The fragile X syndrome is the most common cause of familial mental retardation. Genetic counseling and gene isolation are hampered by a lack of DNA markers close to the disease locus. Two somatic cell hybrids that each contain a human X chromosome with a breakpoint close to the fragile X locus have been characterized. A new DNA marker (DXS296) lies between the chromosome breakpoints and is the closest marker to the fragile X locus yet reported. The Hunter syndrome gene, which causes iduronate sulfatase deficiency, is located at the X chromosome breakpoint that is distal to this new marker, thus localizing the Hunter gene distal to the fragile X locus. 相似文献

Pyrolysis mass spectrometry in combination with computerized multivariate statistical analysis enables qualitative and quantitative analysis of nonvolatile organic materials containing molecular assemblies of a complexity and size far beyond the capabilities of direct mass spectrometry. - Hydromorphological rehabilitation through installing large woody material (LWM) is increasingly being used to reverse degradation of stream ecosystems. There have been many criticisms of stream rehabilitation projects, because many have not met their goals and many others have not been monitored well enough to assess whether their goals were met.
- In a before–after–control design (with samples collected 1 year before and two successive years after LWM installation), instream biotopes and their macroinvertebrate assemblages were used as structural and functional units to assess the effectiveness of LWM installed at the Rolleston Brook, a headwater tributary of the River Welland in Leicestershire, UK.
- The project was successful in enhancing the coefficient of variation of channel water depth and width, wetted surface area, number of instream biotopes, and the biotope diversity in the rehabilitated reach.
- LWM installation led to significant increases in macroinvertebrate total density, total biomass, and taxon richness. Macroinvertebrate community composition was also enhanced, so that it became more similar to that of the control reach.
- Small increases in the number of instream biotopes (appearance of gravel and leaf litter) and changes in biotope proportions (decreasing percentage of silt) were significantly related to changes in the macroinvertebrate community metrics in the rehabilitated reach.
- The results show that using macroinvertebrate community composition is more effective than only using taxon richness and/or diversity metrics for understanding the relationship between LWM installation and macroinvertebrate community responses. To be effective, samples must also be collected in a predefined sampling protocol stratified at the instream biotope level. This approach would be of great benefit in evaluating biodiversity conservation value, and could be incorporated into the advice provided by Natural England concerning restoration and protection of English rivers that are designated as Sites of Special Scientific Interest under UK legislation or Special Areas of Conservation under European legislation.

Pasteurella multocida strains are classified using the Heddleston lipopolysaccharide (LPS) serotyping scheme into 16 serovars. Understanding the structural and genetic basis for this LPS typing scheme is important because protection against infections caused by P. multocida is generally considered to be serovar specific. Here we show that the serovar 14 type strain P2225 and the serovar 1 strains X73 and VP161 express similar LPS structures. However, the serovar 14 LPS lacks the terminal phosphocholine (PCho) residues present on the serovar 1 LPS and contains the 1,4-linked β-galactose but not the 1,6-linked β-galactose. Sequencing analysis of the LPS biosynthesis outer core loci of P2225 and the serovar 1 type strain X73 showed that they were nearly identical. However, the phosphocholine biosynthesis gene, pcgA of P2225 contained a 19bp nucleotide deletion. Complementation of P2225 with an intact pcgA resulted in an LPS structure identical to that expressed by serovar 1 strain VP161 and highly similar to that expressed by strain X73, with a 1,6-linked β-galactose and both terminal PCho residues. This study has shown unequivocally that strains belonging to serovar 1 and 14 share a common LPS outer core locus and that minor changes within this locus can dramatically alter the LPS structure expressed on the surface of P. multocida, and thus has implications into our understanding of the potential to generate cross-protective vaccines. 相似文献

The biodiversity of developing countries is increasingly threatened by introductions of invasive alien species. This study on the allodiversity in Lake Naivasha, Kenya reviews the pathways, establishment rates and outcomes of introduced species, and provides the basis for determining conservation actions that, if implemented, could prevent potentially harmful effects of similar events in other East African lakes. Introductions into Naivasha commenced in the 1920s with the release of a sport fish and have since produced an allodiversity of 23 species. This includes species that are no longer present (e.g., some tilapia species), presumed no longer present (e.g., the Nile perch Lates niloticus) or whose distribution is highly localised and ecologically neutral (e.g., the coypu Myocastor coypus). It also includes species that established successfully and invoked major changes in lake ecology (e.g., the red swamp crayfish Procambarus clarkii) and a species that is producing apparent economic benefits to the local population (i.e., the common carp Cyprinus carpio). The most frequent donor continents were the Americas and most species were the result of secondary introductions. The main introduction vector was active release that aimed to enhance fishery production. Alien species now dominate each main level of the lake’s food web and produce impacts that are rarely restricted to a single ecosystem service. With a few exceptions, the majority of introductions translate into socioeconomic costs that contribute to rising social conflicts and exacerbating poverty. Development of appropriate conservation management tools within a regulatory framework could help protect Naivasha from further damage and could be used elsewhere in East African lakes to ensure that subsequent introductions enhance ecosystem services without affecting biodiversity. 相似文献
