Pseudorabies virus (PRV), an alpha-herpesvirus, causes substantial economic losses in the swine industry and is currently the focus of eradication and control programs. Some of these programs rely on the ability of veterinarians to differentiate animals exposed to virulent strains of PRV from animals exposed to avirulent vaccine strains of PRV on the basis of a serologic response to nonessential glycoproteins that are deleted in some vaccine strains of PRV. Genetic recombination resulting in the creation of virulent strains of PRV with the same negative immunologic markers as vaccine strains could disrupt these programs. Two strains of PRV were coinoculated either into tissue culture or into sheep to facilitate recombination. Progeny viruses were selected to detect a specific recombinant phenotype. We were able to detect genetic recombination between vaccine strains of PRV following in vitro or in vivo coinoculation of 2 strains of PRV. The selected recombinants had marker-deleted phenotypes in strains with restored virulence genes. Summary The role of soluble organic carbon (SOC) in denitrification in four mineral soils and one organic soil was evaluated in laboratory studies. Denitrification capacities and SOC concentrations were determined by nitrate loss from air-dried flooded soil treated with a solution containing 100 g/ml N03
–-N, while the rate of consumption was measured by Warburg manometry on 20 g air-dried soils to which 10 ml water had been added. High correlation coefficients (r > 0.93) were obtained between denitrification capacities, SOC, and oxygen consumption in the five soils. A mineral soil was amended with extracts of an organic soil. After incubating for 1 week, denitrification capacity was enhanced and SOC concentrations decreased in that soil. Extracted mineral soil had a lower denitrification capacity than an unextracted one. Decreases in concentrations of SOC were related to color change. Infrared spectra of precipitates from soil extracts indicated that absorption at wave number 1420–1440 cm -1 was also related to the color changes. It was implied that low molecular weight fulvic acid like compounds represented the SOC mineralized in denitrification, and that their supply to soil solution by solubilization of organic matter influenced the denitrification rate in the soil. 相似文献

Potential uses of dredged sediments have raised questions about leaching of contaminants from such material once transported to other locations and exposed to other environments. In order to estimate leachable, i.e. available fractions of chemical elements in such materials, a number of analytical protocols were proposed, of which none is internationally accepted as a standard procedure. As a consequence, comparability of the results obtained by different protocols in various laboratories is difficult to demonstrate. In the present study some commonly used protocols including a leaching test with (i) seawater, following the modified German Standard Method (DIN), (ii) a single extraction with 25% acetic acid, following the method proposed by the United Nations Environment Programme (UNEP), and (iii) a three-step sequential extraction, following both the original and the modified protocols of the Standards, Measurements and Testing Programme (formerly Community Bureau of Reference – BCR), were performed for assessing the availability of lead, zinc, nickel and copper leachable from dredged marine sediments. A comparison of the results obtained by the different protocols indicated that the single extraction with acetic acid is a reliable simplified technique and allows more rapid evaluations of dredged areas of coastal lagoons in the Northern Adriatic. Uncoated double-stranded DNA dissolved in a salt solution was deposited on graphite and imaged in air with the scanning tunneling microscope (STM). The resolution was such that the major and minor grooves could be distinguished. The pitch of the helix varied between 27 and 63 angstroms in the images obtained. We review Phanerozoic sea-level changes [543 million years ago (Ma) to the present] on various time scales and present a new sea-level record for the past 100 million years (My). Long-term sea level peaked at 100 +/- 50 meters during the Cretaceous, implying that ocean-crust production rates were much lower than previously inferred. Sea level mirrors oxygen isotope variations, reflecting ice-volume change on the 10(4)- to 10(6)-year scale, but a link between oxygen isotope and sea level on the 10(7)-year scale must be due to temperature changes that we attribute to tectonically controlled carbon dioxide variations. Sea-level change has influenced phytoplankton evolution, ocean chemistry, and the loci of carbonate, organic carbon, and siliciclastic sediment burial. Over the past 100 My, sea-level changes reflect global climate evolution from a time of ephemeral Antarctic ice sheets (100 to 33 Ma), through a time of large ice sheets primarily in Antarctica (33 to 2.5 Ma), to a world with large Antarctic and large, variable Northern Hemisphere ice sheets (2.5 Ma to the present). 相似文献

Molecular and antigenic analyses of three influenza viruses isolated from outbreaks of severe respiratory disease in racing greyhounds revealed that they are closely related to H3N8 equine influenza virus. Phylogenetic analysis indicated that the canine influenza virus genomes form a monophyletic group, consistent with a single interspecies virus transfer. Molecular changes in the hemagglutinin suggested adaptive evolution in the new host. The etiologic role of this virus in respiratory disease was supported by the temporal association of rising antibody titers with disease and by experimental inoculation studies. The geographic expansion of the infection and its persistence for several years indicate efficient transmission of canine influenza virus among greyhounds. Evidence of infection in pet dogs suggests that this infection may also become enzootic in this population. 相似文献

The polyketide epothilone is a potential anticancer agent that stabilizes microtubules in a similar manner to Taxol. The gene cluster responsible for epothilone biosynthesis in the myxobacterium Sorangium cellulosum was cloned and completely sequenced. It encodes six multifunctional proteins composed of a loading module, one nonribosomal peptide synthetase module, eight polyketide synthase modules, and a P450 epoxidase that converts desoxyepothilone into epothilone. Concomitant expression of these genes in the actinomycete Streptomyces coelicolor produced epothilones A and B. Streptomyces coelicolor is more amenable to strain improvement and grows about 10-fold as rapidly as the natural producer, so this heterologous expression system portends a plentiful supply of this important agent. 相似文献
