Carbohydrate digestion by α-glucosidase and subsequent glucose uptake at the brush border are critical for postprandial blood glucose control. Any specific inhibitors are useful as hyperglycemia modulating agents. In this study, it was postulated that an array of active components in mulberry leaf extract (MLE) may provide higher potency in inhibiting intestinal glucose absorption compared to the single component 1-deoxynojirimycin (DNJ), which is recognized as a promising inhibitor of intestinal glucose absorption. Both MLE and DNJ were active in inhibiting α-glucosidase. However, in Caco-2 cells, only MLE showed significant inhibition of 2-deoxyglucose uptake, whereas DNJ was ineffective. For glucose loading, co-administration of MLE resulted in potent inhibitions of glucose responses compared to those by DNJ in Sprague Dawley (SD) rats, but this was not found for maltose loading. These novel findings add evidence that the unabsorbed phytochemicals in MLE compete with glucose for intestinal glucose transporters, but DNJ itself does not. We also evaluated the timing of MLE consumption. By administering MLE for 30 min before glucose loading, the incremental area under the curve (IAUC) was significantly lowered in the rats, as compared to a simultaneously administered group. Similarly, cellular glucose uptake was significantly reduced in Caco-2 cells following pretreatment.  相似文献

The effects of cumin and ginger as antioxidants on dough mixing properties and cookie quality were evaluated. Antioxidant activities in cookies were estimated by total phenolic compounds (TPC) contents and free radical scavenging activities. The cookie dough development was evaluated using Mixolab equipment which showed that addition of cumin did not change dough stability and C2, but decreased C3 and C4. While the addition of 5% ginger decreased dough stability (from 8.4 in the control sample to 6.7 min with 5% ginger addition), C2 (from 0.49 in the control sample to 0.31 N·m with 5% ginger addition), C3, and C4. Cookies formulated with addition of cumin and ginger had increased spread ratios, were softer, and had lower L* and b* values (were darker) than the control. Sensory analysis showed that cookies with cumin and ginger additions had overall acceptability similar to that of the the control with a slightly darker appearance, as confirmed by color determination. Using cumin and ginger significantly increased TPC contents from 78.5 in the control to 93.0 and 109.8 mg of gallic acid equivalent/100 g, respectively. Similar results were observed in the antioxidant activity measured by 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH), which increased from 41.0% in the control to 51.5% and 64.6%, respectively, for cookies with 5% addition of cumin and ginger.  相似文献

Variation of polymeric proteins affects wheat end‐use quality. This research investigated associations of polymeric proteins with dough mixing strength and breadmaking characteristics in a near‐homogenous population of 139 recombinant inbred lines (RILs) derived from a cross between two hard red spring wheat breeding lines. Flours from the RILs grown at three locations were analyzed for molecular weight (MW) distribution of SDS‐extractable and unextractable proteins using size‐exclusion HPLC protocol. Correlations were calculated between mixing and breadmaking properties and HPLC absorbance data obtained a 0.01‐min retention time interval to identify protein fractions that had a significant effect on the quality traits. Very high MW polymeric proteins in the unextractable fraction had more distinct and positive associations with dough mixing strength and bread loaf volume than did other polymeric protein fractions, whereas extractable polymeric had negative influence. Consequently, the ratio of unextractable very high MW polymeric proteins to extractable polymeric proteins had greater correlations with dough mixing parameters than other HPLC absorbance area data. Covariate‐effect biplots also visually validated positive effects of unextractable very high MW polymeric proteins and negative effects of extractable polymeric proteins on mixing properties and loaf volume across three growing locations.  相似文献

Cultivated Chinese wheat germplasm has been a valuable genetic resourcein international plant breeding. Landraces endemic to China are a geneticresource that is distinct from other wheat germplasm. Patterns of geneticdiversity among cultivated Chinese accessions and relationship to othergermplasm pools are unknown, despite the proven value and potential novelty. Theobjective of this work was to determine the level of genetic diversity withinimproved Chinese germplasm in the context of several other wheat germplasmpools. We analyzed a set of improved accessions cultivated from the 1940s to the1990s in Shaanxi province, China, using six simple sequence repeat (SSR) primerpairs and 30 restriction fragment length polymorphism - probe enzymecombinations (RFLP-PEC) previously used to characterize 21 geographically basedgermplasm pools. Shaanxi germplasm consists of three groups based on foreignintroductions from Italy, Australia, Denmark, and Russia. There was a decreasein genetic diversity among Shaanxi accessions cultivated in the 1970s and 1980sto the 1990s, and accession classifications based on primary decade ofcultivation were found to be significantly undifferentiated. The analysis of themean genetic distance among 22 geographically based pools of germplasm suggestsseveral regions are significantly undifferentiated. A vast majority of the totalamount of variation was found within pools; therefore, pools appear to belargely differentiated based on small differences in band relative frequency andfew if any unique bands. Previous studies have identified some Chinese landracepools as morphologically and genetically unique. The Shaanxi pool does not havethe same unique morphological or genetic features, nor is it more similar to thelandrace pools than other improved germplasm pools.  相似文献

Plants tolerant to polluted environments were selected, based on several criteria, to restore a coastal forest ecosystem severely damaged by air pollutants discharged from an industrial complex. In addition, a restoration plan was prepared synthesizing these results and the diagnostic ecological indicators in the area for which restoration is required. Pollution-tolerant plants of 11 tree and subtree species, 10 herb species and one shrub species were selected from field surveys in the vicinity of two representative industrial complexes in Korea, Ulsan and Yeocheon. Nine species were selected for tolerance to SO2 fumigation and six species were selected for tolerance to Al3+. Growth and photosynthetic responses of sample plants transplanted into polluted and unpolluted sites showed that 15 species out of the 26 sample plants showed a disposition for tolerance. Most of these are endemic plants and they are composed of diverse species in structure and function. This result implies that these tolerant species could play important roles in the restoration of the study area, which has several specific features. On the other hand, results from transplant tests indicate that a field survey is the most reasonable method for selection of tolerant plants to restore a pollution-damaged ecosystem, as was shown in another restoration program. Results of ecological analysis on vegetation map indicate that the spatial range within the first ridge is the sector for which restoration is required. This sector was classified into four zones on the basis of topographic conditions: lower and upper slopes of both slopes facing and opposite the pollution source. Guidelines for soil amelioration and arrangement of tolerant plants were prepared considering the degree of vegetation degradation, leaf damage of major plant species and soil pollution in each zone under the restoration plan.  相似文献

The combined and separate effects of Cd and wood ash on Archaea from coniferous forest humus were studied in a microcosm experiment. Nonmetric multidimensional scaling of the denaturing gradient gel analysis of polymerase chain reaction amplified 0.9 kb 16S ribosomal DNA fragments revealed changes in archaeal communities due to the ash treatments. Cd with or without ash did not further influence the result. Representatives of the ash and control communities were cloned, grouped by restriction fragment length polymorphism analysis and finally sequenced. All sequences belonged to non-thermophilic Crenarchaea.  相似文献

The effect of spraying exogenous plant hormone methyl jasmonate (MeJA) upon radish sprout (Raphanus sativus L.) was investigated in aspects of total phenolic content (TPC), isothiocyanate content, antioxidant activity of the radish extract, and enzymatic activities of phenylalanine ammonia lyase (PAL) and myrosinase. The MeJA treatment significantly increased the TPC that resulted in the increased DPPH* (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging capacity. In addition, the PAL activity also increased by 60% at 24 h after MeJA treatment. However, the same treatment decreased the amount of 4-methylthio-3-butenylisothiocyanate (MTBITC), a major isothiocyanate in radish sprout and the activity of myrosinase, an enzyme related to produce isothiocyanates.  相似文献

In this study, we report Monopis crocicapitella (Clemens, 1859) (Tineidae), which was collected from bat guano in a cave in the southern region of Korea, for the first time from East Asia. We briefly redescribe and illustrate the external morphology and genital structures of both sexes. Pine (Pinus densiflora) bark was chemically modified by grafting with acrylonitrile (AN) through Fenton reaction and conversion of the cyano group to an amidoxime group by reaction with hydroxylamine. Treatments of pine bark with 160 mM H2O2 at 40°C yielded ~70% grafting in 6-h reaction. After alkali hydrolysis of amidoximated pine bark (AOPB), its adsorption capacity (q e) for copper ions was largely increased from 0.5 to 1.8 mmol/g. Adsorption of the hydrolyzed AOPB (HAOPB) followed the pseudo-second-order kinetic model. Among the metal ions tested, the order of adsorption capacity (q e) predicted by the kinetic model at equilibrium time was Hg2+ > UO2 2+ > Cd2+ > Cu2+ > Zn2+ > Ni2+. The adsorption process was pH-dependent, and the adsorption equilibrium was observed in the pH range of 6–7 for all the metal ions. Even though pine bark is a good adsorbent for Hg2+ and UO2 2+ ions, it had a poor affinity for the other ions tested. It was obviously found that HAOPB had very high adsorption abilities for heavy metal and uranyl ions.  相似文献

Here, we report evidence for the production of ozone in human disease. Signature products unique to cholesterol ozonolysis are present within atherosclerotic tissue at the time of carotid endarterectomy, suggesting that ozone production occurred during lesion development. Furthermore, advanced atherosclerotic plaques generate ozone when the leukocytes within the diseased arteries are activated in vitro. The steroids produced by cholesterol ozonolysis cause effects that are thought to be critical to the pathogenesis of atherosclerosis, including cytotoxicity, lipid-loading in macrophages, and deformation of the apolipoprotein B-100 secondary structure. We propose the trivial designation "atheronals" for this previously unrecognized class of steroids.  相似文献
