respectively. This indicates that the compounds can change the bacterial population in the sludge, and hereby alter the properties of the sludge.  相似文献

In the light of an increasing demand for staple food, especially rice, in southeast China, investigations on the specific site potential expressed as the relationship between soil and crop yield parameters gain increasing importance. Soil texture and several soil chemical parameters as well as plant properties such as crop height, biomass and grain yield were investigated along two terraced catenas with contrasting soil textures cropped with wet rice. We were aiming at identifying correlative relationships between soil and crop properties. Data were analyzed both statistically and geostatistically on the basis of semivariograms. Statistical analysis indicated a significant influence of the relief position on the spatial distribution of soil texture, total carbon and total nitrogen contents. Significant correlations were found for the catena located in a sandstone area (Catena A) between rice yield and silt as well as total nitrogen content. Corresponding relationships were not detectable for paddy fields that developed from Quaternary clays (Catena B). As suggested by the nugget to sill ratio, spatial variability of soil texture, total carbon and nitrogen was mainly controlled by intrinsic factors, which might be attributed to the erosional transport of fine soil constituents, indicating the importance of the relief position and slope in soil development even in landscapes that are terraced. The crop parameters exhibited short ranges of influence and about one third of their variability was unexplained. Comparable ranges Of selected crop and soil parameters, found only for Catena A, are indicative of close spatial interactions between rice yield and soil features. Our findings show that especially in sandstone-dominated areas, a site-specific management can contribute to an environmentally safe rice production increase.  相似文献

The speciation of phosphorus (P) in native and degraded peat soils is an analytical challenge, and synchrotron‐based P K‐edge X‐ray absorption near‐edge structure (XANES) is a suitable method to gain information on P species in soils and organic materials. The objective of the present study was to test if P K‐edge XANES reflected differences in P fractions in fen peat due to sequential extraction and peat degradation. We investigated each one top‐ and subsoil sample of a Fibric Histosol, which differed in the degree of humification (H8 vs. H5) and concentration of total P (Pt) (1944 mg kg–1 vs. 436 mg kg–1). In the topsoil, residual P, H2SO4‐P, and NaOH‐P accounted for roughly the same proportions of Pt (≈30%). In the subsoil, residual P (64% of Pt) was more abundant than NaOH‐P (21% of Pt) and H2SO4‐P (10% of Pt). Among many different P reference standards, the P XANES spectra reflected differences in mineral P more distinctive than in organic P compounds. Phosphorus XANES spectra of the residues after each sequential extraction step all showed a prominent white‐line peak at around 2152 eV. Stepwise removal of resin‐P, NaHCO3‐P, and NaOH‐P were reflected mainly by the peak intensity but scarcely by distinct spectral features. Extraction with H2SO4 led to the disappearance of spectral features of Ca and Mg phosphates which is a first direct hint to these compounds in the peat. In conclusion, a combined sequential fractionation and spectroscopic (31P NMR, P K‐ and L‐edge XANES with linear‐combination fits) approach is proposed to overcome limitations of the present study and gain more insight into the P species in peat soils.  相似文献

The concentration of water soluble carbohydrates (WSC) is a main determinant of the ensiling potential of forages and the aerobic stability of silages. The aims of the present study were to assess the impact of genotype on the seasonal changes of WSC concentration, to adapt and calibrate the FONSCH (FOrage NonStructural CarboHydrates) model – originally developed for forage grasses – for silage maize, and finally to conduct a long-term simulation study for quantifying weather-related variability of WSC content. A 3-year field experiment (2001–2003) was conducted in northern Germany to evaluate differences in WSC content among commercial silage maize varieties and to provide calibration data for modelling. Eight varieties covering three maturity groups (early, mid-early and mid-late) were investigated. On six dates throughout the vegetation period the plants were harvested, separated into ear and stover, and freeze–dried for subsequent determination of WSC. Ear, stover, and whole crop WSC content was influenced by genotype and environmental conditions. Genotypic effects were most pronounced in the early grain filling period. At silage maturity, an effect of variety on whole crop WSC content was found in the mid-early group only, accounting for a difference of 44.8 g WSC kg−1 DM. FONSCH model calibration showed good agreement between observed and calculated WSC contents, with RMSE below 27.6 g WSC kg−1 DM and r2 above 0.84. Parameter estimates revealed a substantial influence of temperature and solar radiation on WSC content, while plant available soil water had minimal impact. Successful model validation with an independent data set allowed us to perform a 30-year simulation study, where variation among varieties and years of up to 68.8 and 186.5 g WSC kg−1 DM, respectively, demonstrated a larger impact of environmental conditions compared to genotype.  相似文献
