The atypical cadherin Fat acts as a receptor for a signaling pathway that regulates growth, gene expression, and planar cell polarity. Genetic studies in Drosophila identified the four-jointed gene as a regulator of Fat signaling. We show that four-jointed encodes a protein kinase that phosphorylates serine or threonine residues within extracellular cadherin domains of Fat and its transmembrane ligand, Dachsous. Four-jointed functions in the Golgi and is the first molecularly defined kinase that phosphorylates protein domains destined to be extracellular. An acidic sequence motif (Asp-Asn-Glu) within Four-jointed was essential for its kinase activity in vitro and for its biological activity in vivo. Our results indicate that Four-jointed regulates Fat signaling by phosphorylating cadherin domains of Fat and Dachsous as they transit through the Golgi.  相似文献

It is increasingly recognised that tackling land degradation through more sustainable land management depends on incorporating multiple perspectives by using a variety of methods at multiple scales, including the perspectives of those who manage and/or use the land. This paper reports experience implementing a previously proposed methodological framework that is designed to facilitate knowledge sharing between researchers and stakeholders about land degradation severity and extent, and sustainable land management options. Empirical findings are presented from the Botswana site of the EU‐funded Desertification Mitigation and Remediation of Land project. The paper reflects upon the challenges and benefits of the proposed framework and identifies a number of benefits, notably related to insights arising from the integration of local and scientific knowledge, and the ownership of the sustainable land management strategies that emerged from the process. However, implementing the framework was not without challenges, and levels of poverty and formal education may limit the implementation of the framework in some developing world contexts. Twentieth-Century Sea Surface Temperature Trends   总被引:4,自引:0,他引:4  
An analysis of historical sea surface temperatures provides evidence for global warming since 1900, in line with land-based analyses of global temperature trends, and also shows that over the same period, the eastern equatorial Pacific cooled and the zonal sea surface temperature gradient strengthened. Recent theoretical studies have predicted such a pattern as a response of the coupled ocean-atmosphere system to an exogenous heating of the tropical atmosphere. This pattern, however, is not reproduced by the complex ocean-atmosphere circulation models currently used to simulate the climatic response to increased greenhouse gases. Its presence is likely to lessen the mean 20th-century global temperature change in model simulations.  相似文献

A transformation system was developed for English elm (Ulmus procera Salisbury) using Agrobacterium tumefaciens C58 pMP90 p35SGUS/INTRON, allowing for the transfer of foreign genes and regeneration of phenotypically normal elm plantlets. The PCR analysis indicated that both nptII and uidA genes were stably inserted in the plant genome. beta-Glucuronidase histochemical and fluorimetric assays revealed expression of the uidA gene in the shoots, leaves, stems and roots of regenerated transgenic plants. The DNA-DNA hybridizations confirmed the presence of the uidA gene in regenerant plants. Factors influencing successful transformation and regeneration of elms included: identifying gene-transfer-proficient Agrobacterium strains for use with elms; developing an infection protocol allowing T-DNA transfer while retaining the ability to remove inciting bacteria; and identifying selection conditions to eliminate non-transformed material and choice of regeneration medium to allow shoot production. The potential utility of an effective elm transformation and regeneration system in the control of Dutch elm disease is discussed.  相似文献

The stable rare earth element (REE) tracer method was evaluated in this study as a new method for studying soil erosion. Erosion and sedimentation amounts for a slope section can be estimated for each rainfall event using multiple tracers. The REE tracer method can be used for quantitative estimates of the distribution of soil erosion from specific areas and for determining sources of sediment in a small watershed.  相似文献

The metabolic rate of three captive manatees was measured at different water and air temperatures. The animals responded metabolically to cold water (15–20°C), but not to cold air (10–20°C). Average metabolic rates in the thermal neutral zone (T1 ? 24°) were 15–22% of predicted weight-specific values, and thermal conductances were 117–229% of predicted levels. Body temperatures averaged 36·4°C. The low heat production and high thermal conductance suggest that manatees are poorly adapted energetically to winter water temperatures in much of Florida. These results support behavioural observations of captive and free-living manatees which suggest that 20°C is the minimum suitable water temperature for manatees. Manatees wintering in colder waters are probably in marginal habitat and are most susceptible to cold-related mortality. Warm water refuges reduce energetic maintenance costs and probably temper the effects of occasional periods of severely cold weather, particularly in northern Florida.  相似文献
