将含高滴度小反刍兽疫病毒的新鲜病料研磨后接种VERO DS细胞系进行病毒分离,出现细胞病变后连续传代,取3代以上培养物,进行RT-PCR和间接免疫荧光试验及生长曲线测定。生长曲线测定分别以0.01和0.1 MOI接种VERO DS细胞,每隔12h测定病毒滴度,连续培养168h,绘制病毒生长曲线,分析生长特性。结果显示,病料接种细胞72h后,出现细胞病变,经RT-PCR和间接免疫荧光试验鉴定为小反刍兽疫病毒?;生长特性研究显示,0.01和0.1 MOI两种病毒接种量,病毒滴度最高均为106 TCID50左右,但峰值出现时间和下降速度存在差异,0.01 MOI接种量出现峰值晚,但是维持高滴度时间较长。本文成功分离一株小反刍兽疫病毒,并对两种接毒量的生长特性差异进行了初步分析,为探索该病毒培养方法和收毒时间提供了借鉴。  相似文献

The experiment was conducted to study the effects of intragastric lycopene on oxidative stress blood after exercise SD rats.Thirty two SD rats with similar weight(233.40 g±6.21 g) and good health for SPF were randomized into one control groups and three trial groups with 8 rats per group.The control group was gastric fed with edible oil,the trial Ⅰ,Ⅱ and Ⅲ groups were gastric fed with edible oil intragastric with 10,20,40 mg/(kg·BW) lycopene,respectively.The pre-experiment period lasted for 10 d,and the experiment period lasted for 42 d.Swimming train six days a week and train six weeks,then that blood were collection for determination of oxidative stress after swimming train 11 hours SD rats.The results showed that lycopene concentration of plasma was increased at first and decreased subsequently,after 4 h lycopene concentration of plasma in SD rat was peak.With the increase of intragastric lycopene doses,glutathione peroxidase,total antioxidant enzyme activity and total antioxidant capacity of plasma showed a trend of rising; malondialdehyde content of plasma showed a trend of decrease with the increase of supplemental lycopene dose after exercise SD rats.Therefore,the antioxidant capacity of blood after exercise SD rats and doses of intragastric lycopene were dose dependent,antioxidant capacity of blood after exercise SD rats showed a trend of rise with the increase of lycopene dose.  相似文献

A series of pyrazolo[5,1-d][1,2,3,5]tetrazin-4(3H)one derivatives were designed, synthesized, and evaluated for their herbicidal activities where some of these compounds provided >80% control of Brassica campestris at 10 microg/mL. Quantitative structure-activity relationship studies were performed on these compounds using physicochemical parameters (electronic, Verloop, or hydrophobic) as independent parameters and herbicidal activity as a dependent parameter, where herbicidal activity correlated best (r > 0.8) with physicochemical parameters in this set of molecules. The herbicidal activity against B. campestris was mainly affected by the molar refractivity (MR) for R1, Taft (Eso) for R2 or R6, Verloop (Lm) for R3 or R5, and electronic parameters (Hammett's constants) for R4. The optimal MR for herbicidal activity is 0.95. The herbicidal activity against Echinochloa crus-galli was mainly related with the substituents' hydrophobic parameter. The optimal pi parameters for R1 and R4 for herbicidal activity are 0.72 and 0.68, respectively. In general, these compounds showed greater herbicidal activity toward B. campestris than E. crus-galli.  相似文献

以农村地区的保育舍为研究对象,运用计算流体力学(CFD)模拟技术对冬季采用横向自然通风的保育舍进行速度场、温度场和颗粒物分布的模拟分析,研究同种通风方式下不同窗户开度猪舍内部的环境质量,并提出优化建议。模拟结果显示:横向自然通风猪舍气流死区较多,但仔猪周围小部分区域的温度和风速值并不能满足国标的要求,影响部分猪只正常的生长发育;对于大跨度的横向自然通风,窗户开度影响颗粒物的排出效果。窗户开度增大,颗粒物的排出效果虽有提升但并不明显,同时也加剧了颗粒物向相邻区域的扩散,最终大部分颗粒物在热升力作用下分布在屋顶周围。该研究结果为夏热冬冷地区冬季自然通风猪舍的结构设计和气流组织优化提供了理论指导。  相似文献

• Developed a two-step synthetic route to anti-plant-virus candidate NK0238. • NK0238 exhibited a broad antivirus spectrum in greenhouse. • NK0238 showed comparable antivirus activities as controls in field trials. • NK0238 was safe to birds, fish, bees and silkworms. • NK0238 has a very good prospect in commercial development. It has previously been shown that tryptophan, the biosynthesis precursor of Peganum harmala alkaloids, and its derivatives have anti-TMV activity both in vitro and in vivo. Further exploration of this led to the identification of NK0238 as a highly effective agent for the prevention and control of diseases caused by plant viruses, but the existing routes are unsuitable for its large-scale synthesis. This study optimized a route for two-step synthesis of this virucide candidate via reaction of l-tryptophan with triphosgene to produce l-tryptophan-N-carboxylic anhydride, which then reacts with n-octylamine to give NK0238 at up to 94% yield and nearly 97% HPLC purity. In addition, the route was used for the preparation of NK0238 on a>40 g scale permitting further assessment of its antivirus activity in the greenhouse and field experiments, and toxicity tests. NK0238 exhibited useful antiviral activities against a variety of viruses both in greenhouse and field experiments. The toxicity tests showed that NK0238 was not acutely toxic to birds, fish, honey bees and silkworms. The optimized route provides a solid foundation for its large-scale synthesis and subsequent efficacy and toxicity studies, its excellent activity and safety make NK0238 a promising drug candidate for further development.  相似文献

基于无人机影像的天山云杉林树高提取及蓄积量的反演   总被引:1,自引:0,他引:1  
【目的】提取影像高程数据建立模型反演天山云杉林分蓄积量,为天然林保护工程实施后天山云杉生态恢复与科学管理提供参考依据。【方法】以新疆天山西部巩留县恰西森林公园的天山云杉(Picea Schrenkiana var. tianshanica)为研究对象,无人机航拍影像与样地每木检尺为数据源,使用点云分类与克里金插值法提取无人机影像高程数据得到天山云杉树高,根据样地实测数据建立胸径树高模型,最终根据胸径树高模型反演天山云杉林林分蓄积量。【结果】利用无人机影像提取树高与实测树高存在显著正相关关系,提取平均精度为88.42%,建立天山云杉胸径-冠幅模型的相关系数为0.696,而胸径-树高模型的相关系数为0.730;验证胸径-树高模型,计算RMSE值为12.386,拟合效果显著。基于胸径-树高模型反演林分蓄积量精度为87.66%,与实测值比对,残差值大部分落在(-2,+2)残差区间。【结论】采用局部最大值算法对天山云杉林树高信息的提取效果较好,建立胸径-树高模型弥补了无人机不能对胸径直接测量的缺陷,进而反演天山云杉林林分蓄积量。  相似文献

胰岛素样生长因子2结合蛋白( IGF2BPs)是一种重要的RNA代谢调控因子,涉及多个不同的生物学过程。本研究鉴定了一个马氏珠母贝IGF2BP1基因,命名为PfIGF2BP1-1,该基因cDNA全长为7348 bp,ORF长 1818 bp,编码605个氨基酸。多序列比对和系统进化分析表明,PfIGF2BP1-1有2个RRM结构域和4个KH结构域,与其他物种来源的IGF2BPs同源。实时荧光定量PCR结果显示,PfIGF2BP1-1 在马氏珠母贝的8个组织中均有表达,在肌肉组织表达最高,其次为足和外套膜组织。利用pET-32a 原核表达载体构建了含有PfIGF2BP1-1成熟多肽区的重组质粒并成功表达蛋白。在IPTG浓度为0.5 mmol/L,37 ℃培养8 h的条件下诱导表达PfIGF2BP1-1蛋白最佳。PfIGF2BP1-1蛋白刺激马氏珠母贝外套膜原代细胞,引起壳基质蛋白基因Accbp、MSI7、Nacrein的表达水平显著上升(p < 0.05),表明PfIGF2BP1-1蛋白可诱导壳矿化相关基因的表达参与马氏珠母贝的生物矿化过程。  相似文献

The gene for penaeidin-5, an antimicrobial peptide comprising 55 amino acids, was isolated from the hemocyte of black tiger shrimp (Penaeus monodon). RT-PCR expression tests revealed that penaeidin-5 was produced in hemocytes, gills, the intestine and muscle. Western blot analysis confirmed the panaeidin-5 was aboundantin hemocytes, the intestine and hemolymph. Immunohistochemistry revealedpenaeidin-5 in the cuticle and gills that are considered primary defense barriers. The deduced amino acid sequence of penaeidin-5 included a proline-rich N-terminal domain and a carboxyl-domain that contained six cysteine residues. Circular dichrosim analysis revealed an -helix in its secondary structure and the predicted 3D structure indicated two-disulfide bridges in the -helix. Based on the sequence of penaeidin-5 peptide cDNA, synthetic penaeidin-5 was prepared to carry out functional tests. The synthetic peptide had efficient bacteriostatic and bactericidal activity against Aerococcus viridans, and also inhibited the growth of two filamentous fungi, Fusarium pisi and Fusarium oxysporum. To measure penaeidin-5 in vivo, black tiger shrimp were challenged with Vibrio alginolyticus and A. viridans. At 3 h post-challenge, penaeidin-5 was induced and bacterial numbers decreased significantly by 12 h and 24 h.  相似文献
