‘Empire’ apples [Malus sylvestris (L.) Mill var. domestica (Borkh.) Mansf.] are susceptible to development of chilling injury, expressed as firm flesh browning, during controlled atmosphere (CA) storage. Because of this susceptibility, fruit are typically stored at 2–4 °C, but the incidence of flesh browning can be increased by 1-methylcyclopropene (1-MCP) treatment at these temperatures. In this study, flesh browning development has been investigated in relationship to ethylene production, internal ethylene concentration (IEC), flesh firmness, total phenolic concentrations, and the activities of polyphenol oxidase (PPO) and peroxidase (POX) in the flesh tissues. Fruit were harvested from two orchards, either untreated or 1-MCP treated, and then stored under CA conditions at either 0.5 or 4 °C. Fruit were removed from storage at 1.5-month intervals for 10.5 months. 1-MCP treated apples were firmer than those of untreated apples, and had lower IECs, at all removals. Flesh browning incidence and severity developed earlier in 1-MCP-treated apples than untreated apples stored at either temperature. Total phenolic concentrations differed by orchard, but no major differences in concentrations were detected between untreated and 1-MCP treated apples. However, PPO activities were higher in the flesh of 1-MCP treated apples than untreated apples from both orchards and at both storage temperatures. POX activity was not consistently affected by 1-MCP treatment or storage temperature. Overall, our results suggest that inhibited ethylene production, either as a result of storage at 0.5 °C, or by treatment with 1-MCP at either temperature, may cause stress and damage to cells and result in higher PPO activity that leads to progressive flesh browning development during CA storage.  相似文献

Balloon flower (Platycodon grandiflorum A. DC) is widely distributed in South Korea and there are some local landraces that are cultivated as a vegetable crop or medicinal plant. Making use of the gene resources of wild-type and landraces is a way to increase the genetic diversity of the cultivars. However, few tools or information are available on an efficient identification system for maintaining and management of these landraces. To improve the genetic resources for balloon flower, 22 simple sequence repeat (SSR) markers, also known as microsatellite markers, were evaluated in a collection of 42 balloon flower landraces, 34 of which were from Korea and eight from China. All microsatellite markers produced the 107 alleles ranging from 2 to 10 with a mean of 4.864 alleles per each locus (NA). The values of observed heterozygosity (HO) and expected heterozygosity (HE) ranged from 0.00 to 0.667 (mean of 0.285) and from 0.024 to 0.741 (mean of 0.416), respectively. An average value of polymorphic information contents (PIC) were 0.382 with a range of 0.023 to 0.703. Results of population structure and phylogenetic and principal coordinate analysis (PCoA) indicated that P. grandiflorum germplasm formed two largely distinct clusters according to their origins and the genetic differentiation. There was a high level of genotypic diversity at broad geographic regions between Korea and China, but the low genetic differentiation was found within the collections from Korea. The results of the genetic diversity will be useful for the selection of the parents for developing balloon flower breeding and the multi-locus SSR markers developed herein will be a valuable resource for germplasm assessments, evaluation of genetic diversity, and population genetic studies of balloon flower.  相似文献

Soybean is a major source of protein meal in the world. Soybean kunitz trypsin inhibitor (SKTI) protein is a responsible for the inferior nutritional quality of unheated or incompletely heated soybean meal. The primary objective of this research was to identify DNA markers linked to the Ti locus controlling presence and absence of kunitz trypsin inhibitor protein. Two mapping populations were developed. Population 1 was derived from a cross between cultivar Jinpumkong2 (TiTi) and C242 (titi). Population 2 was made from a mating between cultivar Clark (TiTi) and C242. The F1 plants were grown in the greenhouse to produce F2 seeds. Each F2 seed from F1 plants was analyzed electrophoretically to determine the presence of the SKTI protein band. One-thousand RAPD primers, 342 AFLP primer sets, and 35 SSR primers were used to map Ti locus in population 1 and 2. The presence of SKTI protein was dominant to the lack of a SKTI protein and kunitz trypsin inhibit protein band was controlled by a single locus. Twelve DNA markers (4 RAPD, 4 AFLP, and 3 SSR) and Ti locus were found to be genetically linked in population 1 consisted with 94 F2 individual plants. Three SSR markers (Satt409, Satt228, and Satt429) were linked with Ti locus within 10 cM. Satt228 marker was tightly linked with Ti locus. Satt228 marker was tightly linked within 0–3.7 cM of the Ti locus and may be useful in a marker assisted selection program.  相似文献

Monosomic sugar beet/wild beet addition lines (2n = 19) with full resistance against the beet cyst nematode have been characterized in different ways. Within the B. procumbens and B. webbiana addition lines three groups could be classified according to their isozymes pattern, growth habit, transmission rate, and resistance level. It is assumed that B. procumbens and B. webbiana each possess three different chromosomes which carry genes for nematode resistance. In the offspring of the addition lines diploid translocation types appear at very low frequencies, Isozyme pattern or growth type of the resistant plants were used for selecting diploid types in the offspring of monosomic addition lines. Effective selection could be made in progenies of susceptible sugar beets pollinated by addition lines because the pollen transmission of the alien chromosome is very low. Using these methods 7 nematode-resistant sugar beet lines could be selected. The transmission rates of the resistance gene ranged from 70.6% to 100%. Threw heterozygous progenies showed a 1:1 segregation indicating monogenic dominant inheritance of resistance. The level of resistance was as high as in the addition lines.  相似文献

试验旨在研究饲粮中添加复合益生素(JS菌)对生长育肥猪生产性能、盲肠菌群、挥发性脂肪酸及蛋白质腐败产物的影响。试验选择120头体重10 kg左右的健康DLY仔猪,随机分为对照组、抗生素组、0.1%JS菌组和0.2%JS菌组,每组5个重复。结果表明:与对照组相比,添加0.2%JS菌显著提高试猪30~50 kg阶段的平均日增重和采食量(P0.05);添加0.1%JS菌显著提高试猪80~100 kg阶段的平均日增重(P0.05);从试验全期来看,抗生素组、0.1%JS菌组和0.2%JS菌组试猪平均日增重和采食量均有所提高,但各组间差异不显著(P0.05)。添加JS菌趋于增加盲肠内容物乳酸杆菌数量,减少大肠杆菌数量(P0.05),从而提高二者的比值,分析PCR-DGGE图谱发现添加JS菌对菌群多样性和相似性也有一定影响。与对照组相比,0.1%JS菌组和0.2%JS菌组盲肠内容物乙酸含量分别提高2.35倍和2.99倍,丁酸含量分别提高58.87%和62.10%,挥发性盐基氮含量分别降低31.91%和28.53%(P0.05)。在本试验条件下,添加JS菌对生长育肥猪的生产性能有一定改善,效果与抗生素基本相当,这可能与其对肠道健康的保护作用有关。  相似文献

This study was conducted to develop analytical models for the prediction of drying stresses and defomations in lumber. Both one- and two-dimensional analytical models were developed with two assumptions: the visco-elastic creep could be neglected, and the diffusion coefficient is constant with moisture changes. The method developed in this study showed that the drying stress of lumber with symmetric moisture profile could be approximately predicted using one-dimensional (1D) and two-dimensional (2D) models. In the case of the 1D model, drying deformations could be determined rather easy and the drying stress can even be predicted by hand calculation. The results of this study might be used for lumber with an asymmetric moisture profile. In order to predict more accurately drying stress and deformation across overall moisture changes, however, this procedure should be incorporated with the other moisture transport models and might be extended to a 3D model.  相似文献

The effects of local climate and silvicultural treatment on the inorganic N availability, net N uptake capacity of mycorrhizal beech roots and microbial N conversion were assessed in order to characterise changes in the partitioning of inorganic N between adult beech and soil microorganisms. Fine root dynamics, inorganic N in the soil solution and in soil extracts, nitrate and ammonium uptake kinetics of beech as well as gross ammonification, nitrification and denitrification rates were determined in a beech stand consisting of paired sites that mainly differed in aspect (SW vs. NE) and stand density (controls and thinning treatments). Nitrate was the only inorganic N form detectable in the soil water. Its concentration was high in control plots of the NE aspect, but only in canopy gaps and not influenced by thinning. Neither thinning nor aspect affected the abundance of root tips in the soil. Maximum nitrate net uptake by mycorrhizal fine roots of beech, however, differed with aspect, showing significantly lower values at the SW aspect with warm–dry local climate. There were no clear-cut significant effects of local climate or thinning on microbial N conversion, but a tendency towards higher ammonification and nitrification and lower denitrification rates on the untreated controls of the SW as compared to the NE aspect. Apparently, the observed sensitivity of beech towards reduced soil water availability is at least partially due to impaired N acquisition. This seems to be mainly a consequence of reduced N uptake capacity rather than of limited microbial re-supply of inorganic N or of changed patterns of inorganic N partitioning between soil bacteria and roots.  相似文献

The recently discovered oak‐specific fine root plant pathogen Phytophthora quercina is a significant factor in the current phase of European oak decline but its origins and ecology are poorly understood. A genome‐wide analysis of 260 amplified fragment length polymorphism (AFLP) markers was used to examine the genetic diversity of 72 isolates from five oak species at 28 sites in Germany (particularly Bavaria), Italy, France, Hungary and the UK. Within‐site diversity was examined at 16 sites. The limited genetic diversity (within and between sites) and lack of genetic substructuring according to geographic origin or host species suggest the rapid spread of a relatively recently introduced species. Two subgroups were distinguished and these may reflect an initial introduction of isolates of two different genetic backgrounds. The relatively low genetic diversity is probably because of the predominantly inbreeding (homothallic) nature of P. quercina. However, evidence of limited intra‐site diversity, temporal variation and the lack of clonality within the European population suggest that some diversity is being maintained by occasional outcrossing and turnover of a reservoir of long‐lived soil‐borne oospore (sexually derived) inoculum.  相似文献

A phytochemical fractionation of the methanol extract of the Morus alba leaves led to the isolation of eleven flavonoids (111). The structure of the new 3′-geranyl-3-prenyl-2′,4′,5,7-tetrahydroxyflavone (1) was elucidated by means of spectroscopic methods. The cytotoxicity of the isolated compounds against human cervical carcinoma HeLa, human breast carcinoma MCF-7, and human hepatocarcinoma Hep3B cells was evaluated. Of note, morusin (9) was the most potent with an IC50 value of 0.64 μM against HeLa cells.  相似文献
