SUMMARY A multivalent footrot vaccine has been developed, containing pilus antigens produced in recombinant Pseudomonas aeruginosa and representing all nine serogroups of Dichelobacter (Bacteroides) nodosus commonly recognised in the field. The responses of sheep to the multivalent vaccine have been compared with those to monovalent vaccines representing only a single serogroup. Antigenie competition between serogroups occurred in sheep immunised with the multivalent formulation, but high levels of protection were still achieved. The study showed that in multivalent footrot vaccines, antigenic competition is predominantly due to the presence of a family of immunologically-related pilus antigens rather than to interference by extraneous proteins.  相似文献

Seven Meat Animal Research Center (MARC) III heifers (410+/-25 kg) fitted with hepatic portal, mesenteric venous, carotid catheters, and an abomasal cannula were used in a 7 x 5 incomplete Latin square design experiment. The objective was to evaluate the effects of increasing levels of ruminally degradable N (RDN) with or without the addition of abomasally infused casein on portal-drained visceral (PDV) flux of nutrients. Treatments consisted of dietary CP percentage levels of 9.5 (control), control plus .72% dietary urea (11.5U), control plus 1.44% dietary urea (13.5U), control plus abomasally infused casein (250 g/d; 11.5C), or control plus .72% dietary urea and abomasally infused casein (250 g/d; 13.5UC). All diets contained (DM basis) 80% ground corn, 15% corn silage, and 5% dry supplement and were provided for ad libitum consumption. Nitrogen intake increased (linear, P < .001) as CP increased from 9.5 to 13.5%. Portal-drained visceral release of ammonia N increased (linear, P < .10) as RDN increased, and was greater (P < .05) when protein was fed compared with heifers fed control (P < .10). Urea N removal by PDV was not affected ( P > . 10) by level of RDN but was greatest when 11.5C was fed and least when 13.5UC was fed. Net alpha-amino N (AAN) release by PDV was greatest when 13.5UC was fed (309 mmol/h), least when 9.5% CP was fed (112 mmol/h), and intermediate for the other groups (205 to 252 mmol/h). These data suggest that removal of N by the PDV may promote microbial protein synthesis when dietary RDN is low. When RDN needs have been met and amino acids are deficient for the host, escape protein should be fed to increase amino acid absorption.  相似文献

A feedlot growth performance experiment and 2 metabolism experiments were conducted to evaluate dietary roughage concentration and calcium magnesium carbonate in steers fed a high-grain diet. In Exp. 1, one hundred ninety-two crossbred yearling steers (320 +/- 10 kg of initial BW) were fed diets based on steam-flaked corn with 0, 0.75, or 1.5% CaMg(CO(3))(2). There were no effects (P > or = 0.13) on ADG, DMI, G:F, or total water intake due to CaMg(CO(3))(2). In Exp. 2, five ruminally and duodenally fistulated steers (263 +/- 9 kg of initial BW) were used in a 5 x 5 Latin square design, with 5 dietary treatments arranged in a 2 x 2 + 1 factorial: 1) 3.8% dietary roughage and no CaMg(CO(3))(2); 2) 7.6% dietary roughage and no CaMg(CO(3))(2); 3) 11.4% dietary roughage and no CaMg(CO(3))(2); 4) 3.8% dietary roughage and 1.5% CaMg(CO(3))(2); and 5) 7.6% dietary roughage and 1.5% CaMg(CO(3))(2). Water consumption was less (quadratic, P = 0.003) when 7.6% dietary roughage was fed compared with 3.8 or 11.4% dietary roughage. Intake of DM was not affected (P > or = 0.16) by dietary roughage or by CaMg(CO(3))(2). Poststomach and total tract starch digestion decreased (linear, P < 0.01) as dietary roughage increased. Ruminal pH tended (P = 0.08) to increase as dietary roughage increased but was not affected (P = 0.60) by CaMg(CO(3))(2). In Exp. 3, DMI and ruminal pH were continuously monitored in a 6 x 6 Latin square design using 6 ruminally and duodenally fistulated Holstein steers (229 +/- 10 kg of initial BW). A 3 x 2 factorial treatment structure was utilized, with factors consisting of dietary roughage concentration (4.5, 9.0, or 13.5%) and CaMg(CO(3))(2) inclusion (0 or 1.0%) to replace MgO and partially replace lime-stone. A dietary roughage x CaMg(CO(3))(2) interaction (P = 0.01) occurred as steers consuming 13.5% roughage, 1.0% CaMg(CO(3))(2) had greater DMI per meal than those consuming 4.5% dietary roughage, no CaMg(CO(3))(2) and 9.0% dietary roughage, 1.0% CaMg(CO(3))(2). Steers consuming 13.5% dietary roughage, 1.0% CaMg(CO(3))(2) and 9.0% dietary roughage, no CaMg(CO(3))(2) had greater meal length (min/meal; P = 0.01) than steers consuming 4.5% dietary roughage, no CaMg(CO(3))(2). Total tract OM digestibility decreased linearly (P = 0.01), and ruminal pH increased linearly (P = 0.01) with increasing dietary roughage concentration. Inclusion of CaMg(CO(3))(2) can replace limestone and MgO but did not produce ruminal pH responses similar to those observed by increasing dietary roughage in high-concentrate diets.  相似文献

Two experiments were conducted to examine the effect of previous BW gain during winter grazing on subsequent growth, carcass characteristics, and change in body composition during the feedlot finishing phase. In each experiment, 48 fall-weaned Angus x Angus-Hereford steer calves were assigned randomly to one of three treatments: 1) high rate of BW gain grazing winter wheat (HGW), 2) low rate of BW gain grazing winter wheat (LGW), or 3) grazing dormant tallgrass native range (NR) supplemented with 0.91 kg/d of cottonseed meal. Winter grazing ADG (kg/d) for HGW, LGW, and NR steers were, respectively, 1.31, 0.54, 0.16 (Exp. 1) and 1.10, 0.68, 0.15 (Exp. 2). At the end of winter grazing, four steers were selected randomly from each treatment to measure initial carcass characteristics and chemical composition of carcass, offal, and empty body. All remaining steers were fed a high-concentrate diet to a common backfat end point. Six steers were selected randomly from each treatment for final chemical composition, and carcass characteristics were measured on all steers. Initial fat mass and proportion in carcass, offal, and empty body were greatest (P < 0.001) for HGW, intermediate for LGW, and least for NR steers in both experiments. Live BW ADG and gain efficiency during the finishing phase did not differ (P = 0.24) among treatments, but DMI (% of mean BW) for NR and LGW was greater (P < 0.003) than for HGW steers. Final empty-body composition did not differ (P = 0.25) among treatments in Exp. 1. In Exp. 2, final carcass and empty-body fat proportion (g/kg) was greater (P < 0.03) for LGW and NR than for HGW steers. Accretion of carcass fat-free organic matter was greater (P < 0.004) for LGW than for HGW and NR steers in Exp. 1, but did not differ (P = 0.22) among treatments in Exp. 2. Fat accretion in carcass, offal, and empty body did not differ (P = 0.19) among treatments in Exp. 1, but was greater (P < 0.05) for LGW and NR than for HGW steers in Exp. 2. Heat production by NR steers during finishing was greater (P < 0.02) than by HGW steers in Exp. 1 and 2. Differences in ADG during winter grazing and initial body fat content did not affect rate of live BW gain or gain efficiency during finishing. Feeding steers to a common backfat thickness end point mitigated initial differences in carcass and empty-body fat content. However, maintenance energy requirements during finishing were increased for nutritionally restricted steers that were wintered on dormant native range.  相似文献

AIM: To obtain an estimate of the incidence of assistance at calving in primiparous (first-calving) beef heifers and the prevalence of breeding heifers at 15 months of age in New Zealand in 2006,and to identify factors contributing to farmers' decisions regarding breeding strategies for heifers, using a survey of beef cattle farmers.

METHODS: A questionnaire was sent to farmers listed in a Massey University database and to members of selected breed societies, as well as published in an industry newspaper; 331 valid responses were received. Information was gathered on the age and number of primiparous heifers, number of heifers assisted, and the importance of various reasons for and against breeding heifers at 15 months of age. Respondents were also required to outline the criteria used for selecting bulls to join with heifers, and the strategies used to manage dystocia in primiparous heifers.

RESULTS: Sixty-five (95% CI=58–71)% of respondents had only 2-year-old primiparous heifers in 2006, whilst a further 11 (95% CI=8–16)% had both 2- and 3-year-old primiparous heifers. The mean reported incidence of assisted calving was 7.0 (95% CI=6.4–7.5)% for 2-year-old primiparous heifers and 1.7 (95% CI=1.2–2.2)% for 3-year-old primiparous heifers. The reported incidence of assistance at calving within individual herdsranged from 0 to 100% for 2-year-old heifers. Respondents with bull-breeding herds most commonly observed their primiparous 2-year-old heifers twice daily, whilst respondents with commercial herds most commonly observed them once daily during calving. The most important reason for breeding heifers at 15 months of age was “increased profit”, whereas the most important reason for not breeding them at that age was “concern about rebreeding performance of 2-year-old heifers”. Estimated breeding value (EBV) for birth weight was the factor considered most frequently when selecting bulls to join with maiden heifers; age of bull and body shape of bull were the next most frequently considered factors. Selection of an appropriate bull was the most common strategy used to manage dystocia in 2-year-old beef heifers.

CONCLUSIONS: The majority of respondents calved their heifers at 2 years of age, and “increased profit” was the primary motivator. Concern about the rebreeding performance of 2-year-old heifers was the most important reason among the remainder of respondents for not breeding heifers at 15 months of age. Dystocia in 2-year-old heifers was “not a problem” or “a minorproblem” in most herds, but there was much variation amongst herds.  相似文献

AIM: To validate the use of para-aminohippuric acid (PAH) as a marker for measuring blood flow in wethers consuming a mixed diet of locoweed and blue grama hay. METHODS: Fourteen sheep, stratified by bodyweight (BW), were assigned to one of three treatments: 0.8 mg swainsonine (SW)/kg BW (HI), 0.2 mg SW/kg BW (LO), and no SW (Control). Sheep were fed various ratios of locoweed and blue grama hay to deliver SW treatments, for 28 days prior to infusion of PAH. Concentrations of SW and activities of alkaline phosphatase (Alk-P) and aspartate aminotransferase (AST) in serum were measured to confirm exposure to SW and subclinical intoxication. A single 20-ml injection of 5% PAH was delivered into the jugular vein after subclinical intoxication had been achieved. Blood samples were collected and serum analysed for PAH immediately prior to injection, then every 5 min from 5-30 min, and every 10 min from 30-60 min, following injection of PAH. RESULTS: Effective delivery of SW was evident from the greater concentrations of SW measured in the serum of HI compared with LO animals (p<0.05). No significant differences were detected in the rate of elimination (range 0.097-0.108 L/min), elimination half-life (range 6.62-7.24 min), apparent volume of distribution for the central compartment (range 7.14-9.72 L), and clearance (range 0.73-0.92 L/min) of PAH, between treatments. CONCLUSIONS: Subclinical intoxication with SW did not affect the pharmacokinetics of PAH. Thus, use of downstream dilution of PAH is a valid method to determine the rate of blood flow in nutrient flux experiments that involve consumption of locoweed.  相似文献

A feedlot experiment was conducted under commercial conditions in the Texas Panhandle using 3,757 feedlot steers (average of 94 steers/pen) to evaluate the effects of feeding zilpaterol hydrochloride with or without monensin and tylosin on feedlot performance and carcass characteristics. The experiment was conducted using a randomized complete block design. Treatments were arranged as a 2 (no zilpaterol vs. zilpaterol) x 2 (monensin and tylosin withdrawn vs. monensin and tylosin fed during the final 35 d on feed) factorial. Steers were fed for a total of 161 to 167 d, and treatments were administered during the final 35 d that cattle were on feed. When included in the diet, zilpaterol, monensin, and tylosin were supplemented at 8.3, 33.1, and 12.2 mg/kg (DM basis), respectively. Zilpaterol was included in the diet for 30 d at the end of the finishing period and withdrawn from the diet for the last 5 or 6 d cattle were on feed. Cattle were harvested and carcass data collected. There were no zilpaterol x monensin/tylosin interactions (P >or= 0.12) for ADG or G:F. Feeding zilpaterol increased ADG (P < 0.001) by 0.20 kg and G:F (P < 0.001) by 0.029 kg/kg during the last 35 d on feed. Likewise, when feedlot variables were measured throughout the entire 161- to 167-d feeding trial, ADG (3.4%; P < 0.001) and G:F (3.9%; P < 0.001) were increased. Feeding zilpaterol increased (P < 0.001) dressing percent and HCW and decreased (P < 0.001) total liver abscess rate compared with controls. In addition, zilpaterol increased (P < 0.001) LM area by an average of 8.0 cm(2). There was a zilpaterol x monensin/tylosin interaction (P = 0.03) for marbling score. Zilpaterol decreased (P < 0.001) marbling score regardless of monensin and tylosin treatment, although withdrawal of monensin and tylosin for 35 d decreased marbling to a greater extent (31 vs. 17 degrees). Zilpaterol decreased (i.e., improved; P < 0.001) calculated yield grade regardless of monensin and tylosin treatment, but feeding zilpaterol in combination with the withdrawal of monensin and tylosin for 35 d decreased calculated yield grade to a greater extent (0.49 vs. 0.29) compared with the zilpaterol, monensin, and tylosin combination treatment (zilpaterol x monensin/tylosin interaction, P = 0.03). Results suggest that monensin and tylosin can be withdrawn from the diet during the zilpaterol feeding period (final 35 d on feed) with minimal effect on animal performance, although feeding zilpaterol in combination with monensin and tylosin seemed to moderate effects on carcass quality.  相似文献

Twenty calves (7 mo old) were blocked by breed, sex, and weight into five groups of four calves and randomly assigned to either a 90% forage (alfalfa) or a 90% grain (50% sorghum and 50% wheat) diet fed at one (1M) or two (2M) times NEm for 140 d. Samples of ruminal epithelial tissue were collected from the anterior ventral sac, and papillae were cut free by hand and used for in vitro incubations and acyl-CoA synthetase assays. Substrates were acetate (90 mM), propionate (60 mM), butyrate (30 mM), and glucose (20 mM). Net productions of beta-hydroxybutyrate and acetoacetate from acetate were greater (P less than .01) with the 2M feeding; however, 14CO2 production from acetate was greater (P less than .05) with the grain diet. Net production of lactate (P = .09) and pyruvate (P less than .01) from propionate increased with the 2M feeding, whereas net lactate production from glucose decreased (P less than .01). Uptakes of VFA were similar with 1M and 2M feeding and were about 10-fold greater than uptakes of glucose. Production of 14CO2 from propionate was two- to fivefold greater than from acetate, butyrate, or glucose. Oxygen consumption was greater (P less than .01) with 2M feeding and unaffected by substrate. Activities of butyryl-CoA synthetase (nmol.mg of tissue-1.h-1) were greater (P less than .05) for animals consuming the forage diets. Addition of butyrate inhibited acetyl- and propionyl-CoA synthetase activity by 63 and 92%, respectively for all dietary treatments. Overall, metabolism of ruminal epithelial tissue tended to increase with the 2M feeding. Influence of dietary forage content on metabolism and activity of acyl-CoA synthetases was minimal, but the high-forage diet caused slight increases.  相似文献
