In Colombia, more than 1 million tons of coffee pulp are produced every year. Its transformation into compost by means of turned piles has led to a final product with poor physical and chemical characteristics and vermicomposting has been suggested as an alternative method of transforming these wastes into a useful organic fertilizer. The ability of the earthworm Eisenia fetida to transform coffee pulp into a valuable compost was evaluated. The influence of bed depth and time on different C fractions, N content and availability of nutrients was studied. The results showed that the C and N contents were not affected by the depth of the bed, whereas time affected both. An increase in the fractionation ratio, determined by calculating the C in the fraction smaller than 100 m as a percentage of C in the samples as a whole, and low values of humic-like substances were recorded during vermicomposting. After ingestion of the pulp by the earthworms, an increase in available P, Ca, and Mg but a decrease in K were detected.  相似文献

A collaborative study was performed in 13 laboratories to validate an enzyme immunoassay (EIA) procedure for rapid detection of Salmonella in foods. The EIA was compared with the standard culture procedure for detection of Salmonella in 6 food types: ground black pepper, soy flour, dried whole eggs, milk chocolate, nonfat dry milk, and raw deboned turkey. Uninoculated and inoculated samples were included in each food group analyzed. There was no significant difference in the proportion of samples positive by the EIA and culture procedures at the 5% level for any of the 6 foods. The enzyme immunoassay screening method has been adopted official first action as a rapid screening method for detection of Salmonella.  相似文献

A collaborative study was conducted to validate the use of the AOAC alkaline phosphatase method for mammalian feces in corn meal, 44.B01-44.B06, for 7 additional products: brown rice cream, oat bran, grits, semolina, pasta flour, farina, and barley plus (a mixture of barley, oat bran, and brown rice). The proposed method determines the presence of alkaline phosphatase, an enzyme contained in mammalian feces, by using phenolphthalein diphosphate as the enzyme substrate in a test agar medium. Fecal matter is separated from the grain products by specific gravity differences in 1% test agar. As the product is distributed on liquid test agar, fecal fragments float while the grain products sink. The alkaline phosphatase cleaves phosphate radicals from phenolphthalein diphosphate, generating free phenolphthalein, which produces a pink to red-purple color around the fecal particles in the previously colorless medium. Collaborators' recovery averages ranged from 21.7 particles (72.3%) for oat bran to 25.3 particles (84.3%) for semolina at the 30 particle spike level. Overall average background was 0.4 positive reactions per food type. The collaborators reported that the method was quick, simple, and easy to use. The method has been approved interim official first action for all 7 grain products.  相似文献

Summary The use of N and P by mixed and by sole cropping (crop rotation) of maize and cowpeas were compared in a field experiment on an Alfisol at the Nyankpala Agricultural Experiment Station in the northern Guinea Savanna of Ghana, using two levels of N (0 and 80 kg N ha-1 year-1 as urea) and P application (0 and 60 kg P ha-1 year-1 as Volta phosphate rock). Maize grain yields were significantly reduced in the mixed cropping system. This yield difference became smaller with the application of N and P fertilizer. The N and P concentrations in maize ear leaves at silking indicated that a deficiency in N and P contributed to the maize yield depression in mixed cropping. Competition for soil and fertilizer N between maize and cowpeas was suggested by: (1) A similarity in total N uptake between the two cropping systems; (2) efficient use of soil nitrate by the cowpeas; and (3) low N2 fixation by the cowpeas, calculated with the aid of an extended-difference method. In general, N2 fixation was low, with the highest values in the sole cropping (53 kg ha-1) and a substantial reduction in the mixed cropping system. The application of N fertilizer further reduced N2 fixation. This was substantiated by nodule counts. The lower N2 fixation in the mixed cropping system was only partly explained by the lower density of cowpeas in this system. In addition, dry spells during the cropping season and shading by the maize component could have reduced the nodulation efficiency. No N transfer from the legume/rhizobium to the non-legume crop was observed. Impaired P nutrition in the mixed compared with the sole-cropped maize might have been due to less P mobility in the soil. This was indicated by lower soil moisture contents in the topsoil under mixed cropping, especially during the dry year of 1986. The results show that mixed cropping of maize and cowpeas did not lead to improved use of soil and fertilizer N and P or to an enhanced N2 fixation. On the contrary, an annual rotation of maize and cowpeas was clearly superior.  相似文献

Summary The influence of more than 100 years of fertilization with farmyard manure on soil organic matter in comparison to unfertilized soil was studied in particle-size fractions using elemental (C and N) analyses and pyrolysis-field ionization mass spectrometry. Distinct differences in C and N concentrations and distribution and in the quality of organic matter between the size fractions and the fertilization treatments were observed. Clay-associated C and N were relatively higher in the unfertilized treatment, whereas the application of farmyard manure preferentially increased soil organic matter associated with the fine and medium silt fractions. Pyrolysis-field ionization mass spectrometry of soil fractions <20 m showed increasing values for lignin monomers and dimers and fatty acids with larger equivalent diameters, whereas the proportion of N compounds, mono- and polysaccharides and phenolics decreased in the larger size fractions. Sand fractions were particularly rich in lignin fragments, mono- and polysaccharides, and alkanes/alkenes. These relationships seemed to be independent of management practices. In the same size fractions of the different treatments, however, a higher relative abundance of N-compounds, mono- and polysaccharides, phenolics, lignin monomers, and alkanes/alkenes was observed in the unfertilized variant. Lignin dimers and fatty acids were more abundant in the farmyard manure treatment. Both trends together imply that soil enrichment in organic matter due to the application of farmyard manure largely reflects an increase in lignin building blocks and partly reflects an increase in lipids such as fatty acids in the silt fractions. Therefore these constituents are of particular importance in assessing the positive effects of farmyard manure on soil fertility.  相似文献

An enzymatic method for the determination of free glutamic acid in meat products and dried soups was collaboratively studied in 11 laboratories. In the presence of the enzyme glutamate dehydrogenase, L-glutamic acid is oxidatively deaminated by nicotinamide adenine dinucleotide (NAD) to 2-oxoglutarate. In a reaction catalyzed by diaphorase, the NADH thus formed converts 2-(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyltetrazolium chloride to a formazan, which is measured in the visible range at 492 nm. Fourteen samples (7 samples of minced sausage and 7 samples of dried cauliflower soup) with glutamate contents varying between 0.4 and 16 g/kg were included in the study. Materials were distributed to participants as blind duplicates and as split level pairs. The mean relative standard deviation (RSDR) for reproducibility for the dried soup material containing glutamate between 7 and 16 g/kg was 4.6%. RSDR values for samples of minced sausage containing glutamate at lower levels (0.4-1.3 g/kg) were between 12 and 16%.  相似文献

An analytical procedure was developed for determination of benzene in polypropylene food packaging and was adapted for determination of benzene in commercial paraffin waxes intended for food-contact use. The polymer was dissolved in hexadecane at 150 degrees C. The wax was melted in an 80 degrees C oven. A simple helium-sparging apparatus was used to remove the volatile chemical from the polymer or wax. The contaminant was collected in methanol, distilled water was added, and the resulting solution was analyzed by headspace gas chromatography. The instrument was equipped with a 30 m fused silica open tubular capillary column and a photoionization detector. Average recoveries of benzene from polymer and paraffin wax at low parts-per-billion concentrations were 63 and 70%, respectively. Limits of detection and quantitation for analysis of polypropylene were 8 and 17 ppb, respectively; the limit of quantitation for analysis of paraffin wax was 2 ppb. In several commercial polypropylene products examined, benzene levels ranged from none detected to 426 ppb. In 3 commercial waxes examined, concentrations of 16-73 ppb benzene were determined. The presence of benzene was confirmed by gas chromatography/mass spectrometry.  相似文献

A closed vessel microwave digestion procedure was developed for shellfish samples. This procedure was compared with wet and dry ash procedures for levels of lead, cadmium, chromium, copper, and zinc. Results obtained for microwave and conventional wet ash digestion were comparable. The dry ashing procedure produced results consistently lower than either of the other methods. Recoveries ranged from 80-92% for microwave and wet ashing procedures and 54-72% for the dry ashing procedure. Accuracy was also determined by analyzing lobster hepatopancreas marine reference material. Values for Pb, Cd, and Cr fell within the range specified for the reference material for all 3 digestion procedures; however, values were lower for Cu and Zn. Results of this study show that microwave digestion is comparable to wet ashing.  相似文献

 Maize (Zea mays L.) and sunflower (Helianthus annuus L.) grown on a calcareous soil showed poor growth and/or were chlorotic in spite of abundant Fe in the roots. It has been hypothesized that microbial siderophores chelate Fe (III) in the soil, and that in this form Fe is transported towards the root apoplast. On the calcareous soil, total and apoplastic root Fe concentrations were high, probably because of a high apoplastic pH depressing Fe (III)-reductase activity and thus the Fe2+ supply to the cytoplasm. On the acidic soil, total and apoplastic root Fe concentrations were low, probably because of a low apoplastic pH favouring Fe (III) reduction, hence plants showed no Fe-deficiency symptoms. The main objective of the present work was to investigate the role of microbial soil activity in plant Fe acquisition. For this purpose, plants were grown under sterile and non-sterile conditions on a loess loam soil. Plants cultivated under non-sterile conditions grew well, showed no Fe-deficiency symptoms and had fairly high Fe concentrations in the roots in contrast to plants grown in the sterile medium. Low root and leaf Fe concentrations in the axenic treatments indicated that the production of microbial siderophores was totally suppressed. Accordingly, sunflowers were severely chlorotic and this was associated with very poor growth, whereas in maize only growth was drastically reduced. In maize under sterile conditions, root apoplastic and total Fe concentrations were not as low as in sunflowers, which may have indicated that phytosiderophores produced in maize partly sustained Fe acquisition, but due to poor growth were not as efficient in supplying Fe as microbial activity under natural conditions. Carbohydrates are known to be important precursors in the development of roasted peanut quality. However, little is known about their genotypic variation. A better understanding of the role of carbohydrates in roasted peanut quality requires first an understanding of the genotypic variation in the soluble carbohydrate components. Ion exchange chromatography was used to isolate 20 different carbohydrate components in 52 genotypes grown in replicated trials at two locations. Inositol, glucose, fructose, sucrose, raffinose, and stachyose were quantitated, and 12 unknown peaks were evaluated on the basis of the peak height of the unknown relative to the cellobiose internal standard peak height. Peaks tentatively identified as verbascose and ajugose could not be properly integrated because of tailing. Of the 18 carbohydrates that were estimated, 9 exhibited significant variation between test environments, 5 among market types, 14 among genotypes within market types, and 11 exhibited some significant form of genotype x environment interaction. Genotypes accounted for 38-78% of the total variation for the known components, suggesting that broad-sense heritability for these components is high. The observed high genotypic variation in carbohydrate components is similar to the high genotypic variation observed for the sweetness attribute in roasted peanuts, which raises the question regarding possible interrelationships. The establishment of such interrelationships could be most beneficial to peanut breeding programs to ensure the maintenance of flavor quality in future peanut varieties.  相似文献
