Early vigour is an important characteristic for direct-seeded rice systems. The genetic control of early vigour was studied using a population of 129 backcross lines derived from a cross between Vandana, an improved indica, and Moroberekan, a traditional japonica. Screening was conducted under controlled conditions in greenhouse and field conditions, and indicators of early vigour, including shoot length, shoot biomass, leaf area, number of roots, root biomass, partitioning coefficients, and growth rates, were measured. Phenotypic correlations suggested that traits that were related and combined could be used to define early vigour. Broad-sense heritability ranged from moderate to high. Many regions were identified containing more than one QTL, suggesting that these traits were controlled by pleiotropic and/or closely linked QTLs. Many QTLs were specific to one environment but G × E interaction analysis showed that the main effects of the environment were large. Differences in temperature between experiments resulted in large differences in seedling age when expressed in thermal time. Different genes (QTLs) may be expected to control growth at different time intervals and thus may partly explain the limited agreement between experiments. However, several regions showed co-location of QTLs from more than one experiment. Comparisons with published studies revealed that these regions were previously identified in different genetic backgrounds and could potentially be used as introgression targets in a marker-assisted breeding program to improve germplasm for direct-seeded environments. 相似文献

Microbial biomass was determined by three biochemical procedures in nine topsoils from a climosequence in tussock grasslands. The pH values of the samples ranged from 4.4 to 6.2 and organic C contents from 2.5 to 20.0%. When determined by a chloroform-fumigation procedure, contents of biomass C and mineral-N (Min-N) flush ranged from 530–2780 and 59–167 μgg?1 dry soil respectively. Adenosine 5'-triphosphate (ATP) content ranged from 2.2 to 10.7 μg g?1 dry soil. All three estimates were significantly correlated with each other and with several soil properties, including organic C and total N contents and CO2 production. They were not significantly correlated with any climatic factor.In spite of these significant correlations, the ratios of the biomass estimates varied appreciably in the different soils. The ratios of biomass C/Min-N flush ranged from 7.8 to 22.8 (average 12.5), biomass C/ATP from 163 to 423 (average 248) and Min-N flush/ATP from 12 to 35 (average 22). These ratios were mostly higher than those found elsewhere for Australian and English soils. The high biomass C/ATP and Min-N flush/ATP ratios did not appear to originate from inefficient extraction of “native” ATP or from the soils' P status. Based on these results, care in the use of factors for obtaining soil microbial biomass content from Min-N flush or ATP values is indicated. 相似文献

We report the design and total chemical synthesis of "synthetic erythropoiesis protein" (SEP), a 51-kilodalton protein-polymer construct consisting of a 166-amino-acid polypeptide chain and two covalently attached, branched, and monodisperse polymer moieties that are negatively charged. The ability to control the chemistry allowed us to synthesize a macromolecule of precisely defined covalent structure. SEP was homogeneous as shown by high-resolution analytical techniques, with a mass of 50,825 +/-10 daltons by electrospray mass spectrometry, and with a pI of 5.0. In cell and animal assays for erythropoiesis, SEP displayed potent biological activity and had significantly prolonged duration of action in vivo. The susceptibility locus for the autoimmune disease lupus on murine chromosome 1, Sle1z/Sle1bz, and the orthologous human locus are associated with production of autoantibody to chromatin. We report that the presence of Sle1z/Sle1bz impairs B cell anergy, receptor revision, and deletion. Members of the SLAM costimulatory molecule family constitute prime candidates for Sle1bz, among which the Ly108.1 isoform of the Ly108 gene was most highly expressed in immature B cells from lupus-prone B6.Sle1z mice. The normal Ly108.2 allele, but not the lupus-associated Ly108.1 allele, was found to sensitize immature B cells to deletion and RAG reexpression. As a potential regulator of tolerance checkpoints, Ly108 may censor self-reactive B cells, hence safeguarding against autoimmunity. 相似文献

A standardized investigation was conducted to compare the biophysical properties of hyaluronate products used in the treatment of joint dysfunction in horses. Results showed that two of the five products, Hyvisc® and Hylartin®V, contained higher-weight molecular hyaluronate (greater than 1.5×106). Other investigations have shown that higher molecular weight hyaluronate is effective in providing physical resistance to shear, in stimulating endogenous hyaluronate synthesis, and in inhibiting excess cell proliferation which may be associated with synovial pannus formation. It is suggested that the therapeutic benefits of hyaluronate products may be dependent on the molecular size of the hyaluronate molecules. Higher molecular weight hyaluronate products would be expected to be more effective than lower molecular hyaluronate products in providing therapeutic benefits. 相似文献

BACKGROUND: Standardized hematologic methods and reference intervals have not been established for cartilaginous fishes (sharks, skates, and rays) despite the large number of animals displayed in zoos and aquariums worldwide. OBJECTIVE: The focus of this study was to validate CBC methods for sandbar shark (Carcharhinus plumbeus) blood, based on criteria established in human medicine, for the following tests: RBC count, total WBC count, PCV, hemoglobin (Hgb) concentration, and WBC differential percentages. METHODS: Replicate CBCs were performed using blood samples from 5 captive sandbar sharks. Three protocols for RBC and total WBC counts were compared, as were different centrifugation times for PCV determination, and 2 methods for Hgb concentration. Means, minimum and maximum values, and CVs were compared to CAP and CLIA performance guidelines for human tests. RESULTS: Total WBC counts in a diluent modified for elasmobranch blood, Hgb concentration by the cyanmethemoglobin method after removal of nuclei, and WBC differential percentages showed acceptable performance. PCV results were acceptable when tubes were centrifuged for at least 5 minutes. Total RBC counts by all 3 methods exceeded the acceptable error for manual counts of human cells. CONCLUSIONS: Standardized CBC tests can be used as health assessment tools for elasmobranchs. Total RBC counts should be viewed as estimates. 相似文献
