本文研究了氨苄青霉素、羧苄青霉素和头孢霉素等3种抗生素对火炬松愈伤组织的生长和分化及不定芽生根的影响。结果表明,头孢霉素最有利于愈伤组织的诱导和生长,羧苄青霉素最有利于芽的分化,氨苄青霉素降低了不定芽的生根频率。所有试验的3种抗生素提高愈伤组织的形成和芽再生,但降低了芽的生根频率。这些结果表明,选择合适的抗生素对优化火炬松遗传转化体系有重要作用。图3表4参25。  相似文献

Agricultural productivity may be raised in a sustainable way by many different technologies such as biological fertilizers, soil and water conservation, biodiversity conservation, improved pest control, and changes in land ownership and distribution. Of these measures, biotechnology applications probably hold the most promise in augmenting conventional agricultural productivity, because biotechnology applications give not only the need to increase production, but also protect the environment and conserving natural resources for future generations. Biotechnology applications will have the possibilities to increase productivity and food availability through better agronomic performance of new varieties, including resistance to pests; rapid multiplication of disease-free plants; ability to obtain natural plant products using tissue culture; diagnosis of diseases of plants and livestock; manipulation of reproduction methods increasing the efficiency of breeding; and the provision of incentives for greater participation by the private sector through investments. Insect resistance through the transfer of a gene for resistance fromBacillus thuringiensis (Bt) is one of the most advanced biotechnology applications already being commercialized in many parts of the world. This paper reviews the development and the status ofBt technology and application ofBt transgenic plants in current agriculture, and discusses specific issues related to the transfer of the technology to the future of genetic engineered trees with emphasis on conifers. Prosopis cineraria is an important species for aridzone agroforestry in India. Information on the chemical composition and nutritive value of the leaves of young trees that will be useful in screening the provenances, is lacking. Chemical composition (N, P, K, Ca, Mg, Na), crude protein, and structural carbohydrates (neutral detergent fibre, acid detergent fibre, cellulose, hemicellulose, and lignin) of the foliage of one-year-old plants of 31 provenances ofProsopis cineraria were determined. The seeds were collected from 16°50 N to 29°55 N latitude and 69°49 E to 78°03 E longitude in India, and grown at Hisar (29°10 N latitude, 75°46 E longitude and 215 m altitude). Mineral contents (except N and P), crude protein, and structural carbohydrates varied significantly (p<0.05) between provenances, and the greatest amount of N, P, crude protein, NDF, ADF and lignin contents were found in Hisar provenance. The in vitro dry matter digestibility varied from 18.2% in Gandhinagar provenance to 34.0% in that of Barmer provenance.  相似文献

Soil carbon (C) stocks in forest ecosystems have been widely estimated to a fixed soil depth (i.e., 0-30 cm) to clarify temporal changes in the C pool. However, surface elevations change as a result of compaction or expansion of the soil under forest management and land use. On the other hand, the calculation of soil C stocks based on “equivalent soil mass” is not affected by compaction or expansion of forest soil. To contribute to the development of a forest C accounting methodology, we compared changes in soil C stocks over 4 years between depth- and mass-based approaches using original soil data collected at 0-30 cm depths in young plantations and secondary forests in West Java, Indonesia. Our methodology expanded on the mass-based approach; rather than using one representative value for the mass-based calculation of soil C stocks, we adjusted individual values, maintaining the coefficient of variance in soil mass. We also considered the effect of an increase or decrease in soil organic matter on equivalent soil mass. Both increasing and decreasing trends in soil C stocks became clearer when the mass-based approach was used rather than the depth-based approach. The trends in soil C stocks based on equivalent soil mass were particularly evident in the surface soil layers (0-5 cm) and in plantation sites, compared with those for soil profiles including subsurface soil layers (0-30 cm) and in secondary forests. These trends in soil C stocks corresponded with temporal trends in litter stocks. We suggest that equivalent mass-basis soil C stock for the upper 30 cm of soil be calculated based on multiple soil layers to reduce estimation errors. Changes in soil organic matter mass had little effect on the estimation of soil C stock on an equivalent mass basis. For the development of a forest C accounting system, the mass-based approach should be used to characterize temporal trends in soil C stocks and to improve C cycle models, rather than simpler methods of calculating soil C stocks. These improvements will help to increase the tier level of country-specific forest C accounting systems.  相似文献

OBJECTIVE: To compare effects of oral supplementation with an experimental potassium-free sodium-abundant electrolyte mixture (EM-K) with that of oral supplementation with commercial potassium-rich mixtures (EM+K) on acid-base status and plasma ion concentrations in horses during an 80-km endurance ride. ANIMALS: 46 healthy horses. PROCEDURE: Blood samples were collected before the ride; at 21-, 37-, 56-, and 80-km inspection points; and during recovery (ie, 30-minute period after the ride). Consumed electrolytes were recorded. Blood was analyzed for pH, PvCO2, and Hct, and plasma was analyzed for Na+, K+, Cl-, Ca2+, Mg2+, lactate, albumin, phosphate, and total protein concentrations. Plasma concentrations of H+ and HCO3-, the strong ion difference (SID), and osmolarity were calculated. RESULTS: 34 (17 EM-K and 17 EM+K treated) horses finished the ride. Potassium intake was 33 g less and Na+ intake was 36 g greater for EM-K-treated horses, compared with EM+K-treated horses. With increasing distance, plasma osmolarity; H+, Na+, K+, Mg2+, phosphate, lactate, total protein, and albumin concentrations; and PvCO2 and Hct were increased in all horses. Plasma HCO3-, Ca2+, and Cl- concentrations were decreased. Plasma H+ concentration was significantly lower in EM-K-treated horses, compared with EM+K-treated horses. Plasma K+ concentrations at the 80-km inspection point and during recovery were significantly less in EM-K-treated horses, compared with EM+K-treated horses. CONCLUSIONS AND CLINICAL RELEVANCE: Increases in plasma H+ and K+ concentrations in this endurance ride were moderate and unlikely to contribute to signs of muscle fatigue and hyperexcitability in horses.  相似文献

Abstract

Pansies are one of the most popular annual bedding plants in the United States. Growth and uptake of essential nutrients as influenced by N‐form ratio was evaluated in pansy as well as what role pansy nutrition plays in the protection of pansy against feeding damage by white‐tailed deer provided by selected repellents. Plants were grown under three N‐form ratios: 100:0, 50:50, and 0:100NO3:NH4. Dry weight was highest for pansy treated with 100:0 and lowest for plants treated with 0:100 NO3:NH4 Mean quality ratings were 4.07 for pansies grown with 100:0,1.80 for pansies grown with 50:50, and 0.78 for pansies grown with 0:100. Potassium (K), magnesium (Mg), and manganese (Mn) content was lower in plants treated with 0:100 than in those treated with 100:0. Ammonium may have competed with these cations for uptake. Boron (B), copper (Cu), molybdenum (Mo), and aluminum (Al) content was highest in plants treated with N‐NH4. There was no significant difference shown in nutrient levels caused by repellent treatments. Feeding damage was shown to be affected by N‐form ratio only on Day 3 of the study. Of the three repellent treatments [Deer and Rabbit Repellent (Thiram), Deer Away purrescent egg spray, and no spray], Thiram provided the greatest protection to pansies over the study period.  相似文献

Amenity turf accounts for up to 4% of land‐use in urban areas, providing key refuges for both above‐ and below‐ground biodiversity. Golf courses occupy the largest surface area of all sports facilities; however, only a limited amount of microbial ecology has been carried out to investigate differences in the size and structure of microbial communities of the soil. The soil microbial community is a key agent in nutrient cycling and delivery of other ecosystem goods and services; however, there has been little work focused on amenity turf ecosystems in the UK. A study of soil microbial community size and structure, on the range of playing areas maintained for the game of golf at a single golf course in relation to depth through the soil profile, was carried out. Soil from different playing areas showed significant differences in the size (measured using chloroform fumigation extraction) of the microbial community (P < 0.01), with a greater concentration of microbial biomass at 0–75 mm from the surface, compared with deeper zones (P < 0.01). Principal component analysis of phospholipid fatty acid (PLFA) biomarkers indicated that the community structure was significantly different at 0–75 mm from the surface on all areas of the golf course investigated (P < 0.05, in all cases). The PLFA biomarkers consistently associated with such discrimination were 16:0 and 18:1ω9 c. These findings suggest that there is a consistently larger and similarly structured microbial community associated with the surface thatch layer, commonly found in amenity turf.  相似文献

Newly developed candidate cultivars of cocksfoot (Dactylis glomerata L.), phalaris (Phalaris aquatica L.) and tall fescue (Festuca arundinacea Schreb. = syn. L. arundinaceum (Schreb) Darbysh.) were evaluated over four years for persistence and productivity against current commercial cultivars in small plots at five locations selected for lower and less reliable rainfall and difficult soils (low pH and high Al) in south‐eastern Australia known to be marginally too dry for these grass species. The five locations were ‐ representing summer dominant rainfall, Inverell, in northern New South Wales (NSW); ‐ representing uniform rainfall; Trungley Hall, (medium rainfall), and Beckom (lower rainfall) both in southern NSW; and; representing a winter dominant pattern ‐ Eversley, (higher rainfall), and Bealiba, (lower rainfall), in central Victoria. The objective was to determine if the new candidate cultivars were more likely to persist and to be productive than current commercial cultivars. The study showed that most phalaris and cocksfoot treatments were highly productive in high rainfall years at one or both sites in southern NSW. However, all treatments had become much less productive by the end of the experimental period due to plant loss under hot, dry conditions in the final summer. At Bealiba in central Victoria, cocksfoot was the most productive species with several cocksfoot treatments of both subsp. hispanica and subsp. glomerata still present at the final harvest despite a hot and dry final summer‐autumn. Tall fescue was the most productive species in the two higher rainfall environments (Inverell, Eversley) although most treatments of all species performed well at those sites. Continental tall fescues were more productive on average than Mediterranean tall fescues at the strongly acidic Eversley site. As a result of this work, two of the new cocksfoot candidates (Moroccan Fine and AVH48 Selection) and one of the tall fescue candidates (Summer Active 1) have been licensed for commercial development and release.  相似文献

The objectives of this study were to characterize the glycemic and insulinemic responses of Thoroughbred broodmares fed late spring pasture only or a mixture of pasture and a high starch or low starch feed and to test hypotheses about differences in the glycemic and insulinemic effects of these dietary regimes. A group of 15 mares were divided into three treatment groups; pasture and high starch feed (PHS), pasture and low starch feed (PLS), and pasture only (PO) and maintained in these groups for 4.5 months prior to this study. These groups were maintained on a single pasture that was temporarily divided into three sections. The study protocol was conducted over two days. On day 1 the mares were fed their respective treatments and on day 2 all mares were allowed access to pasture only. On both days plasma glucose and insulin were measured in samples taken over a 7.5 h period. Baseline measurements for glucose and insulin were not different between any of the treatment groups on either day (P > 0.05). The baseline insulin concentrations of these pasture-kept mares (26.7 ± 8.3 mIU/L) were high compared to those reported from stall-kept horses. Plasma glucose and insulin on day 1 were influenced by treatment group, sample time, and an interaction between treatment and time (P < 0.05). On day 2 there was no significant influence of treatment group (P > 0.05). Glucose and insulin rose to higher (P < 0.01) peak concentrations in the PHS group on day 1 when compared to the PLS and PO groups, with no difference (P > 0.05) detected between the PLS and PO groups. These results are reflected in greater areas under the concentration-time curves for glucose and insulin in the PHS group on day 1 (P < 0.05). On day 2 there were no differences in any of the glucose and insulin characteristics for any of the treatment groups (P > 0.05). These results indicate a clear difference in the glycemic and insulinemic effect of the PHS feed compared to the PLS and PO groups. Of further interest are the glucose and insulin characteristics of these pasture-kept mares that indicate a low insulin sensitivity and high insulin secretory response. This study provides further information on factors influencing glycemic and insulinemic responses in horses.  相似文献

A genomic library of DNA extracted from Mycobacterium paratuberculosis was constructed in the expression vector lambda gt 11. The library was screened by plaque hybridization with labelled M. paratuberculosis genomic DNA as probe. Strongly hybridizing plaques were isolated and their DNA extracted and characterised for M. paratuberculosis specificity by hybridization to DNA from other Mycobacteriaceae. A clone was obtained which was specific for M. paratuberculosis. DNA from this clone could detect 7 ng M. paratuberculosis DNA.  相似文献
