In vitro comparison of the effects of two forms of hydroxyethyl starch solutions on platelet function in dogs

OBJECTIVE: To evaluate the effect of 2 hydroxyethyl starch (HES) preparations (ie, HES solution with a molecular weight of 600 kd and a degree of substitution of 0.7 [HES 600/0.7] and a calcium-containing polyionic HES solution with a molecular weight of 670 kd and a degree of substitution of 0.75 [HES 670/0.75]) on canine platelet function. SAMPLE POPULATION: Blood samples from 10 healthy adult dogs. PROCEDURES: Dilution of citrated whole blood was performed with saline (0.9% NaCl) solution, HES 600/0.7, and HES 670/0.75 at ratios of 1:9 (ie, 1 part saline solution or colloid to 9 parts whole blood) and 1:3. Measurements of time to platelet plug formation in a capillary tube (ie, closure time) were made by use of a bench-top platelet function analyzer with collagen and ADP platelet agonists. RESULTS: Mean baseline closure time was 68.0 +/- 15.3 seconds. A 1:3 dilution of whole blood with saline solution, HES 600/0.7, and HES 670/0.75 resulted in mean closure times of 85.8 +/- 15.7 seconds, 100.6 +/- 18.6 seconds, and 101.6 +/- 16.2 seconds, respectively. Closure time following 1:3 dilution of whole blood with saline solution was significantly different from baseline and from 1:9 dilution with saline solution. Closure time following 1:3 dilution of whole blood with HES 670/0.75 was significantly different from baseline, 1:3 and 1:9 dilutions with saline solution, and 1:9 dilutions with HES 600/0.7 or HES 670/0.75. CONCLUSIONS AND CLINICAL RELEVANCE: Saline solution, HES 600/0.7, and HES 670/0.75 affect canine platelet function by prolonging closure times; HES solutions prolonged closure time to a greater extent than saline solution.     

Use of bone morphogenetic proteins for augmentation of bone regeneration

A large body of preclinical and clinical data now documents that recombinant BMPs can be used for skeletal regeneration in humans and animals. Recombinant human BMP-2 and BMP-7 have been approved for use in human patients with long-bone fractures and nonunions and in patients undergoing lumbar fusion or various maxillofacial and dental regenerative procedures. These products have also been made available for veterinary use.     

禾谷镰刀菌产生的细胞壁降解酶的特性研究

禾谷镰刀菌(Fusarium gramimearum<\i>)是引起玉米茎腐病、穗腐病的主要病原菌之一。该病原菌在活体外能产生PG、PMG、PGTE和PMTE四种细胞壁降解酶,这些酶都具有酶的典型性动力学特性。细胞壁降解酶是F.gramimearum<\I>致病的一个重要因子。     

Is N‐feedback involved in the regulation of nitrogenase activity in Medicago truncatula?

To determine whether senescing leaves provoke an active nitrogen (N) remobilization that results in the reduction of nitrogenase activity, 60% of Medicago truncatula lower leaves were either darkened or individually excised for two weeks. Although a considerable amount of N was remobilized, N₂ fixation activity was found to be increased to maintain the N source/sink balance, indicating an absence of the negative N‐feedback regulation of nitrogenase activity in the senescing M. truncatula.     

Predictions of soil water and crop growth dynamics using the agroecosystem models THESEUS and OPUS

The agroecosystem models THESEUS and OPUS were tested with data obtained from three agricultural experimental field plots on sandy soils without groundwater located at the moraine landscape in East Brandenburg, Germany. At each of these plots, a separate agricultural management practice was applied. Measurements of soil water contents, pressure heads, above‐ground crop biomass, and crop yield from these three plots were compared with the corresponding simulation results of both models. The comparisons of simulated with measured outputs were analyzed using the modeling‐efficiency index IA. According to these analyses, both models simulated adequately the time courses of volumetric soil water contents and above‐ground crop biomass, but the time courses of pressure heads were predicted with a lower quality by both models. As for the pressure heads, the yields simulated with both models showed greater discrepancies in comparison with the observed ones. This indicates the need of a site‐specific parameter calibration of the crop‐growth modules, especially for that included in OPUS .     

基于ETM+数据的新疆于田地区地表温度反演研究

地表温度是地球系统的热状况与水热平衡的关键因子.热红外遥感是获取地表热状况信息的一种非常重要的手段.利用ETM+数据和同时相气象数据获得两个地表温度反演的重要参数:大气透射率和地表比辐射率.运用覃志豪等的单窗算法反演出于田地区的地表温度,并用陆面实际地表温度进行了检验.结果表明,水体表面温度最低,绿洲次之,戈壁最高.地表温度以绿洲为中心从里到外呈环状分布.说明利用ETM+数据反演地表温度是可行的 .并对进一步理解我国西北干旱、半干旱区的土壤-植被-大气系统能量交换状况及区域热量分布差异有着重要意义.     

人HIF-1α基因重组减毒沙门氏菌载体的构建及稳定性检测

利用RT-PCR方法从低氧预处理的A549细胞中获得HIF-1α基因片段,将其定向插入pIRES-SEQ质粒NheⅠ、MLuⅠ2个酶切位点之间,得到真核表达载体pIRES-HIF-1α;采用电转化方法将其电转化入减毒沙门氏菌Ty21α中,获得重组减毒沙门氏菌菌株TPH;转染A549细胞,RT-PCR法检测HIF-1α基因表达,ELISA方法检测HIF-1α蛋白表达;TPH菌株在氨苄青霉素(+)和氨苄青霉素(-)LB培养板上分别传代40代,每10代提取质粒进行PCR及酶切鉴定.结果表明:试验成功构建了携带HIF-1α基因的重组减毒沙门氏菌TPH,其可在体外细胞中稳定表达HIF-1α基因及蛋白,TPH可稳定遗传.     

β-蒎烯合成诺蒎酸的反应条件研究

研究以松节油中的β-蒎烯为原料,通过KMnO4氧化合成诺蒎酸,并对氧化反应步骤中各因素对反应的影响进行了探讨。制备诺蒎酸的最佳条件为:β-蒎烯、KMnO4、NaOH的物质的量之比1∶2.5∶1,水为溶剂,温度25~30℃,反应时间4~5 h,得率达到35%。
