Identification of vaccine candidates against serogroup B meningococcus by whole-genome sequencing

Neisseria meningitidis is a major cause of bacterial septicemia and meningitis. Sequence variation of surface-exposed proteins and cross-reactivity of the serogroup B capsular polysaccharide with human tissues have hampered efforts to develop a successful vaccine. To overcome these obstacles, the entire genome sequence of a virulent serogroup B strain (MC58) was used to identify vaccine candidates. A total of 350 candidate antigens were expressed in Escherichia coli, purified, and used to immunize mice. The sera allowed the identification of proteins that are surface exposed, that are conserved in sequence across a range of strains, and that induce a bactericidal antibody response, a property known to correlate with vaccine efficacy in humans.     

GONG Observations of Solar Surface Flows

Doppler velocity observations obtained by the Global Oscillation Network Group (GONG) instruments directly measure the nearly steady flows in the solar photosphere. The sun's differential rotation is accurately determined from single observations. The rotation profile with respect to latitude agrees well with previous measures, but it also shows a slight north-south asymmetry. Rotation profiles averaged over 27-day rotations of the sun reveal the torsional oscillation signal-weak, jetlike features, with amplitudes of 5 meters per second, that are associated with the sunspot latitude activity belts. A meridional circulation with a poleward flow of about 20 meters per second is also evident. Several characteristics of the surface flows suggest the presence of large convection cells.     

Single-Electron Transport in Ropes of Carbon Nanotubes

The electrical properties of individual bundles, or "ropes," of single-walled carbon nanotubes have been measured. Below about 10 kelvin, the low-bias conductance was suppressed for voltages less than a few millivolts. In addition, dramatic peaks were observed in the conductance as a function of a gate voltage that modulated the number of electrons in the rope. These results are interpreted in terms of single-electron charging and resonant tunneling through the quantized energy levels of the nanotubes composing the rope.     

Lipopolysaccharide binding protein in the acute phase response of experimental murine Trypanosoma brucei brucei infection

Cellular responses to lipopolysaccharide (LPS) are enhanced by LPS-binding protein (LBP). The present study investigated the acute phase response of LBP during Trypanosoma brucei brucei infection in mice. Mean plasma concentrations of LBP increased two-fold by the seventh day following infection, but decreased to intermediate levels by the 14th day. There were no significant differences in LBP concentrations of infected/antibiotic-treated and infected/untreated mice. At 35 days post-infection, the infected mice were treated with the anti-trypanosomal diminazine aceturate (Berenil^®). LBP levels of the mice then decreased to pre-infection levels within one-week. This demonstrated that LBP is an acute phase protein during murine trypanosomosis. Furthermore, opportunistic secondary bacterial infection during trypanosomosis did not seem to play an important role in the changes in plasma LBP levels. We speculate that the marked concomitant increases in plasma LBP and endotoxin-like activity following murine trypanosome infection might play an important role in the pathogenesis of trypanosomosis.     

Bone haemangio-sarcoma in a young Belgian malinois

Bone haemangiosarcoma in a young Belgian malinois initially presented as an abscess involving the Tight 10th rib. The dog responded promptly to abscess drainage and antibiotics. Anaemia and thrombocytopenia began one week later and ultrasonography revealed splenic and liver lesions. Exploratory surgery identified tumour masses in the spleen, liver, lung and rib. Histopathological diagnosis was hemangiosarcoma, possibly of bone origin.
