Due to the importance of N in forest productivity ecosystem and nutrient cycling research often includes measurement of soil N transformation rates as indices of potential availability and ecosystem losses of N. We examined the feasibility of using soil temperature and moisture content to predict soil N mineralization rates (Nmin) at the Coweeta Hydrologic Laboratory in the southern Appalachians. We conducted seasonal laboratory incubations of A and AB horizon soils from three sites with mixed-oak vegetation using temperature and moisture levels characteristic of the season in which the soils were collected. The incubations showed that temperature and temperature-moisture interactions significantly affected net soil Nmin. We used the laboratory data to generate equations relating net Nmin to soil temperature and moisture data. Using field-collected temperature and moisture data we then calculated Nmin on similar forest sites and compared predicted rates with in situ, closed-core Nmin measurements. The comparison showed that the in situ Nmin was greater than rates predicted from laboratory generated equations (slope =3.22; r2=0.89). Our study suggests that while climatic factors have a significant effect on soil Nmin, other factors also influence rates measured in the laboratory and in situ.  相似文献

Abstract. Continuous cultivation of soils of the semiarid tropics has led to significant land degradation. Soil erosion and nutrient loss caused by high runoff volumes have reduced crop yields and contributed to offsite damage. We compared a number of soil management practices (tillage, mulch and perennial/annual rotational based systems) for their potential to improve crop production and land resource protection in an Alfisol of the semiarid tropics of India. Runoff and soil erosion were monitored and surface soil and sediment were analysed for nitrogen and carbon to determine enrichment ratios. Amelioration of soils with organic additions (farmyard manure, rice straw) or rotating perennial pasture with annual crops increased soil carbon and nitrogen contents and reduced runoff, soil erosion and nutrient loss. Soil erosion totalled less than 7 t ha–1, but enrichment ratios were often greater than 2 resulting in up to 27 kg N ha–1 and 178 kg C ha–1 being lost in sediment. Up to an extra 250 mm of water per year infiltrated the soil with organic additions and was available for crop water use or percolation to groundwater. The results show that there are good opportunities for reducing degradation and increasing productivity on farms.  相似文献

Upflow anaerobic sludge blanket (UASB)reactors were used to treat municipal wastewater attemperatures of 6, 11, 15, 20 and 32 °C and athydraulic retention times (HRTs) ranging from 48 to 3 h overan operational period of approximately 860 days. The Monodmodel was used to evaluate substrate utilization. TheArrhenius model was used to calculate the activationenergies from which temperature coefficients weredetermined. It was found that the maximum specific substrateutilization rate constant (k) decreased from 0.387 d-1(at 32 °C) to 0.041 d-1 (at 6 °C). Thedecay rate constant (kd), yield coefficient (Yg),half velocity constant (Ks) and maximum specific growthrate (μm ) were also impacted by temperature in theUASB treatment of municipal wastewater.  相似文献

Aggregates and gel network structure of globin hydrolysates   总被引:4,自引:0,他引:4  
A gel with excellent functional properties was prepared successfully using the hydrolysates of globin. In the present study, the structures of intermediate aggregates and gel network were observed directly with an electronic microscope. It was shown clearly that the intermediate aggregates were in a thin rod shape with a length of 130--140 nm, which was in good accordance with the results of the light scattering obtained in a previous study. The diameter of intermediate aggregates was 4--5 nm. Each unit of the intermediate aggregate was composed of beta-chain and peptide beta-1 in a ratio of 1:1. Its molecular weight was 26922 Da, and it had a diameter of 4.1 nm. The thin rod-shaped aggregates were formed with units through the hydrophobic interaction. The length of intermediate aggregate was >30--33 times the diameter. Furthermore, the cross-linked structure formed by peptide alpha-1 and the thin rod-shaped aggregates was also confirmed by the photography of the electronic microscope. These results supported the model proposed in previous papers as proper to depict exactly the formation and structure of the gel network of globin hydrolysates.  相似文献

A high-performance liquid chromatographic (HPLC) method for the determination of anthocyanidins from berries and red wine is described. Delphinidin, cyanidin, petunidin, pelargonidin, peonidin, and malvidin contents of bilberry (Vaccinium myrtillus), black currant (Ribes nigrum), strawberry (Fragaria ananassa cv. Jonsok), and a Cabernet sauvignon (Vitis vinifera) red wine were determined. The aglycon forms of the anthocyanins present in the samples were revealed by acid hydrolysis. A reversed phase analytical column was employed to separate the anthocyanidins before identification by diode array detection. The suitability of the method was tested by determining the recovery (95-102% as aglycons and 69-104% from glycosides) for each anthocyanidin. Method repeatability was tested by charting the total aglycon content of two samples over a period of 14 analyses and determining the coefficients of variation (1.41% for bilberry and 2.56% for in-house reference material). The method developed proved thus to be effective for reliable determination of anthocyanidins from freeze-dried berry samples and red wine. The total anthocyanidin content of the tested samples was as follows: in-house reference material, 447 +/- 8 mg/100 g; strawberry, 23.8 +/- 0.4 mg/100 g; black currant, 135 +/- 3 mg/100 g; bilberry, 360 +/- 3 mg/100 g; and Cabernet sauvignon red wine, 26.1 +/- 0.1 mg/100 mL.  相似文献

The effects of product and preparation variables on the in-cup chemical composition of tea extracts is of interest because the appearance and taste characteristics and the possible health effects of a tea liquor arise from the chemical components extracted from the leaf during tea preparation. A comprehensive study was therefore undertaken to determine the contributions of product and preparation variables on the total soluble solids, caffeine, and polyphenol contents of tea extracts. The results of this study show that the variety, growing environment, manufacturing conditions, and grade (particle size) of the tea leaves each influence the tea leaf and final infusion compositions. In addition, the composition of the tea infusion was shown to be influenced by whether the tea was contained in a teabag and, if so, the size and material of construction of the bag. Finally, the preparation method, including the amounts of tea and water used, infusion time, and amount of agitation, was shown to be a major determinant of the component concentrations of tea beverages as consumed. An illustration of the variation introduced by these product and preparation factors is provided by comparing solids, caffeine, and polyphenol contents of green and black tea infusions when commercial products are prepared according to the instructions given on their packaging.  相似文献

Efforts in grasspea (Lathyrus sativus) improvement have increased since the development of lines that are very low in the neurotoxin Beta-N-oxalyl-L-alpha-beta-diamino propionic acid (ODAP); also referred to as Beta-N oxalyl-amino-L-alanine (BOAA). Many programs now address several related aspects of improvement simultaneously. These include reduced ODAP concentrations, insect and disease resistance, nitrogen fixation, agronomic practices, fodder and forage production, and components for increased yielding ability. The coordinated, multidisciplinary approach now being applied to the genetic improvement of grasspea should allow the potential of this largely neglected grain legume to be fully realized.  相似文献

In the Canary Islands, water scarcity is one of the constraints for agricultural activity. Non-conventional water resources generally represent more water volume than conventional ones. The distribution of these resources frequently permits the possibility of a conjunctive use of desalinated (DW) water and reclaimed municipal wastewater (RW). Field testing with both water qualities and different irrigation systems is necessary for optimal site-specific management. The objective of this work was to evaluate soil salinity and phosphorus distribution, and alfalfa yield in a 20 month field experiment carried out in the island of Gran Canaria, using municipal RW and freshwater (FW) under subsurface drip irrigation (SDI). Phosphorus speciation was performed both in irrigation waters and in soils (Olsen's inorganic, organic, and microbial). RW had large EC values (2.4 dS m−1) with a remarkable nutrient load contribution and an average total P around 3 mg L−1, predominantly hydrolysable forms, while FW had very low salinity and negligible amounts of P. For the RW treatment a salt gradient was established, causing plant mortality between the irrigation lines. The study of P speciation allows describing P distribution and plant uptake in terms of P forms. Large values of microbial P were produced for the two irrigation waters around the emitters, especially for FW.A faster P-cycling could have contributed to the significantly larger inorganic P contents observed in FW irrigated soils, in spite no external sources were added by the irrigation water.  相似文献

Ammonia oxidizing bacteria (AOB) control the rate limiting step of nitrification, the conversion of ammonia (NH4+) to nitrite (NO2). The AOB therefore have an important role to play in regulating soil nitrogen cycling. Tillage aerates the soil, stimulating rapid changes in soil N cycling and microbial communities. Here we report results of a study of the short term responses of AOB and net nitrification to simulated tillage and NH4+ addition to soil. The intensively farmed vegetable soils of the Salinas Valley, California, provide the context for this study. These soils are cultivated frequently, receive large N fertilizer inputs and there are regional concerns about groundwater N concentrations. An understanding of N dynamics in these systems is therefore important. AOB population sizes were quantified using a real-time PCR approach. In a 15 day experiment AOB populations, increased rapidly following tillage and NH4+ addition and persisted after the depletion of soil NH4+. AOB population sizes increased to a similar degree, over a 1.5-day period, irrespective of the amount of NH4+ supplied. These data suggest selection of an AOB community in this intensively farmed and C-limited soil, that rapidly uses NH4+ that becomes available. These data also suggest that mineralization may play an especially important role in regulating AOB populations where NH4+ pool sizes are very low. Methodological considerations in the study of soil AOB communities are also discussed.  相似文献

The structural characteristics of the gum exudate of Acacia senegal (gum arabic) have been investigated by monitoring the composition and physicochemical properties before and after treatment with proteolytic enzyme and various alkaline systems. Molecular mass ( M w) and radius of gyration ( R g) measurements were performed using gel permeation chromatography (GPC) coupled to refractive index, UV absorbance, and multiangle light scattering detectors and indicated that the macromolecules present have a compact structure. It was found that treatment with proteolytic enzyme caused the arabinogalactan-protein component (AGP) with average molecular mass approximately 2 x 10 (6) Da to degrade, yielding material of molecular mass approximately 4 x 10 (5) Da, whereas the bulk of the material corresponding to the protein-deficient arabinogalactan component (AG) with molecular mass 4 x 10 (5) remained unaffected. Barium hydroxide was found to hydrolyze the polysaccharide component (AG) itself in addition to the proteinaceous component as demonstrated in control experiments using dextran. However, sodium borohydride/sodium hydroxide treatments were unable to hydrolyze dextran and were assumed to hydrolyze only the proteinaceous component of gum arabic. The AGP component was completely degraded, yielding material of molecular mass approximately 4.5 x 10 (4) Da. It has been concluded, therefore, that the enzyme did not fully hydrolyze all of the protein present and that the AGP component of gum arabic consists of carbohydrate blocks of approximately 4.5 x 10 (4) Da linked to a polypeptide chain consistent with the wattle blossom structure. Because the AGP was degraded to differing extents using a mild and more severe sodium borohydride/sodium hydroxide treatment, it was concluded that the polysaccharide moieties were linked through both O-serine and O-hydroxyproline residues. The gum arabic sample was deglycosylated by treatment with anhydrous hydrogen fluoride and revealed the presence of two putative core proteins of approximately 3 x 10 (4) and approximately 5 x 10 (3) Da, respectively, which correspond to proteins of approximately 250 and 45 amino acids in length. A new model for the structure of the AGP component has been proposed.  相似文献
