The cryopreservation of spermatozoa constitutes a valuable tool for the captive breeding management of valuable and/or threatened species. Chinchilla lanigera is a species almost extinct in the wild, and the domestic counterpart has one of the most valuable pelts in the world. The objectives of this study were to: (i) compare the functional activity of post‐thawed chinchilla spermatozoa cryopreserved at ?196°C either with glycerol (G) or ethylene glycol (EG) as cryoprotectants (1 m final concentration) and (ii) investigate the effects of incubating the gametes for 4 h in the presence or in the absence of the cryoprotectants; evaluations were performed taking into account motility, viability, response to hypo‐osmotic shock and acrosome integrity of the cells. Parameters reflecting post‐thaw (0 h) sperm functional activity were significantly lower than those of freshly ejaculated gametes. When comparing the cryoprotectant efficiency of G vs EG, neither cryoprotectant agent offered appreciable advantages. After 4 h of incubation, in the presence or absence of the cryoprotectant agent, a rapid and significant decrease was found in all functional parameters and remained at ~ 20–30% motile, viable and viable acrosome intact cells. Viability was significantly lower when the cryoprotectant was removed from the media (possibly due to the centrifugation process). With respect to the maintenance of sperm membrane integrity, only ~ 10% of cells showed membrane resistance to hypo‐osmotic conditions after the 4 h incubation period. These results constitute new insights for cryopreservation protocols and the development of assisted reproductive techniques in this species.  相似文献

Global minimum temperatures (TMIN) are increasing faster than maximum temperatures, but the ecological consequences of this are largely unexplored. Long-term data sets from the shortgrass steppe were used to identify correlations between TMIN and several vegetation variables. This ecosystem is potentially sensitive to increases in TMIN. Most notably, increased spring TMIN was correlated with decreased net primary production by the dominant C4 grass (Bouteloua gracilis) and with increased abundance and production by exotic and native C3 forbs. Reductions in B. gracilis may make this system more vulnerable to invasion by exotic species and less tolerant of drought and grazing.  相似文献

Abstract  The migratory behaviour of hatchery-reared Atlantic salmon, Salmo salar L., post-smolts during the first phase of the marine migration was examined to assess their susceptibility to salmon lice, Lepeophtheirus salmonis (Krøyer), infestations. Swimming depths of eight post-smolts relative to the measured salinity and temperature were monitored for an average of 11.4 h following release outside the mouth of the River Eio using depth sensitive acoustic transmitters. Vertical salinity and temperature distributions were simultaneously recorded along the migratory route. Mean swimming depth was 1.7 m (individual mean 0.5–2.1 m). There was no overall preference among all the post-smolts for specific salinity concentrations. Typically post-smolts migrated the majority (68%) of their time at salinities <20 psu (brackish water), and as a result outside the reported salinity tolerances of sea lice. Furthermore, post-smolts chose the warmest water layer during their coastal migration.  相似文献

  • 1. Aerial photograph classification was used to map perennial thick canopy seagrass presence/absence over a large area (85 km2) off the coast of Western Australia. Within those areas mapped as seagrass, a geostatistical nonparametric interpolation method was applied to map the probability of seagrass species presence from underwater tow video. Multiple species mixtures were mapped at fixed probability thresholds of 0.95, 0.75, 0.50, and 0.25. Taxa included Amphibolis spp., Posidonia coriacea, P. sinuosa, P. australis and ephemeral species (Halophila and Zostera tasmanica (newly named as Heterozostera polychlamys)).
  • 2. The most commonly occurring species were respectively Amphibolis spp., Posidonia coriacea, P. sinuosa, P. australis, and the ephemeral species. Amphibolis, P. coriacea, and the ephemeral species were mapped predominantly as mixed assemblages (71–89% mixed), whereas P. sinuosa and P. australis were typically mapped as single species.
  • 3. Different species growth habits led to distinctive differences in large area distributions. All species were highly variable over short distances (<500 m), and spatial dependence persisted over more than 5 km. However, Posidonia sinuosa meadows were oriented with the longest axis running north–south, and a shorter axis running east–west perpendicular to the coastline (spatial dependence to 2.8 km and 0.8 km, respectively). The ephemeral species were less successfully mapped, largely owing to the potentially different growth patterns of the grouped species, and because their full extent could not be captured by the aerial photograph classification.
  • 4. The individual biology of each species results in unique landscape features where Posidonia sinuosa forms larger continuous and predominantly monospecific meadows, whereas the more common Amphibolis and P. coriacea form multi‐species patchy meadows. These mapped features suggest that the emergence of species patterns in seagrass landscapes is influenced by differences in clonal growth among seagrass species.
  • 5. Probabilistic species mapping provided information unavailable from discretely classified maps, and facilitates targeted sampling for improving map accuracy, and for more realistically evaluating species and mixed species distribution predictions. The kriging approach, although not well suited for all types of vegetation data, performed well for clonal seagrasses.

A boundary between compositionally distinct regions at a depth of about 1600 kilometers may explain the seismological observations pertaining to Earth's lower mantle, produce the isotopic signatures of mid-ocean ridge basalts and oceanic island basalts, and reconcile the discrepancy between the observed heat flux and the heat production of the mid-ocean ridge basalt source region. Numerical models of thermochemical convection imply that a layer of material that is intrinsically about 4 percent more dense than the overlying mantle is dynamically stable. Because the deep layer is hot, its net density is only slightly greater than adiabatic and its surface develops substantial topography.  相似文献

Before and during the first week of the March-April 1980 eruptions of Mount St. Helens, Washington, infrared thermal surveys were conducted to monitor the thermal activity of the volcano. The purpose was to determine if an increase in thermal activity had taken place since an earlier airborne survey in 1966. Nine months before the eruption there was no evidence of an increase in thermal activity. The survey during the first week of the 1980 eruptions indicated that little or no change in thermal activity had taken place up to 4 April. Temperatures of ejected ash and steam were low and never exceeded 15 degrees C directly above the vent.  相似文献

Summary Two cloned cassava genes (pCAS6, pHNL) and a barley gene (pBLT63) which is highly conserved in higher plants, were evaluated as potential multilocus probes for RFLP analysis of phylogenetic relationships of species within the genus Manihot. For M. rubricaulis, M. chlorosticta and M. esculenta subsp. flabellifolia dendrograms based on 13 probe-restriction enzyme combinations show very little variation between accessions of the same species. A close relationship is demonstrated between the Mexican species M. chlorosticta and the M. esculenta subsp. flabellifolia accessions, which does not support the classification of flabellifolia as a true South American wild species.  相似文献
